Recently an increasing number of new adenovirus types associated with type-dependent pathogenicity have been identified. However, identification of these clinical isolates represents the very first step to characterize novel pathogens. click here For deeper analyses, these adenoviruses need to be further characterized in basic virology experiments or they could be applied in translational research. To achieve this goal, it is essential to get genetic access and to enable genetic modification of these novel adenovirus genomes (deletion, insertion, and mutation). Here we demonstrate a high-throughput approach to get genetic access to new adenoviruses via homologous recombination. We first defined the cloning conditions regarding homology arm-length and input adenoviral genome amounts. Then we cloned four naturally occurring adenoviruses (Ad70, Ad73, Ad74, and Ad75) into easy-to-manipulate plasmids and genetically modified them by reporter gene insertion. Three recombinant adenoviruses (Ad70, Ad73, and Ad74) containing a reporter cassette were successfully reconstituted. These novel reporter-labeled adenoviruses were further characterized using the inserted luciferase reporter with respect to receptor usage, presence of anti-adenovirus antibodies, and tropism in vitro. The identified receptor usage, the relatively low prevalence of anti-adenovirus antibodies, and the various cancer cell line transduction pattern are important features of these new pathogens providing essential information for their therapeutic application.Railway wheel tread flat is one of the main faults of railway wheels, which brings great harm to the safety of vehicle operation. In order to detect wheel flats dynamically and quantitatively when trains are running at high speed, a new wheel flat detection system based on the self-developed reflective optical position sensor is demonstrated in this paper. In this system, two sensors were mounted along each rail to measure the wheel-rail impact force of the entire circumference by detecting the displacement of the collimated laser spot. In order to establish a quantitative relationship between the sensor signal and the wheel flat length, a vehicle-track coupling dynamics analysis model was developed using the finite element method and multi-body dynamics method. The effects of train speed, load, wheel flat lengths, as well as the impact positions on impact forces were simulated and evaluated, and the measured data can be normalized according to the simulation results. The system was assessed through simulation and laboratory investigation, and real field tests were conducted to certify its validity and correctness. The system can determine the position of the flat wheel and can realize the quantification of the detected wheel flat, which has extensive application prospects.The grey squirrel is an invasive alien species that seriously threatens the conservation of the native red squirrel species. With the aim of characterizing the reproductive physiology of this species due to its great reproductive success, the function of the ovarian nerve growth factor (NGF) system was analyzed in a grey squirrel population living in central Italy. During the breeding and nonbreeding seasons, the ovarian presence, distribution, and gene expression of NGF, neurotrophic tyrosine kinase receptor 1 (NTRK1), and nerve growth factor receptor (NGFR), as well as NGF plasma concentrations, were evaluated in female grey squirrels. NGF was found in the luteal cells and in the thecal and granulosa cells of follicles, while NTRK1 and NGFR were only observed in follicular thecal and granulosa cells. NGF and NGFR transcripts were almost two-fold greater during the breeding season, while no seasonal differences were observed in NTRK1 gene expression. During the breeding season, NGFR was more expressed than NTRK1. Moreover, no changes were observed in NGF plasma levels during the reproductive cycle. The NGF system seems to be involved in regulating the ovarian cycle mainly via local modulation of NGF/NGFR, thus playing a role in the reproductive physiology of this grey squirrel population.Common methods of ocean remote sensing and seafloor surveying are mainly carried out by airborne and spaceborne hyperspectral imagers. However, the water column hinders the propagation of sunlight to deeper areas, thus limiting the scope of observation. As an emerging technology, underwater hyperspectral imaging (UHI) is an extension of hyperspectral imaging technology in air conditions, and is undergoing rapid development for applications in shallow and deep-sea environments. It is a close-range, high-resolution approach for detecting and mapping the seafloor. In this paper, we focus on the concepts of UHI technology, covering imaging systems and the correction methods of eliminating the water column's influence. The current applications of UHI, such as deep-sea mineral exploration, benthic habitat mapping, and underwater archaeology, are highlighted to show the potential of this technology. This review can provide an introduction and overview for those working in the field and offer a reference for those searching for literature on UHI technology.This publication is intended to present a new way of estimating the fatigue life of various construction materials. Carpinteri's proposal was modified by replacing the fatigue limits ratio with the value of the normal to shear stress ratio for a given number of cycles. In this study, the proposed criterion and calculation model was verified for the selected group of aluminium alloys. The purpose of the analysis of the experimental studies was to check the effectiveness of the proposed method of estimating fatigue life under the applied bending and torsional load conditions. The results of the fatigue calculations are presented in graphical form by means of diagrams showing the comparison of design and experimental strength. Before fatigue life was calculated, the critical plane orientation according to Carpinteri's model and the proposed model were determined. After analyzing the results of the comparison of design and experimental durability, it can be stated that the proposed fatigue life estimation algorithm gives satisfactory results for multiaxial cyclic loads.
