Worldwide demand for tef (Eragrostis tef) as a functional food for human consumption is increasing, thanks to its nutritional benefits and gluten-free properties. As a result, tef in now grown outside its native environment in Ethiopia and thus information is required regarding plant nutrition demands in these areas, as well as resulting grain health-related composition. In the current work, two tef genotypes were grown in Israel under irrigation in two platforms, plots in the field and pots in a greenhouse, with four and five nitrogen treatments, respectively. Nutritional and health-related quality traits were analyzed, including mineral content, fatty acid composition, hydrophilic and lipophilic antioxidative capacity, total phenolic content and basic polyphenolic profile. Our results show that tef genotypes differ in their nutritional composition, e.g. higher phenolic contents in the brown compared to the white genotype. Additionally, nitrogen availability positively affected grain fatty acid composition and iron levels in both experiments, while negatively affecting total phenolics in the field trials. To conclude, nitrogen fertilization is crucial for crop growth and productivity, however it also implicates nutritional value of the grains as food. These effects should be considered when fertilizing tef with nitrogen, to optimize both crop productivity and nutritional effects.Nocardia is an opportunistic pathogen that most frequently affects the lungs. A retrospective study was conducted based on medical records of consecutive adult patients (N = 60) with nocardiosis hospitalized during 2007-2018 at a tertiary hospital in central Israel. A matched comparison group of 120 patients was randomly selected among hospitalized patients with community-acquired pneumonia. Multivariable conditional logistic regression models were fitted. Immunosuppressive pharmacotherapy was positively associated with nocardiosis (matched odds ratio [OR] 4.40, 95% confidence interval [CI] 2.25-8.62, p  less then  0.001), particularly corticosteroid therapy (matched OR 4.69, 95% CI 2.45-8.99, p  less then  0.001). Systemic corticosteroid therapy was strongly associated with pulmonary nocardiosis (matched OR 5.90, 95% CI 2.75-12.66, p  less then  0.001). The positive association between solid organ transplantation and nocardiosis was attenuated following adjustment for systemic corticosteroids in a multivariable model. The association between corticosteroid therapy and nocardiosis appeared stronger in patients with chronic pulmonary disease (OR 5.74, 95% CI 2.75-12.66, p  less then  0.001) than in the pooled analysis of all nocardiosis cases. In conclusion, corticosteroid therapy was strongly correlated with nocardiosis, particularly among individuals with chronic pulmonary disease and in pulmonary nocardiosis.Due to climate change, winter temperatures are predicted to increase worldwide. Ziziphus spina-christi is a thermophilic tree that has recently become more abundant and widespread in the eastern Mediterranean, presumably due to a gradual increase in winter temperatures. We aim to define the temperature limitations for seed germination and the growth and survival of young seedlings to broaden our understanding of the future geographical distribution of this species. We studied effects of temperature on germination, growth, and photosynthesis in a controlled environment with four different day/night temperature regimes (34/28 °C, 28/22 °C, 22/16 °C and 16/10 °C). Effects of endocarp on germination and seed germination in the field were also studied. Results showed that germination has a lower thermal optimum (34-22 °C, 63.5-67.5% germination) than growth and photosynthesis (34-28 °C). link2 Moderate cold stress (22/16 °C), did not affect germination capacity, but strongly reduced seedling growth (71%) and photosynthetic capacity (44.6%). This may explain the recent increase in the tree's relative abundance and further highlight the potential spread of this species at higher altitudes and latitudes across the Mediterranean.This study compared the tumor immune microenvironments (TIMEs) of primary gastric cancer (PGC) and paired metastatic gastric cancer (MGC). CD4+ and CD8+ T-cell density and PD-L1 expression were evaluated by multiplex immunohistochemistry, DNA mismatch repair (MMR) by immunohistochemistry, and immune-related genes by RNA sequencing. Twenty-three patients who underwent surgical treatment for PGC and MGC were enrolled in this study. CD8+ T-cell, PD-L1+ cell, and PD-L1+CK+ cell densities were significantly lower in MGC than PGC. PD-L1 positivity using a combined positive score (≥ 1%) and deficient MMR were observed in 52.2% and 8.7% of PGC samples, respectively, whereas both occurred in only 4.3% of MGC samples. The most frequent TIME types were inflamed (34.8%) and adaptive immune resistance (34.8%) in PGC, and immune desert (65.2%) and immunological ignorance (73.9%) in MGC. In transcriptome analysis, the expression of the T-cell inflamed gene set and co-stimulatory gene module was down-regulated in MGC compared to PGC. The total CD8+ T-cell density was an independent prognostic marker in both PGC and MGC (univariate P = 0.002, multivariate P = 0.006). In the present study, electroencephalography was conducted in young animals (mice, rats and nonhuman primates [NHPs]) and in aged mice to examine the effects of the selective M1 PAM VU0453595 in comparison with the acetylcholinesterase inhibitor donepezil, M1/M4 agonist xanomeline (in NHPs), and M1 PAM BQCA (in rats) on sleep/wake architecture and arousal. In young wildtype mice, rats, and NHPs, but not in M1 mAChR KO mice, VU0453595 produced dose-related increases in high frequency gamma power, a correlate of arousal and cognition enhancement, without altering duration of time across all sleep/wake stages. Effects of VU0453595 in NHPs were observed within a dose range that did not induce cholinergic-mediated adverse effects. In contrast, donepezil and xanomeline increased time awake in rodents and engendered dose-limiting adverse effects in NHPs. Finally, VU0453595 attenuated age-related decreases in REM sleep duration in aged wildtype mice. The first isolate of the Gent1984/Diepholz reassortant emerged in Northern Germany in September 2011 whereas the last German Gent/1984-like isolate was collected in October 2011.Binding affinities of metal-ligand complexes are central to a multitude of applications like drug design, chelation therapy, designing reagents for solvent extraction etc. While state-of-the-art molecular modelling approaches are usually employed to gather structural and chemical insights about the metal complexation with ligands, their computational cost and the limited ability to predict metal-ligand stability constants with reasonable accuracy, renders them impractical to screen large chemical spaces. In this context, leveraging vast amounts of experimental data to learn the metal-binding affinities of ligands becomes a promising alternative. Here, we develop a machine learning framework for predicting binding affinities (logK1) of lanthanide cations with several structurally diverse molecular ligands. Six supervised machine learning algorithms-Random Forest (RF), k-Nearest Neighbours (KNN), Support Vector Machines (SVM), Kernel Ridge Regression (KRR), Multi Layered Perceptrons (MLP) and Adaptive Boosting (AdaBoost)-were trained on a dataset comprising thousands of experimental values of logK1 and validated in an external 10-folds cross-validation procedure. This was followed by a thorough feature engineering and feature importance analysis to identify the molecular, metallic and solvent features most relevant to binding affinity prediction, along with an evaluation of performance metrics against the dimensionality of feature space. Having demonstrated the excellent predictive ability of our framework, we utilized the best performing AdaBoost model to predict the logK1 values of lanthanide cations with nearly 71 million compounds present in the PubChem database. Our methodology opens up an opportunity for significantly accelerating screening and design of ligands for various targeted applications, from vast chemical spaces.GRAS genes belong to the plant-specific transcription factors (TF's) family that are known to be involved in plant growth and development. In this study, we have identified 37 genes from the bottle gourd genome that encodes for GRAS TF's. Except for the SCLA, we were able to identify at least one gene from each of the 17 subfamilies. Gene structure and chromosomal analysis showed that maximum seven genes are present on Chr7 followed by six genes on Chr1. The subcellular location analysis revealed that most of the genes were localized in the nucleus, except for a few in chloroplast and mitochondria. Additionally, we have identified one tandem gene duplication event on Chr7 and three major motifs that were present in all the GRAS genes. Furthermore, the protein-protein interaction prediction and gene expression analysis showed five candidate hub-genes interact with various other genes and thus probably control the expression of interacting partners in different plant tissues. Overall, this study provides a comprehensive analysis of GRAS transcription factors in bottle gourd genome which could be further extended to other vegetable crops.
