Britton var. crispa (Thunb.) H. Deane, is traditionally cultivated as an edible/medicinal crop in East Asia. Its essential oil contains many bioactive compounds that are expected to have high pharmacological functionality, as well as antimicrobial and insecticidal activity. Spider mites are a major pest group for perilla cultivation. The two-spotted spider mite, Tetranychus urticae Koch, possesses divergent detoxification enzymes and has developed resistance against most acaricides. The essential oil content of perilla halves from the pre-flowering phase to the flowering phase, and ultraviolet (UV)-B radiation generally increases defense compounds. To clarify the effects of this change in essential oil content and the effects of UV-B pretreatment, we investigated the developmental success and egg production of T. urticae on leaves from the preflowering and flowering phases cultivated with and without nighttime UV-B irradiation. Both the parameters significantly increased on leaves from the flowering phase in comparison with that from the preflowering phase, suggesting that constitutively produced essential oil provided protection against mite pests in a growth phase-specific manner. The defense system also extended the developmental period of mites on red perilla leaves, but not on green perilla leaves, in preflowering phase. Although egg production was lower on red perilla leaves pretreated with UV-B, no negative effects were caused on the developmental success and duration on red and green perilla and the egg production on green perilla by UV-B pretreatment. Our findings reveal a significant impact of investment allocation of perilla plants and a small contribution of UV-B irradiation to the plant defense system.Background Diabetes management has not been evaluated in French nursing homes (NHs) for 10 years. Objectives The present study aimed to compare the management of diabetes with guidelines in older patients living in NHs. Design Observational, retrospective and multicentre study carried out in 13 NH in the Cote d'Or region of France. Settings and subjects Between January and June 2018, all NH residents older than 65 years and known to have diabetes (n = 148) were included. Methods Epidemiological, clinical and biological data and diabetes characteristics were collected from the medical records. Results The average glycated haemoglobin (HbA1C) was 7.2 ± 1.2%. In total, 51% of patients had HbA1C less then 7% (n = 70), of which 39 took one or more antidiabetic drugs. In total, 28 of those patients (40%) were at risk of developing hypoglycaemia as a result of their treatment. In all, 44.6% of patients were treated with insulin. Glinides were the most commonly prescribed oral antidiabetic drug (OAD) (27%). Capillary blood glucose monitoring (CBGM) was not carried out daily for 75% of patients taking a potentially hypoglycaemia-inducing OAD. Conclusions We found that glycaemic control was too tight in at least 36.5% of the total population and that 40% of patients with HbA1C less then 7% were potentially overtreated. The use of dipeptidyl peptidase 4 (DPP-4) inhibitors was still insufficient, as was CBGM. Avoiding hypoglycaemia is one of the priorities in the management of older patients with diabetes. Dalbavancin is a long-acting lipoglycopeptide, approved for the treatment of acute bacterial skin and skin structure infections caused by designated susceptible strains of Gram-positive microorganisms. It demonstrated promising pharmacokinetic/pharmacodynamic properties and safety data to be considered for the treatment of bone infections. However, information on the effectiveness and safety of dalbavancin in the treatment of invasive Gram-positive infections, such as osteomyelitis, is very limited. In this review, we describe the novel characteristics and potential utility of dalbavancin in the treatment of osteomyelitis and summarize its long-term clinical outcomes from published case reports, series and comparative studies.Epigenetic changes during B cell differentiation generates distinct DNA methylation signatures specific for B cell subsets, including memory B cells (MBCs) and plasma cells (PCs). Waldenström's macroglobulinemia (WM) is a complex B cell malignancy uniquely comprised of a mixture of lymphocytic and plasmacytic phenotypes. Here we integrated genome-wide DNA methylation, transcriptome, mutation and other phenotypic features of tumor cells from 35 MYD88-mutated WM patients in relation to normal plasma and B cell subsets. We discovered that WM patients naturally segregate into two groups according to DNA methylation patterns, related to normal MBC and PC profiles, and reminiscent of other memory and plasma cell-derived malignancies. Concurrent analysis of DNA methylation changes in normal and WM development were used to capture tumor-specific events, highlighting a selective reprogramming of enhancer regions in MBC-like WM and repressed and heterochromatic regions in PC-like WM. MBC-like WM hypomethylation was enriched in motifs belonging to PU.1, TCF3 and OCT2 transcription factors and involved elevated MYD88/TLR pathway activity. PC-like WM displayed marked global hypomethylation and selective overexpression of histone genes. Finally, WM subtypes exhibited differential genetic, phenotypic and clinical features. MBC-like WM harbored significantly more clonal CXCR4 mutations (P=0.015), deletion 13q (P=0.006), splenomegaly (P=0.02) and thrombocytopenia (P=0.004), while PC-like WM harbored more deletion 6q (P=0.012), gain 6p (P=0.033), had increased frequencies of IGHV3 genes (P=0.002), CD38 surface expression (P=4.1e-5), and plasmacytic differentiation features (P=0.008). Together our findings illustrate a novel approach to subclassify WM patients using patterns of DNA methylation and reveal divergent molecular signatures among WM patients.The weevil Metamasius spinolae (Gyllenhal) is the most important insect pest of cultivated prickly pear in Mexico. A previous work reported that the pheromone of this weevil species was composed by three components. In this study, we reinvestigated the aggregation pheromone of M. spinolae using gas chromatography-electroantennography and gas chromatography-mass spectrometry to locate and identify new potential pheromonal compounds. The behavioral activity of identified compounds was evaluated in laboratory and field trials. Metamasius spinolae males released four compounds 2-methyl-4-heptanone, 6-methyl-2-hepten-4-one, 2-methyl-4-octanone, and 2-hydroxy-2-methyl-4-heptanone. In the laboratory assays, depending on the concentration, the compounds were attractive, neutral, or repellent to M. spinolae. Field evaluation showed that traps baited with 2-hydroxy-2-methyl-4-heptanone singly or in most of the binary or tertiary blends where this compound was present captured a higher number of M. spinolae compared to live males and the other compounds identified. In conclusion, our results indicate that 2-hydroxy-2-methyl-4-heptanone is the main component of the aggregation pheromone of M. spinolae. By performing inter-species comparative RNA-seq analyses and functional assays, we explored the complexity of the aortic microenvironment landscape and the fine-tuning of various factors interplaying to control HSC generation both in time and space in vivo, including the ligand-receptor couple ADM-RAMP2 and SVEP1. The relationships between E. sudra and five fruit tree species (Rosaceae) were studied for 3 yr. The highest relative density of E. PKI 14-22 amide,myristoylated mw sudra was recorded on leaves of Amygdalus persica L. and Cerasus pseudocerasus Lindl. Likewise, it had the highest survival rate and shortest developmental time when it fed on these two plants species, indicating that they were the most preferred by E. sudra than the other plant species. The relative density and growth performance of E. sudra were negatively correlated with the levels of tannins and flavonoids in the leaves of the host species. Both plant species had relatively lower flavonoids and tannins, and this may have contributed to the enhanced survival and population growth of E. sudra. These results can guide the development of improved management strategies for this pest.Deep vein thrombosis and pulmonary embolism, collectively defined as venous thromboembolism (VTE), are the third leading cause of cardiovascular death in the United States. Common genetic variants conferring increased varying degrees of VTE risk have been identified by genome-wide association studies (GWAS). Rare mutations in the anticoagulant genes PROC, PROS1 and SERPINC1 result in perinatal lethal thrombosis in homozygotes and markedly increased VTE risk in heterozygotes. However, currently described VTE variants account for an insufficient portion of risk to be routinely utilized for clinical decision making. To identify new rare VTE risk variants, we performed a whole exome study of 393 individuals with unprovoked VTE and 6114 controls. This study identified four genes harboring an excess number of rare damaging variants in patients with VTE; PROS1, STAB2, PROC and SERPINC1. At STAB2, 7.8% of VTE cases and 2.4% of controls had a qualifying rare variant. In cell culture, VTE associated variants of STAB2 had a reduced surface expression compared to reference STAB2. Common variants in STAB2 have been previously associated with plasma von Willebrand factor and coagulation factor VIII levels in GWAS, suggesting that haploinsufficiency of stabilin-2 may increase VTE risk through elevated levels of these procoagulants. In an independent cohort, we found higher von Willebrand factor levels and equivalent propeptide levels in individuals with rare STAB2 variants compared to controls. Taken together, this study demonstrates the utility of gene-based collapsing analyses to identify loci harboring an excess of rare variants with functional connections to a complex thrombotic disease.
