Here, essential oils of C. flexuosus and C. martinii were analysed to identify bioactive constituents and bioactivities using a network pharmacology approach. Essential oils were isolated using hydro-distillation in a mini Clevenger apparatus. Analysis of essential oils by GC-MS revealed 20 and 15 chemical constituents in C. flexuosus and C. martinii, respectively. An ingredient-target protein-pathway network was constructed comprising 10 oil constituents (citral, geraniol, geranyl acetate, limonene, linalool, α-terpineol, borneol, α-pinene, myrcene, and n-decanol), 14 target proteins, 51 related pathways, and 108 connections. Analyses of the network showed geraniol, geranyl acetate, limonene, linalool, and citral as major active constituents. A core sub-network constructed from the ingredient-target protein-pathway network revealed bioactivities including anti-cancer, anti-inflammatory and neuroprotective. The protein association network pointed out the major target proteins viz., THRB, FXR, ALOX15, and TSHR and pathways like metabolic, and neuroactive ligand-receptor interaction pathways of essential oil constituents. The target proteins and pathways provided insights into the mechanism of action of bioactive constituents. Based on the results of the study, geraniol was correlated with neuroprotective, citral to chemo-preventive, and limonene to anti-inflammatory activities. Thus, the study offers a new way for the assessment of the bioactivities of Cymbopogon species essential oils leading to the development of new biomedicines.

To measure serum concentrations of a-MSH, BDNF and AGRP in a group of children with BBS or PWS.

We recruited 12 subjects with PWS, 12 subjects with BBS, 28 obese controls (OC) and 26 lean controls (LC) matched for age, sex and puberty. Serum a-MSH, BDNF and AGRP levels were measured by the ELISA method.

The mean a-MSH level was lower in PWS than those of OC and LC (3729 ± 1319, 5211 ± 829 and 5681 ± 565pg/ml, respectively, p < 0.001), and mean a-MSH was lower in OC than LC (p < 0.05). The mean BDNF level of PWS was higher than those of OC and LC (565 ± 122, 482 ± 102 and 391 ± 74pg/ml, respectively, p < 0.001). On the other hand, mean a-MSH level of BBS was lower than those of OC and LC (4543 ± 658, 5211 ± 829 and 5681 ± 565pg/ml, respectively, p < 0.001), and mean a-MSH was lower in OC than LC (p &lr to detect a-MSH deficiency in individuals suspected to have BBS or PWS, and also in those with POMC, PCSK1, and LEPR deficiency.

Therefore, we designed an Ad5 vector with a specific tropism to CD4 + cells containing an expression system limited to HIV-infected cells. We replaced the knob region of Ad5 fiber protein with the extracellular region of the HIV-1 envelope. We also used a specific Tat-inducible promoter to express two anti-HIV-1 shRNAs. Tropism of recombinant Ad5 was assayed by a comparison of the shRNA expression level in CEM and PBMC cells (as CD4 + cells) and HEK293 cells (as CD4 cells). HIV-1 inhibition was assayed by the determination of p24 antigen in the HIV-infected CEM cells transduced with the recombinant Ad5 vector. New-onset dizziness was reported by 251 (16.6%) patients, among whom 110 (43.8%) complained of lightheadedness, 70 (27.9%) of disequilibrium, 41 (16.3%) of presyncope, and 30 (12%) of vertigo. Whether this holds true taking into account carriage of the HSD17B13TA splice variant, also affecting lipogenesis, and achievement of viral clearance (SVR), is unknown.

PNPLA3, TM6SF2, MBOAT7, GCKR and HSD17B13 variants were determined in a cohort of 328 cirrhotic patients free of HCC before starting treatment with direct acting antivirals (DAA).

SVR in the study cohort was 96%. At the end of follow-up, N = 21 patients had been diagnosed an HCC; none of the genes included in the GRS was individually associated with HCC development. However, in a Cox proportional hazards model, a GRS > 0.457 predicted HCC independently of sex, diabetes, albumin, INR and FIB4. The fit of the model improved adding treatment outcome and carriage of the HSD17B13TA splice variant, with sex, GRS > 0.457, HSD17B13TA splice variant and failure to achieve an SVR (hazard ratio = 6.75, 4.24, 0.24 and 7.7, respectively) being independent predictors of HCC.

However, currently, there are no criteria available for concomitant surgical intervention for an enlarged ascending aorta in infancy. We report a rare case of TOF with pulmonary atresia complicated by severe enlargement of the ascending aorta in a girl aged 2 years and 5 months. We performed the Rastelli procedure with ascending aorta replacement. Pathological findings of the ascending aorta included fragmentation of elastic fibers and loss of smooth muscle cells. Postoperatively, the patient remained healthy and asymptomatic. This case indicates that ascending aortic enlargement in conotruncal anomalies may be associated with pathological lesion even in early childhood. Replacement of the diseased aorta could be considered as a concomitant procedure with definitive repair for conotruncal anomalies presenting with severe ascending aortic enlargement.
