Perturbation in JAK-STAT signaling has been reported in the pathogenesis of cutaneous T cell lymphoma (CTCL). JAK3 is predominantly associated with the intra-cytoplasmic part of IL-2Rγc located in the plasma membrane of hematopoietic cells. Here we demonstrate that JAK3 is also ectopically expressed in the nucleus of malignant T cells. We detected nuclear JAK3 in various CTCL cell lines and primary malignant T cells from patients with Sézary syndrome, a leukemic variant of CTCL. Nuclear localization of JAK3 was independent of its kinase activity whereas STAT3 had a modest effect on nuclear JAK3 expression. Moreover, JAK3 nuclear localization was only weakly affected by blockage of nuclear export. An inhibitor of the nuclear export protein CRM1, Leptomycin B, induced an increased expression of SOCS3 in the nucleus, but only a weak increase in nuclear JAK3. Importantly, immunoprecipitation experiments indicated that JAK3 interacts with the nuclear protein POLR2A, the catalytic subunit of RNA Polymerase II. Kinase assays showed tyrosine phosphorylation of recombinant human Histone H3 by JAK3 in vitro-an effect which was blocked by the JAK inhibitor (Tofacitinib citrate). In conclusion, we provide the first evidence of nuclear localization of JAK3 in malignant T cells. Our findings suggest that JAK3 may have a cytokine-receptor independent function in the nucleus of malignant T cells, and thus a novel non-canonical role in CTCL.Although the hallmarks of Alzheimer's disease (AD) are amyloid beta plaques and neurofibrillary tangles, there is growing evidence that neuroinflammation, mitochondrial dysfunction and oxidative stress play important roles in disease development and progression. A major risk factor for the development of AD is diabetes, which is also characterized by oxidative stress and mitochondrial dysfunction along with chronic, low-grade inflammation. Increasing evidence indicates that in immune cells, the induction of a pro-inflammatory phenotype is associated with a shift from oxidative phosphorylation (OXPHOS) to glycolysis. However, whether hyperglycemia also contributes to this shift is not clear. This raises the need for the healthcare community to proactively address the care needs of older persons with Parkinson's disease. Though it is tempting to categorise different phenotypes of Parkinson's disease, a strong evidence based for the same is lacking. There is considerable literature describing the varying clinical manifestations in old age. This article aims to review the literature looking for strategies in personalising the management of an older person with Parkinson's disease.

This article aims to review the literature looking for strategies in personalising the management of an older person with Parkinson's disease.This study examines the effect of the exogenous application of polyamine putrescine together with the application of different ratios of nitrate/ammonium (NO3-/NH4+), on the physiology of cauliflower subjected to heat stress. The 5050 NO3-/NH4+ ratio was the best ratio against heat stress. As a result of the joint application of these compounds, a higher photosynthetic rate, a higher accumulation of both photosynthesis-related compounds and pigments, total proteins, and a change in the status of nutrients were obtained. Particularly, the decrease in content of calcium, chloride and sulphate in plants under heat stress is ameliorated by the ammonium effect. Additionally, it is important to highlight that cauliflower waste contains a higher content of mineral nutrients than floret cauliflower. These effects were more marked in young leaves. Furthermore, a synergistic effect for coping with heat stress between the polyamine and the nutritional treatment was observed. For this, both the application of putrescine and the feeding of plants with a 5050 NO3-/NH4+ ratio before heat stress is proposed for the first time as an agricultural practice for increasing the thermotolerance of cauliflower cv Moonshine. On the other hand, due to the lower lipid peroxidation rate obtained in cauliflower leaves, these plants could be used for health purposes as ointments or other nutraceutical products, making the cultivation of this kind of cruciferous more sustainable.Limited evidence suggests that serum iron and hepcidin concentrations are dysregulated in obesity and inflammation. The objective of the present study was to compare C-reactive protein, interleukin-6, circulating levels of hepcidin, serum lipids, and iron status in obese vs. normal-weight women of childbearing age. Healthy women aged 18-30 years were recruited for the study (n = 47 25 obese and 22 normal weight). Fasting blood samples were obtained to measure serum lipids (total cholesterol, HDL, LDL cholesterol, triglycerides, non-HDL cholesterol), complete blood count, serum iron, total iron-binding capacity, transferrin saturation, serum ferritin, hepcidin, C-reactive protein, and interleukin-6. Obese women had significantly higher mean serum C-reactive protein (p less then 0.001), interleukin-6 (p less then 0.001), hepcidin (p = 0.024), triglycerides (p less then 0.001) and total cholesterol/HDL ratio (p less then 0.001) but lower HDL (p = 0.001) and serum iron/hepcidin ratio (p = 0.011) compared with normal-weight women.
