Piperlongumine (PL), a major alkaloid in the fruit of Piper longum Linn. has gained worldwide attention for its anticancer properties, however, its mechanism of action in the chemoprevention of colon cancer has not been investigated yet. Therefore, the present study was designed to elucidate the underlying molecular mechanism of PL in preventing DMH/DSS induced experimental colon cancer in mice. In the current study well established DMH/DSS induced experimental colon cancer mouse model was used to demonstrate the chemopreventive potential of PL. The expression of NF-κB and its downstream target proteins was evaluated mainly through western blotting. In addition, CAM assay, immunohistochemical staining and gelatin zymography was used to show anti-angiogenic and anti-invasive potential of PL. Additionally, important tumor biomarkers such as TSA, LASA, LDH and IL-6 levels were also estimated. The results of current study showed that PL was capable to inhibit NF-κB activation as well as its nuclear translocation. PL administration to DMH/DSS treated mice also inhibited the NF-κB downstream signaling cascades such as including COX-2 pathway, JAK/STAT pathway, β-catenin, Notch signaling pathway, angiogenesis and epithelial to mesenchymal transition pathway. The findings of the present study have claimed PL as promising chemopreventive agent for colon cancer with pleiotropic action. The current study emphasizes that regular consumption of PL can be an effective approach in the prevention of colon cancer in humans.Treatment of bone cancer pain (BCP) caused by bone metastasis in advanced cancers remains a challenge in clinical oncology, and the underlying mechanisms of BCP are poorly understood. This study aimed to investigate the pathogenic roles of circular RNAs (circRNAs) in regulating cancer cell proliferation and BCP development. Eight differentially expressed circRNAs in the rat spinal cord were validated by agarose gel electrophoresis and Sanger sequencing. Expression of circRNAs and mRNAs was detected by quantitative RT-PCR. MTS assay and flow cytometry were performed to analyze cell proliferation and apoptosis, respectively. Differentially expressed mRNA profiles were characterized by deep RNA sequencing, hierarchical clustering, and functional categorization. The interactions among circRNAs, microRNAs (miRNAs), and mRNAs were predicted using TargetScan. Additionally, western blot was performed to determine the protein levels of Pax8, Isg15, and Cxcl10. Multiple circRNAs were differentially expressed in the spinal cords of BCP model rats; of these, circSlc7a11 showed the greatest increase in expression. The overexpression of circSlc7a11 significantly promoted cell proliferation and repressed apoptosis of LLC-WRC 256 and UMR-106 cells, whereas circSlc7a11 silencing produced the opposite effects. Altered expression of circSlc7a11 also induced substantial changes in the mRNA expression profiles of LLC-WRC 256 cells; these changes were linked to multiple apoptotic processes and signaling pathways, such as the chemokine signaling pathway, and formed a complex circRNA/miRNA/mRNA network. The results indicated that a temperature of 900 °C in both stages resulted in the highest H2 content (32.4 mol%) in syngas. When different equivalence ratios (ERs) were investigated, it was found that the highest H2 content in syngas (25.4 mol%) was achieved at an ER of 0.3. Our study aims to evaluate air pollution changes, with focus on nitrogen dioxide (NO2), before and during the lockdown in Rome and in the surroundings. Significant NO2 declines were observed during the COVID-19 pandemic with reductions of - 50%, - 34%, and - 20% at urban traffic, urban background, and rural background stations, respectively. Tropospheric NO2 vertical column density (VCD) from the TROPOspheric Monitoring Instrument (TROPOMI) was used to evaluate the spatial-temporal variations of the NO2 before and during the lockdown for the entire area where the surface stations are located. The evaluation is concerned with the pixels including one or more air quality stations to explore the capability of the unprecedented high spatial resolution to monitor urban and rural sites from space with relation to the surface measurements. Good agreement between surface concentration and TROPOMI VCD was obtained in Rome (R = 0.64 in 2019, R = 0.77 in 2020) and in rural sites (R = 0.71 in 2019). Inversely, a slight correlation (R = 0.20) was observed in rural areas during the lockdown due to very low levels of NO2. Finally, the TROPOMI VCD showed a sharp decline in NO2, larger in urban (- 43%) than in rural sites (- 17%) as retrieved with the concurrent surface measurements averaging all the traffic and urban background (- 44%) and all the rural background stations (- 20%). These results suggest air pollution improvement in Rome gained from implementing lockdown restrictions.
