Cetylpyridinium chloride (CPC), a quaternary ammonium compound and cationic surfactant, is used in personal hygiene products such as toothpaste, mouthwash, and nasal spray. Although public exposure to CPC is frequent, its pulmonary toxicity has yet to be fully characterized. Due to high risks of CPC inhalation, we aimed to comprehensively elucidate the in vitro and in vivo toxicity of CPC. The results demonstrated that CPC is highly cytotoxic against the A549 cells with a half-maximal inhibitory concentration (IC50 ) of 5.79 μg/ml. Following CPC exposure, via intratracheal instillation (ITI), leakage of lactate dehydrogenase, a biomarker of cell injury, was significantly increased in all exposure groups. Further, repeated exposure of rats to CPC for 28 days caused a decrease in body weight of the high-exposure group and the relative weights of the lungs and kidneys of the high recovery group, but no changes were evident in the histological and serum chemical analyses. The bronchoalveolar lavage fluid (BALF) analysis showed a significant increase in proinflammatory cytokines interleukin (IL)-6, IL-1β, and tumor necrosis factor (TNF)-α levels. ITI of CPC induced focal inflammation of the pulmonary parenchyma in rats' lungs. Our study demonstrated that TNF-α was the most commonly secreted proinflammatory cytokine during CPC exposure in both in vitro and in vivo models. Polymorphonuclear leukocytes in the BALF, which are indicators of pulmonary inflammation, significantly increased in a concentration-dependent manner in all in vivo studies including the ITI, acute, and subacute inhalation assays, demonstrating that PMNs are the most sensitive parameters of pulmonary toxicity.

Blood clotting disorders and acute respiratory failure have surfaced as the major complications among the severe cases of coronavirus disease 2019 (COVID-19) caused by SARS-CoV-2 infection. Remarkably, more than 70% of deaths related to COVID-19 are attributed to clotting-associated complications such as pulmonary embolism, strokes and multi-organ failure. These vascular complications have been confirmed by autopsy. This study summarizes the current understanding and explains the possible mechanisms of the blood clotting disorder, emphasizing the role of (1) hypoxia-related activation of coagulation factors like tissue factor, a significant player in triggering coagulation cascade, (2) cytokine storm and activation of neutrophils and the release of neutrophil extracellular traps and (3) immobility and ICU related risk factors.We analyze unidirectional and reciprocally connected pairs of neurons in the chemical connectomes of the male and hermaphrodite Caenorhabditis elegans, using recently published data. We further reveal that the C. elegans connectome is wired so that afferent connections onto neurons with large numbers of presynaptic neighbors (in-degree) comprise an above-average number of synapses (synaptic multiplicity). Notably, the larger the in-degree of a neuron the larger the synaptic multiplicity of its afferent connections. Allicin, the main active ingredient of allium plants, has broad-spectrum antifungal activity. However, the unstable properties of allicin limit its wide application. In this study, 1-[(R)-ethylsulfinyl]sulfanylethane, a structurally stable derivative of allicin, was used to explore its antifungal activity and potential mechanism on the expansion of Penicillium expansum. We demonstrated the antifungal activity of 1-[(R)-ethylsulfinyl]sulfanylethane through in vitro antifungal experiments. At the concentration is 2 mg/L, 1-[((R)-ethylsulfinyl]sulfanyl]ethane can completely inhibit spore germination and mycelial growth, whereas the concentration of allicin needs to reach 16 mg/L. Fungal Biochemical assay indicated that decrease of mitochondrial membrane potential, overgeneration of reactive oxygen species, decrease of adenosine triphosphate and glutathione content, increase of superoxide dismutase, catalase, and malondialdehyde content.
