In order to facilitate point-of-care diagnosis, this study aims at presenting a label-free electrochemical biosensor as a powerful nanobiodevice for SARS-CoV-2 spike protein detection. Utilizing the IgG anti-SARS-CoV-2 spike antibody onto the electrode surface as a specific platform in an ordered orientation through staphylococcal protein A (ProtA) is highly significant in fabricating the designed nanobiodevice. In this sense, the screen-printed carbon electrode modified with Cu2O nanocubes (Cu2O NCs), which provide a large surface area in a very small space, was applied in order to increase the ProtA loading on the electrode surface. Accordingly, the sensitivity and stability of the sensing platform significantly increased. The electrochemical evaluations proved that there is a very good linear relationship between the charge transfer resistance (Rct) and spike protein contents via a specific binding reaction in the range 0.25 fg mL-1 to 1 μg mL-1. Moreover, the assay when tested with influenza viruses 1 and 2 was performed in 20 min with a low detection limit of 0.04 fg mL-1 for spike protein without any cross-reactivity. The designed nanobiodevice exhibited an average satisfactory recovery rate of ~ 97-103% in different artificial sample matrices, i.e., saliva, artificial nasal, and universal transport medium (UTM), illustrating its high detection performance and practicability. The nanobiodevice was also tested using real patients and healthy samples, where the results had been already obtained using the standard polymerase chain reaction (PCR) procedure, and showed satisfactory results. Graphical abstract.Apical-basal polarity is a key feature of most epithelial cells and it is regulated by highly conserved protein complexes. In mammalian podocytes, which emerge from columnar epithelial cells, this polarity is preserved and the tight junctions are converted to the slit diaphragms, establishing the filtration barrier. In Drosophila, nephrocytes show several structural and functional similarities with mammalian podocytes and proximal tubular cells. However, in contrast to podocytes, little is known about the role of apical-basal polarity regulators in these cells. In this study, we used expansion microscopy and found the apical polarity determinants of the PAR/aPKC and Crb-complexes to be predominantly targeted to the cell cortex in proximity to the nephrocyte diaphragm, whereas basolateral regulators also accumulate intracellularly. Knockdown of PAR-complex proteins results in severe endocytosis and nephrocyte diaphragm defects, which is due to impaired aPKC recruitment to the plasma membrane. Similar, downregulation of most basolateral polarity regulators disrupts Nephrin localization but had surprisingly divergent effects on endocytosis. Our findings suggest that morphology and slit diaphragm assembly/maintenance of nephrocytes is regulated by classical apical-basal polarity regulators, which have distinct functions in endocytosis.

Conventional anchorage with exclusively intraorally anchored appliances for non-compliance molar distalization combines apalatal acrylic button with periodontal anchorage. This type of anchorage is critically discussed because of the temporary hygienic impairment of the palate and the uncertain anchoring quality of the button. Apurely dentally/periodontally anchored PendulumK appliance was developed, which is exclusively anchored via four occlusal rests. The aims of this pilot study were to examine the suitability of the skeletonized PendulumK for distalization of maxillary molars, and to investigate the quality of this alternative anchoring modality.

In all, 10patients received skeletonized PendulumK appliances attached to all maxillary premolars for bilateral molar distalization. Supporting anchorage through an acrylic button adjacent to the anterior palate was not used. Cells of the strain were coccoid, motile, and strictly aerobic. The strain was able to grow in presence of 2.6-5.4 M NaCl (optimum concentration is 3.4 M) at 30-50 °C (optimum temperature is 42 °C) and pH 7-9.5 (optimum pH is 9.0). The 16S rRNA gene sequence of strain JWXQ-INN-674T showed 95.3-96.6% similarity to members of the genus Natronorubrum of the family Natrialbaceae. The whole genome sequencing of strain JWXQ-INN-674T revealed a genome size of 4.56 M bp and a DNA G + C content of 62.5 mol%. Genome relatedness of strain JWXQ-INN-674T and other species in the genus Natronorubrum was analyzed by average nucleotide identity and digital DNA-DNA hybridization with the values of 76.8-90.6 and 23.1-39.3%, respectively. The strain possessed the polar lipids phosphatidylglycerol and methylated phosphatidylglycerol phosphate lipid. No glycolipids were detected. Based on phylogenetic analysis, phenotypic characteristics, chemotaxonomic properties and genome relatedness, the isolate was proposed as the type strain of a novel species of genus Natronorubrum, Natronorubrum halalkaliphilum sp.
