Novel scientific fields that include laboratory work are perfect playgrounds for developing highly motivating research-based teaching modules. We believe that research-based learning enriched by digital tools is the best approach for teaching new emerging essentials of academic education. This is especially true when the scientific field as such is still not canonized with text books and best-practice examples. Our experience shows that iGEM/BIOMOD competitions represent an excellent basis for designing research-based courses in xenobiology. Therefore, we present a report on "iGEM-Synthetic Biology" offered at the Technische Universität Berlin as an example.Focal adhesion kinase (FAK) is a non-receptor tyrosine kinase located at the extracellular matrix cell adhesion site. This kinase mediates downstream signalling cascades on the cell-extracellular matrix of integrins, cytokine receptors, growth factor receptors and G-protein-coupled receptors. Several studies have suggested the importance of FAK in cancer cell adhesion, motility, proliferation and survival and is over-expressed in cancer cells. There is a growing body of evidence indicating involvement of FAK-mediated signalling and functions in development of tumour cells, making FAK an emerging viable therapeutic target. There is substantial research impetus on development of small molecule FAK inhibitors that impact and inhibit the downstream pathways of FAK, subsequently modulating cancer progression and survival. A variety of scaffolds including hybrid scaffolds have been designed and synthesized with some translating into clinical trials. In addition to the reduction of metastasis and angiogenesis, these inhibitors are effective in inducing tumour cell apoptosis. In this paper, we provide an overview of FAK and analysis of design, synthesis and structure-activity relationship of small molecule FAK inhibitors reported till date. We have discussed FAK inhibitors in clinical trials and highlighted future prospects in the development of FAK inhibitors to augment the armamentarium of cancer therapeutics.

In the AGBRESA study, which assessed artificial gravity through centrifugation as potential countermeasure for space travel, 24 healthy persons (eight women) were submitted to 60day strict -6° head-down-tilt bed rest. Participants were assigned to three groups of eight subjects a control group, continuous artificial gravity training on a short-arm centrifuge (30min/day), or intermittent centrifugation (6×5min/day). We assessed cardiac morphology, function, strain, and haemodynamics by cardiac magnetic resonance imaging (MRI) and echocardiography. We observed no differences between groups and, therefore, conducted a pooled analysis. Consistent with deconditioning, restirain measurements, that appear functional rather than mediated through cardiac remodelling. Thus, strain measurements are of limited utility in assessing influences of physical deconditioning or exercise interventions on cardiac function.A new strategy that utilizes temperature-responsive wax-based Janus particles as microsurfactants to simultaneously achieve enhanced emulsion stability, as well as, on-demand coalescence of emulsion droplets is presented. The dumbbell structure with different surface wetting properties on each side of the Janus particle enables the particles to strongly adsorb at the liquid-liquid interface, leading to excellent stability against coalescence for both water-in-oil (W/O) and oil-in-water (O/W) emulsions. Moreover, these Janus microparticles are composed of a hydrophilic acrylate resin and a hydrophobic wax compartment which transitions from a frozen to an oil-soluble molten state above the melting point. This allows the particle-adsorbed droplets to coalesce above a designated temperature, depending on the type of wax material used. It is envisioned that the excellent emulsion stability and the tunable and rapid response to local temperature enabled by the wax-based Janus particle offers new and exciting opportunities in the advancing technologies including micro-reactors and drug delivery systems to name a few.An improved glucose-chelator-albumin bioconjugate (GluCAB) derivative, GluCAB-2Mal , has been synthesized and studied for in vivo 64 Cu-PET/CT imaging in breast cancer mice models together with its first-generation analogue GluCAB-1Mal . The radioligand works on the principle of tumor targeting through the enhanced permeability and retention (EPR) effect with a supportive role played by glucose metabolism. [64 Cu]Cu-GluCAB-2Mal (99 % RCP) exhibited high serum stability with immediate binding to serum proteins. In vivo experiments for comparison between tumor targeting of [64 Cu]Cu-GluCAB-2Mal and previous-generation [64 Cu]Cu-GluCAB-1Mal encompassed microPET/CT imaging and biodistribution analysis in an allograft E0771 breast cancer mouse model. Tumor uptake of [64 Cu]Cu-GluCAB-2Mal was clearly evident with twice as much accumulation as compared to its predecessor and a tumor/muscle ratio of up to 5 after 24 h. In symptomatic carriers (SCs) we explored Lyonization to explain their symtomatology. From a DNA-Bank of 3,000 samples, intragenic restriction fragment length (RFLPs) and short tandem repeats (STRs) of F8/F9 genes were assessed by PCR-PAGE and GeneScan. In SHA patients, F8 inversions were detected by inverse shifting-PCR/diagnostic and complementary tests. In SCs, we evaluated hemorrhagic symptoms, clotting FVIII/FIX and X-chromosome inactivation (XCI) patterns were assessed by HUMARA assay and the search of XIST promoter pathogenic variants. Informativeness of linkage analysis for HA carrier diagnosis with RFLP's/STR's increased to 74% and reached 80% with five RFLPs for HB. Combined Inv22/Inv1 diagnosed 113 possible carriers, three de novo Inv22-1, and confirmed 45 mothers as obligate or sporadic carriers.
