Although pathogenic fungi are different from animals and plants in the genomic heritability, sequence feature, and epigenetic modification, an increasing number of phytopathogenic fungi have been demonstrated to share DNA methyltransferases (MTases) responsible for DNA methylation with animals and plants. Fungal plant pathogens predominantly possess four types of DNA MTase homologs, including DIM-2, DNMT1, DNMT5, and RID. Numerous studies have indicated that DNA methylation in phytopathogenic fungi mainly distributes in transposable elements (TEs), gene promoter regions, and the repetitive DNA sequences. As an important and heritable epigenetic modification, DNA methylation is associated with silencing of gene expression and transposon, and it is responsible for a wide range of biological phenomena in fungi. This review highlights the relevant reports and insights into the important roles of DNA methylation in the modulation of development, pathogenicity, and secondary metabolism of fungal plant pathogens. Recent evidences prove that there are massive links between DNA and histone methylation in fungi, and they commonly regulate fungal development and mycotoxin biosynthesis.The study aimed to understand the experiences of adolescent smokers who participated in a self-reflection program for smoking cessation and to develop the theoretical basis for constructing similar programs. The participants, ten students from middle and high schools located in cities A and S, were interviewed right after the program ended. Data were collected from August to December 2019 and analyzed using a phenomenological approach to understand participant experiences in depth. The analysis revealed five major themes 'Uniqueness of the Program,' 'Perception of Smoking Cessation,' 'Positive Reflection on Life,' 'Understanding Others,' and 'A Search for Hope and Vision in Life.' The findings revealed that their smoking behaviors were changed through self-reflection and enhancement of self-efficacy and that the program facilitated the formation of identity and vision for the future, which may indirectly strengthen the motivation for adolescent smokers to quit smoking. These findings suggest the need for a smoking cessation program that enhances self-concept and self-esteem. Moreover, it highlights the importance of follow-up research to ensure effectiveness and the need to develop programs with creative content.Combining atomistic and coarse-grained (CG) models is a promising approach for quantitative prediction of polymer properties. However, the gaps between the length and time scales of atomistic and CG models still need to be bridged. Here, the scale gaps of the atomistic model of polyethylene melts, the bead-spring Kremer-Grest model, and dissipative particle dynamics with the slip-spring model were investigated. A single set of spatial and temporal scaling factors was determined between the atomistic model and each CG model. The results of the CG models were rescaled using the set of scaling factors and compared with those of the atomistic model. For each polymer property, a threshold value indicating the onset of static or dynamic universality of polymers was obtained. The scaling factors also revealed the computational efficiency of each CG model with respect to the atomistic model. The performance of the CG models of polymers was systematically evaluated in terms of both the accuracy and computational efficiency.Myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS) is a devastating illness whose biomedical basis is now beginning to be elucidated. We reported previously that, after recovery from frozen storage, lymphocytes (peripheral blood mononuclear cells, PBMCs) from ME/CFS patients die faster in culture medium than those from healthy controls. We also found that lymphoblastoid cell lines (lymphoblasts) derived from these PBMCs exhibit multiple abnormalities in mitochondrial respiratory function and signalling activity by the cellular stress-sensing kinase Target Of Rapamycin Complex 1 (TORC1). These differences were correlated with disease severity, as measured by the Richardson and Lidbury weighted standing test. The clarity of the differences between these cells derived from ME/CFS patient blood and those from healthy controls suggested that they may provide useful biomarkers for ME/CFS. Here, we report a preliminary investigation into that possibility using a variety of analytical classification tools, including linear discriminant analysis, logistic regression and receiver operating characteristic (ROC) curve analysis. We found that results from three different tests-lymphocyte death rate, mitochondrial respiratory function and TORC1 activity-could each individually serve as a biomarker with better than 90% sensitivity but only modest specificity vís a vís healthy controls. However, in combination, they provided a cell-based biomarker with sensitivity and specificity approaching 100% in our sample. This level of sensitivity and specificity was almost equalled by a suggested protocol in which the frozen lymphocyte death rate was used as a highly sensitive test to triage positive samples to the more time consuming and expensive tests measuring lymphoblast respiratory function and TORC1 activity. This protocol provides a promising biomarker that could assist in more rapid and accurate diagnosis of ME/CFS.A simple, sensitive and time-saving differential-pulse adsorptive stripping voltammetric (DPAdSV) procedure using a screen-printed carbon electrode modified with carboxyl functionalized multiwalled carbon nanotubes (SPCE/MWCNTs-COOH) for the determination of diclofenac (DF) is presented. The sensor was characterized using optical profilometry, SEM, and cyclic voltammetry (CV). The use of carboxyl functionalized MWCNTs as a SPCE modifier improved the electron transfer process and the active surface area of sensor. Under optimum conditions, very sensitive results were obtained with a linear range of 0.1-10.0 nmol L-1 and a limit of detection value of 0.028 nmol L-1. The SPCE/MWCNTs-COOH also exhibited satisfactory repeatability, reproducibility, and selectivity towards potential interferences. Moreover, for the first time, the electrochemical sensor allows determining the real concentrations of DF in environmental water samples without sample pretreatment steps.
