tivation time course of the current. Taken together, the compound like CIL, which was tailored to block hyperpolarization-activated cation (HCN) channels effectively, was also capable of altering the amplitude and gating of IK(DR), thereby influencing the functional activities of electrically excitable cells, such as GH3 cells.Apurinic/apyrimidinic endonuclease 1 (APE1), the main mammalian AP-endonuclease for the resolution of DNA damages through the base excision repair (BER) pathway, acts as a multifunctional protein in different key cellular processes. The signals to ensure temporo-spatial regulation of APE1 towards a specific function are still a matter of debate. BAY-3827 in vivo Several studies have suggested that post-translational modifications (PTMs) act as dynamic molecular mechanisms for controlling APE1 functionality. Interestingly, the N-terminal region of APE1 is a disordered portion functioning as an interface for protein binding, as an acceptor site for PTMs and as a target of proteolytic cleavage. We previously demonstrated a cytoplasmic accumulation of truncated APE1 in acute myeloid leukemia (AML) cells in association with a mutated form of nucleophosmin having aberrant cytoplasmic localization (NPM1c+). Here, we mapped the proteolytic sites of APE1 in AML cells at Lys31 and Lys32 and showed that substitution of Lys27, 31, 32 and 35 with alanine impairs proteolysis. We found that the loss of the APE1 N-terminal domain in AML cells is dependent on the proteasome, but not on granzyme A/K as described previously. The present work identified the proteasome as a contributing machinery involved in APE1 cleavage in AML cells, suggesting that acetylation can modulate this process.Ceria (CeO2)-based materials are widely used in applications such as catalysis, fuel cells and oxygen sensors. Its cubic fluorite structure with a cell parameter similar to that of silicon makes it a candidate for implementation in electronic devices. This structure is stable in a wide temperature and pressure range, with a reported structural phase transition to an orthorhombic phase. In this work, we study the structure of CeO2 under hydrostatic pressures up to 110 GPa simultaneously for the nanometer- and micrometer-sized powders as well as for a single crystal, using He as the pressure-transmitting medium. The first-order transition is clearly present for the micrometer-sized and single-crystal samples, while, for the nanometer grain size powder, it is suppressed up to at least 110 GPa. We show that the stacking fault density increases by two orders of magnitude in the studied pressure range and could act as an internal constraint, avoiding the nucleation of the high-pressure phase.Terminal drought is the main stress limiting pea (Pisum sativum L.) grain yield in Mediterranean environments. This study aimed to investigate genotype × environment (GE) interaction patterns, define a genomic selection (GS) model for yield under severe drought based on single nucleotide polymorphism (SNP) markers from genotyping-by-sequencing, and compare GS with phenotypic selection (PS) and marker-assisted selection (MAS). Some 288 lines belonging to three connected RIL populations were evaluated in a managed-stress (MS) environment of Northern Italy, Marchouch (Morocco), and Alger (Algeria). Intra-environment, cross-environment, and cross-population predictive ability were assessed by Ridge Regression best linear unbiased prediction (rrBLUP) and Bayesian Lasso models. GE interaction was particularly large across moderate-stress and severe-stress environments. In proof-of-concept experiments performed in a MS environment, GS models constructed from MS environment and Marchouch data applied to independent material separated top-performing lines from mid- and bottom-performing ones, and produced actual yield gains similar to PS. The latter result would imply somewhat greater GS efficiency when considering same selection costs, in partial agreement with predicted efficiency results. GS, which exploited drought escape and intrinsic drought tolerance, exhibited 18% greater selection efficiency than MAS (albeit with non-significant difference between selections) and moderate to high cross-population predictive ability. GS can be cost-efficient to raise yields under severe drought.Single-cell RNA sequencing is a powerful technology for obtaining transcriptomes at single-cell resolutions. However, it suffers from dropout events (i.e., excess zero counts) since only a small fraction of transcripts get sequenced in each cell during the sequencing process. This inherent sparsity of expression profiles hinders further characterizations at cell/gene-level such as cell type identification and downstream analysis. To alleviate this dropout issue we introduce a network-based method, netImpute, by leveraging the hidden information in gene co-expression networks to recover real signals. netImpute employs Random Walk with Restart (RWR) to adjust the gene expression level in a given cell by borrowing information from its neighbors in a gene co-expression network. Performance evaluation and comparison with existing tools on simulated data and seven real datasets show that netImpute substantially enhances clustering accuracy and data visualization clarity, thanks to its effective treatment of dropouts. While the idea of netImpute is general and can be applied with other types of networks such as cell co-expression network or protein-protein interaction (PPI) network, evaluation results show that gene co-expression network is consistently more beneficial, presumably because PPI network usually lacks cell type context, while cell co-expression network can cause information loss for rare cell types. Evaluation results on several biological datasets show that netImpute can more effectively recover missing transcripts in scRNA-seq data and enhance the identification and visualization of heterogeneous cell types than existing methods.Total hip arthroplasty (THA) and total knee arthroplasty (TKA) represent two of the most common procedures in orthopedic surgery. The growing need to avoid physical impairment in elderly patients undergoing this kind of surgery puts the focus on the possibility to undertake a preoperative physical activity program to improve their fit and physical health at the time of surgery. A systematic review has been carried out with online databases including PubMed-Medline, Cochrane Central and Google Scholar. The aim was to retrieve available evidence concerning preoperative physical activity and exercise, before total knee or total hip arthroplasty in patients older than 65 years, and to clarify the role of this practice in improving postoperative outcomes. Results of the present systematic analysis showed that, for TKA, most of the studies demonstrated a comparable trend of postoperative improvement of Visual Analogue Scale (VAS), range of movement (ROM) and functional scores, and those of quality of life. There is insufficient evidence in the literature to draw final conclusions on the topic.
