The bacterial flagellum is a motility organelle consisting of a long helical filament as a propeller and a rotary motor that drives rapid filament rotation to produce thrust. Salmonella enterica serovar Typhimurium has two genes of flagellin, fljB and fliC, for flagellar filament formation and autonomously switches their expression at a frequency of 10-3-10-4 per cell per generation. We report here differences in their structures and motility functions under high-viscosity conditions. A Salmonella strain expressing FljB showed a higher motility than one expressing FliC under high viscosity. To examine the reasons for this motility difference, we carried out structural analyses of the FljB filament by electron cryomicroscopy and found that the structure was nearly identical to that of the FliC filament except for the position and orientation of the outermost domain D3 of flagellin. The density of domain D3 was much lower in FljB than FliC, suggesting that domain D3 of FljB is more flexible and mobile than that of FliC. These differences suggest that domain D3 plays an important role not only in changing antigenicity of the filament but also in optimizing motility function of the filament as a propeller under different conditions.Chronic inflammation, a pervasive feature of the aging process, is defined by a continuous, multifarious, low-grade inflammatory response. It is a sustained and systemic phenomenon that aggravates aging and can lead to age-related chronic diseases. In recent years, our understanding of age-related chronic inflammation has advanced through a large number of investigations on aging and calorie restriction (CR). A broader view of age-related inflammation is the concept of senoinflammation, which has an outlook beyond the traditional view, as proposed in our previous work. In this review, we discuss the effects of CR on multiple phases of proinflammatory networks and inflammatory signaling pathways to elucidate the basic mechanism underlying aging. Based on studies on senoinflammation and CR, we recognized that senescence-associated secretory phenotype (SASP), which mainly comprises cytokines and chemokines, was significantly increased during aging, whereas it was suppressed during CR. Further, we recognized that cellular metabolic pathways were also dysregulated in aging; however, CR mimetics reversed these effects. These results further support and enhance our understanding of the novel concept of senoinflammation, which is related to the metabolic changes that occur in the aging process. Furthermore, a thorough elucidation of the effect of CR on senoinflammation will reveal key insights and allow possible interventions in aging mechanisms, thus contributing to the development of new therapies focused on improving health and longevity.The emergence and spread of mobile colistin (COL) resistance (mcr) genes jeopardize the efficacy of COL, a last resort antibiotic for treating deadly infections. COL has been used in livestock for decades globally. Bacteria have mobilized mcr genes (mcr-1 to mcr-9). Mcr-gene-containing bacteria (MGCB) have disseminated by horizontal/lateral transfer into diverse ecosystems, including aquatic, soil, botanical, wildlife, animal environment, and public places. selleck inhibitor The mcr-1, mcr-2, mcr-3, mcr-5, mcr-7, and mcr-8 have been detected in isolates from and/or directly in environmental samples. These genes are harboured by Escherichia coli, Enterobacter, Klebsiella, Proteus, Salmonella, Citrobacter, Pseudomonas, Acinetobacter, Kluyvera, Aeromonas, Providencia, and Raulotella isolates. Different conjugative and non-conjugative plasmids form the backbones for mcr in these isolates, but mcr have also been integrated into the chromosome of some strains. Contact with the mcr-containing reservoirs, consumption of contaminated animal-/plant-based foods or water, international animal-/plant-based food trades and travel, are routes for transmission of MGCB.The natural deep eutectic solvent (NADES) based on choline chloride (ChCl) and lactic acid (LA) was used for the preparation of chitosan (CS) films by the solution casting method. The content of NADES in films was from 0 to 82 wt%. The impact of NADES on the morphology and crystalline structure of films was investigated using scanning electron microscopy as well as wide-angle and small-angle X-ray scattering. The experimental results allow to propose CS chains swelling in NADES. FTIR spectroscopy confirms the interactions between CS and NADES components via the formation of hydrogen and ion bonds. The thermal properties of the composite films were studied by simultaneous thermogravimetric and differential thermal analysis. Thermomechanical analysis demonstrated appearance of two transitions at temperatures between -23 and -5 °C and 54-102 °C depending on NADES content. It was found that electrical conductivity of film with 82 wt% of NADES reaches 1.7 mS/cm. The aim of this study was to compare the acute physiological, metabolic and perceptual responses to two different SIE cycling protocols with different recovery durations. Methods Twelve healthy, active male participants took part in this study and completed four testing sessions in the laboratory separated by a minimum of 72h. Two SIE protocols were applied in randomized order SIE6×10"/4'-six "all-out" repeated 10-s bouts, interspersed with 4-min recovery; and SIESERIES-two series of three "all-out" repeated 10-s bouts, separated by 30-s recovery and 18-min recovery between series. Protocols were matched for the total work time (1 min) and recovery (20 min). Results In SIESERIES, peak oxygen uptake and peak heart rate were significantly higher (p less then 0.05), without differences in peak blood lactate concentration and mean rating of perceived exertion compared to SIE6×10"/4'. There were no differences in peak power output, peak oxygen uptake and peak heart rate between both series in SIESERIES.
