The distribution of DEGs among three comparison groups was similar, and many DEGs were mapped to the phagosome pathway, indicating that eyestalk ablation triggers activation of the neuroendocrine-immune (NEI) system. Interestingly, several important pathways were uniquely enriched in the brain tissue, suggesting that the brain may play a crucial role in the NEI system in response to eyestalk ablation. This is the first report on the transcriptomic regulation of the nervous system in response to eyestalk ablation in L. vannamei. The genes and pathways identified in this study will help to elucidate the molecular mechanisms of neuroendocrine-immune responses to eyestalk ablation in penaeid shrimp.The transcriptome of the caenogastropod mollusk Littorina littorea was scanned for the presence of sequences encoding Toll-like receptors (TLRs) and corresponding proteins involved in downstream TLR signaling pathway. In the transcriptomic snapshots of hemocytes and kidney tissues, 45 complete TLRs encoded by 35 genes were identified. Out of the 59 non-TLR molecules involved in a canonical TLR signaling pathway, 35 genes were classified as homologous and could be placed within the TLR-mediated MyD88-and MAPK-dependent circuitries. No reference vertebrate adapters TIRAP, TRIF and TRAM were identified in the transcriptome. The results of RNA-seq experiments with an immune challenge (rediae of the digenean Himasthla elongata) indicate that four TLRs (LlTLR1, 3, 5 and 8) and a set of upregulated genes involved in signal transduction (LlMyd88, LlTNFα, LlCASP8, LlFADD, LlNFKBIA (IkBα), LlIRAK1, LlSTAT1, LlMAPK14 (P38), LlMAP2K1 (MEK1/2), LlIRF3 and LlIRF5) may participate in the anti-digenean immune response of L. littorea.The presence of diclofenac in the aquatic environment and the risks for aquatic wildlife, especially fish, have been raised in several studies. One way to manage risks without enforcing improved wastewater treatment would be to substitute diclofenac (when suitable from a clinical perspective) with another non-steroidal anti-inflammatory drug (NSAID) associated with less environmental risk. While there are many ecotoxicity-studies of different NSAIDs, they vary extensively in set-up, species studied, endpoints and reporting format, making direct comparisons difficult. We previously published a comprehensive study on the effects of diclofenac in the three-spined stickleback (Gasterosteus aculeatus). Our present aim was to generate relevant effect data for another NSAID (naproxen) using a very similar setup, which also allowed direct comparisons with diclofenac regarding hazards and risks. Sticklebacks were therefore exposed to naproxen in flow-through systems for 27 days. Triplicate aquaria with 20 fish per aquer than expected in surface waters. Naproxen bioconcentrated less than diclofenac, in line with the observed effect data. An analysis of recent sales data and reported concentrations in treated sewage effluent in Sweden suggest that despite higher dosages used for naproxen, a complete substitution would only be expected to double naproxen emissions. In summary, naproxen and diclofenac produce highly similar effects in fish but the environmental hazards and risks are clearly lower for naproxen. Hence, if there are concerns for environmental risks to fish with diclofenac, a substitution would be advisable when naproxen presents an adequate alternative from a clinical point-of-view.
Simultaneous multi-slice (SMS) imaging accelerates MRI data acquisition by exciting multiple image slices with a single radiofrequency pulse. Overlapping slices encoded in acquired signal are separated using a mathematical model, which requires estimation of image reconstruction kernels using calibration data. Several parameters used in SMS reconstruction impact the quality and fidelity of final images. Therefore, finding an optimal set of reconstruction parameters is critical to ensure that accelerated acquisition does not significantly degrade resulting image quality.
Gradient-echo echo planar imaging data were acquired with a range of SMS acceleration factors from a cohort of five volunteers with no known neurological pathology. Images were collected using two available phased-array head coils (a 48-channel array and a reduced diameter 32-channel array) that support SMS. Data from these coils were identically reconstructed offline using a range of coil compression factors and reconstruction kernel paraing algorithm, coil compression caused small reduction in correlation between single-band and SMS unaliased images. Similarly, group analysis of fMRI results did not show a significant influence of coil compression on resulting image quality.
This study demonstrated that the hyperparameters used in SMS reconstruction need to be fine-tuned once the experimental factors such as the RF receive coil and SMS factor have been determined. A cursory evaluation of SMS reconstruction hyperparameter values is therefore recommended before conducting a full-scale quantitative study using SMS technologies.

It is caused by biallelic mutations in the FAM20A gene. Herein, we report two unrelated patients with ERS. Our patients presented with the characteristic features of the syndrome, and amelogenesis imperfecta and gingival hyperplasia were the main complaint. Strikingly, they both had long face, thick lips, notched upper central incisors, and thick alveolar ridge which have never been reported before in patients with ERS. Gingival biopsy showed psammomatous calcifications, and renal ultrasound revealed bilateral nephrocalcinosis in the two patients. SB525334 Mutational analysis of the FAM20A gene identified two homozygous mutations including a novel one (c.915_918delCTTT, p.Phe305Leufs*76 and c.1219 + 3_1219+6delAGGT). Our data expand the phenotypic and mutational spectrum of FAM20A gene and reinforce the importance of kidney examination and follow up for all patients with amelogenesis imperfecta unless FAM20A mutations were ruled out.
