Enhancers located in introns are abundant and play a major role in the regulation of gene expression in mammalian species. By contrast, the functions of intronic enhancers in plants have largely been unexplored and only a handful of plant intronic enhancers have been reported and characterized. We performed a genome-wide prediction of intronic enhancers in Arabidopsis thaliana using open chromatin signatures based on DNase I sequencing. We identified 941 candidate intronic enhancers associated with 806 genes in seedling tissue and 1,271 intronic enhancers associated with 1,069 genes in floral tissue. We validated the function of 15 of 21 (71%) of the predicted intronic enhancers in transgenic assays using a reporter gene. We also created deletion lines of three intronic enhancers associated with two different genes using CRISPR/Cas. Deletion of these enhancers, which span key transcription factor binding sites, did not abolish gene expression but caused varying levels of transcriptional repression of their cognate genes. Remarkably, the transcriptional repression of the deletion lines occurred at specific developmental stages and resulted in distinct phenotypic effects on plant morphology and development. Clearly, these three intronic enhancers are important in fine-tuning tissue- and development-specific expression of their cognate genes.
Metabolic and endocrine alterations in women with polycystic ovary syndrome (PCOS) affect adipose tissue mass and distribution. PCOS is characterised by hyperandrogenism, obesity and adipocyte dysfunction. Hyperandrogenism in PCOS drives dysfunctional adipocyte secretion of potentially harmful adipocytokines. Glucocorticoids and sex-steroids modulate adipocyte development and function. For their part, adipocyte products interact with adrenal and ovarian steroidogenic cells. Currently, the relationship between adipocyte and steroidogenic cells is not clear, and for these reasons, it is important to elucidate the interrelationship between these cells in women with and without PCOS.
This comprehensive review aims to assess current knowledge regarding the interrelationship between adipocytes and adrenal and ovarian steroidogenic cells in animal models and humans with or without PCOS.
We searched for articles published in English and Portuguese in PubMed. Keywords were as follows polycystic ovary syndrome, sal harmful sequelae of PCOS in humans. Further studies are required to investigate the possible therapeutic application of adipokines in women with obese and non-obese PCOS. Meanwhile, when appropriate, metformin use alone, or associated with flutamide, may be considered for therapeutic purposes.Spotted owls (SOs, Strix occidentalis) are a flagship species inhabiting old-growth forests in western North America. In recent decades, their populations have declined due to ongoing reductions in suitable habitat caused by logging, wildfires, and competition with the congeneric barred owl (BO, Strix varia). The northern spotted owl (S. o. caurina) has been listed as "threatened" under the Endangered Species Act since 1990. Here, we use an updated SO genome assembly along with 51 high-coverage whole-genome sequences to examine population structure, hybridization, and recent changes in population size in SO and BO. We found that potential hybrids identified from intermediate plumage morphology were a mixture of pure BO, F1 hybrids, and F1 × BO backcrosses. Also, although SO underwent a population bottleneck around the time of the Pleistocene-Holocene transition, their population sizes rebounded and show no evidence of any historical (i.e., 100-10,000 years ago) population decline. This suggests that the current decrease in SO abundance is due to events in the past century. Although this result is surprising, it is unclear where the ancestors of western BO lived after the separation. In this study, fumigant toxicity and sublethal effects of Eucalyptus camaldulensis Dehnh. (Myrtaceae) and Mentha piperita L. (Lamiaceae) essential oils (EOs) were investigated against different growth stages of T. granarium. To assess the sublethal effects, insects were exposed to an LC20 or LC50 concentration of each essential oil, and the ability of these oils to deter feeding, oviposition, and damage to wheat seeds and overall mass were surveyed. At LC50 concentrations, M. piperita EO showed higher fumigant toxicity than E. camaldulensis EO against eggs, 2nd instar larvae, 4th instar larvae, and adults of T. granarium. Furthermore, the adults were more sensitive to the tested EOs than immatures. In free-choice tests, both larvae and adults showed a preference for control-treated wheat seeds than for seeds treated with an LC20 or LC50 concentration of EOs from E. camaldulensis or M. piperita. In a no-choice test, adult females exposed to EOs showed lower fecundity and fertility in comparison to control females not exposed to EOs. Treatment of wheat seeds with E. camaldulensis or M. piperita EOs resulted in a dose-specific reduction in the number of damaged seeds and seed weight loss when compared to control. According to our results, both tested EOs, especially EOs extracted from M. piperita, showed good potential for use in integrated pest management strategies against T. granarium.In October 2020, three captive male white-tailed deer, Odocoileus virginianus [Zimmermann] (artiodactyla Cervidae), were found dead in central Pennsylvania and a fourth was euthanized due to extreme lethargy. The deer presented with high burdens of Dermacentor albipictus (Packard) (Ixoda Ixodidae) (winter tick). There were no other clinical symptoms and deer were in otherwise good physical condition with no observed alopecia. Winter tick epizootics have been associated with mortalities of moose, Alces alces [Linnaeus] (artiodactyla cervidae), and more recently elk, Cervus canadensis [Erxleben] (artiodactyla cervidae), in Pennsylvania, but have not been reported in white-tailed deer. Mild winters are favorable to winter ticks and deer producers and managers should be aware of possible infestations as a result.
