In this work, a combination of molecular dynamics, targeted mutagenesis, fluorescent Ca2+ imaging and patch-clamp electrophysiology has been applied to G153S mutant muscle nAChR to investigate the role of hydrogen bonds formed by Ser 153 with C-loop residues near the acetylcholine-binding site. Introduction of L199T mutation to the C-loop in the vicinity of Ser 153 changed hydrogen bonds distribution, decreased acetylcholine potency (EC50 2607 vs. 146 nM) of the double mutant and decay kinetics of acetylcholine-evoked cytoplasmic Ca2+ rise (τ 14.2 ± 0.3 vs. 34.0 ± 0.4 s). These results shed light on molecular mechanisms of nAChR activation-desensitization and on the involvement of such mechanisms in channelopathy genesis.Fusarium genus comprises important saprophytic and phytopathogenic fungi and is widespread in nature. The present study reports the occurrence of Fusarium spp. in soils from two mangrove forests in northern Peninsular Malaysia and analyzed physico-chemical properties of the mangrove soil. Based on TEF-1α sequences, nine Fusarium species were identified Fusarium solani species complex (FSSC) (n = 77), Fusarium verticillioides (n = 20), Fusarium incarnatum (n = 10), Fusarium proliferatum (n = 7), Fusarium lateritium (n = 4), Fusarium oxysporum (n = 3), Fusarium rigidiuscula (n = 2), Fusarium chlamydosporum (n = 1), and Fusarium camptoceras (n = 1); FSSC isolates were the most prevalent. Phylogenetic analysis of the combined TEF-1α and ITS sequences revealed diverse phylogenetic affinities among the FSSC isolates and potentially new phylogenetic clades of FSSC. Soil analysis showed varied carbon content, pH, soil moisture, and salinity, but not nitrogen content, between sampling locations. Regardless of the physico-chemical properties, various Fusarium species were recovered from the mangrove soils. These were likely saprophytes; however, some were well-known plant pathogens and opportunistic human pathogens. Thus, mangrove soils might serve as inoculum sources for plant and human pathogenic Fusarium species. To allow clinicians to adapt treatment plans for patients, Ontario Health (Cancer Care Ontario) issued a series of interim funding measures for the province's New Drug Funding Program (NDFP), which covers the cost of most hospital-delivered cancer drugs. To assess the utility of the measures and the need for their continuation, we conducted an online survey of Ontario oncology clinicians. The survey was open 3-25 September 2020 and generated 105 responses. Between April and June 2020, 46% of respondents changed treatment plans for more than 25% of their cancer patients due to the pandemic. Clinicians report broad use of interim funding measures. The most frequently reported strategies used were treatment breaks for stable patients (62%), extending dosing intervals (59%), and deferring routine imaging (56%). Most clinicians anticipate continuing to use these interim funding measures in the coming months. The survey showed that adapting cancer drug funding policies has supported clinical care in Ontario during the pandemic.In the paper, the thermal compensation loops on a composite, superconducting NbTi cable were investigated. This type of cable is used in the superconducting, fast ramping magnets of the SIS100 synchrotron, part of the Facility for Antiproton and Ion Research (FAIR) under construction in Darmstadt, Germany. The influence of space restrictions and electromagnetic cross-talk on the design of the thermal compensation loop was discussed. Plastic deformation of cable components during bending was analyzed by numerical simulations and experiments. A three-dimensional numerical model of the cable was prepared with individual superconducting wires in contact with a central cooling pipe. The bending of a straight cable into a compensation loop shape was simulated, followed by cyclic operation of the cable during thermal cycles. The maximum strains in the superconducting strands and cooling tube were analyzed and discussed.Forward osmosis (FO) modules currently suffer from performance efficiency limitations due to concentration polarisation (CP), as well as pressure drops during operation. There are incentives to further reduce CP effects, as well as optimise spacer design for pressure drop improvements and mechanical support. In this study, the effects of applying transmembrane pressure (TMP) on FO membrane deformation and the subsequent impact on module performance was investigated by comparing experimental data to 3D computational fluid dynamics (CFD) simulations for three commercial FO modules. At a TMP of 1.5 bar the occlusion of the draw-channel induced by longitudinal pressure hydraulic drop was comparable for the Toray (16%) and HTI modules (12%); however, the hydraulic perimeter of the Profiera module was reduced by 46%. CFD simulation of the occluded channels indicated that a change in hydraulic perimeter due to a 62% increase in shear strain resulted in a 31% increase in the Reynolds number. This reduction in channel dimensions enhanced osmotic efficiency by reducing CP via improved draw-channel hydrodynamics, which significantly disrupted the external concentration polarization (ECP) layer.
