These observations bring new insights into the molecular mechanisms of heterosis during the cabbage head development.MicroRNAs (miRNAs or miRs) have a critical role in regulating stem cells (SCs) during development and altered expression can cause developmental defects and/or disease. Indeed, aberrant miRNA expression leads to wide-spread transcriptional dysregulation which has been linked to many cancers. Mounting evidence also indicates a role for miRNAs in the development of the cancer SC (CSC) phenotype. Our goal herein is to provide a review of (i) current research on miRNAs and their targets in colorectal cancer (CRC), and (ii) miRNAs that are differentially expressed in colon CSCs. MicroRNAs can work in clusters or alone when targeting different SC genes to influence CSC phenotype. Accordingly, we discuss the specific miRNA cluster classifications and isomiRs that are predicted to target the ALDH1, CD166, BMI1, LRIG1, and LGR5 SC genes. miR-23b and miR-92A are of particular interest because our previously reported studies on miRNA expression in isolated normal versus malignant human colonic SCs showed that miR-23b and miR-92a are regulators of the LGR5 and LRIG1 SC genes, respectively. We also identify additional miRNAs whose expression inversely correlated with mRNA levels of their target genes and associated with CRC patient survival. Altogether, our deliberation on miRNAs, their clusters, and isomiRs in regulation of SC genes could provide insight into how dysregulation of miRNAs leads to the emergence of different CSC populations and SC overpopulation in CRC.Bryophytes are widely used to monitor air quality. Due to the lack of a cuticle, their cells can be compared to the roots of crop plants. This study aimed to test a hypothetical relation between metal tolerance and cell shape in biomonitoring mosses (Hypnum cupressiforme, Pleurozium schreberi, Pseudoscleropodium purum) and metal sensitive species (Physcomitrium patens, Plagiomnium affine). The tolerance experiments were conducted on leafy gametophytes exposed to solutions of ZnSO4, ZnCl2, and FeSO4 in graded concentrations of 1 M to 10-8 M. Plasmolysis in D-mannitol (0.8 M) was used as a viability measure. The selected species differed significantly in lamina cell shape, cell wall thickness, and metal tolerance. In those tested mosses, the lamina cell shape correlated significantly with the heavy metal tolerance, and we found differences for ZnSO4 and ZnCl2. Biomonitoring species with long and thin cells proved more tolerant than species with isodiametric cells. For the latter, "death zones" at intermediate metal concentrations were found upon exposure to ZnSO4. Species with a greater tolerance towards FeSO4 and ZnSO4 had thicker cell walls than less tolerant species. Hence, cell shape as a protoplast-to-wall ratio, in combination with cell wall thickness, could be a good marker for metal tolerance.Additively-manufactured Ti-6Al-4V (Ti64) exhibits high strength but in some cases inferior elongation to those of conventionally manufactured materials. CDK inhibitors in clinical trials Post-processing of additively manufactured Ti64 components is investigated to modify the mechanical properties for specific applications while still utilizing the benefits of the additive manufacturing process. The mechanical properties and fatigue resistance of Ti64 samples made by electron beam melting were tested in the as-built state. Several heat treatments (up to 1000 °C) were performed to study their effect on the microstructure and mechanical properties. Phase content during heating was tested with high reliability by neutron diffraction at Los Alamos National Laboratory. Two different hot isostatic pressings (HIP) cycles were tested, one at low temperature (780 °C), the other is at the standard temperature (920 °C). The results show that lowering the HIP holding temperature retains the fine microstructure (~1% β phase) and the 0.2% proof stress of the as-built samples (1038 MPa), but gives rise to higher elongation (~14%) and better fatigue life. The material subjected to a higher HIP temperature had a coarser microstructure, more residual β phase (~2% difference), displayed slightly lower Vickers hardness (~15 HV10N), 0.2% proof stress (~60 MPa) and ultimate stresses (~40 MPa) than the material HIP'ed at 780 °C, but had superior elongation (~6%) and fatigue resistance. Heat treatment at 1000 °C entirely altered the microstructure (~7% β phase), yield elongation of 13.7% but decrease the 0.2% proof-stress to 927 MPa. The results of the HIP at 780 °C imply it would be beneficial to lower the standard ASTM HIP temperature for Ti6Al4V additively manufactured by electron beam melting.Cisplatin is the most widely used chemotherapeutic agent. However, it often causes nephrotoxicity, which results in acute kidney injury (AKI). Therefore, we urgently need a drug that can reduce the nephrotoxicity induced by cisplatin. Loganin is a major iridoid glycoside isolated from Corni fructus that has been used as an anti-inflammatory agent in various pathological models. However, the renal protective activity of loganin remains unclear. In this study, to examine the protective effect of loganin on cisplatin-induced AKI, male C57BL/6 mice were orally administered with loganin (1, 10, and 20 mg/kg) 1 h before intraperitoneal injection of cisplatin (10 mg/kg) and sacrificed at three days after the injection. The administration of loganin inhibited the elevation of blood urea nitrogen (BUN) and creatinine (CREA) in serum, which are used as biomarkers of AKI. Moreover, histological kidney injury, proximal tubule damages, and renal cell death, such as apoptosis and ferroptosis, were reduced by loganin treatment. Also, pro-inflammatory cytokines, such as interleukin (IL)-1β, IL-6, and tumor necrosis factor (TNF)-α, reduced by loganin treatment. Furthermore, loganin deactivated the extracellular signal-regulated kinases (ERK) 1 and 2 during AKI. Taken together, our results suggest that loganin may attenuate cisplatin-induced AKI through the inhibition of ERK1/2.Owing to its low flame temperature and high airflow velocity, the activated combustion high-velocity air-fuel (AC-HVAF) spraying process has garnered considerable attention in recent years. Analyzing the velocity field, temperature field, and composition of AC-HVAF spray coatings plays a vital role in improving the quality of coatings. In this study, an actual spray gun is adopted as a prototype, and the radial air inlets are introduced to improve the reaction efficiency so that the chemical reaction can be completed in the combustion chamber. Furthermore, a complete three-dimensional (3D) model is established to examine the effects of radial inlets and porous ceramic sheet on the combustion and flow fields. The hexahedral cells are used to discretize the entire model for reducing the influence of false-diffusion on the calculation results. The gas flow field is simulated by the commercial Fluent software, and the results indicate that the porous ceramic sheet effectively reduces the turbulent dissipation of the airflow with a good rectification effect (the ceramic sheet ensures a consistent airflow direction).
