Objective The COVID-19 pandemic seems to be developing into a planetary-scale mental health experiment, undermining the foundations of our being human, obliging us to keep physically apart from one another, and inducing us to see other people as a potential threat. Methods In the world of pediatric oncology, we have found ourselves up against new challenges. In this article, we discuss the difficulties found in conducting psychological support sessions while complying with physical distancing rules, as well as wearing protective face masks, and even gloves; or while using modern remote communication telecommunications. Results The classic reasons behind requests for psychological support have been compounded by other, novel problems, such as fewer relational resources for families at home, less chance to socialize, hospital stays with only one parent, the suspension of all teaching and group activities on the ward, economic difficulties caused by the pandemic, the rising levels of family conflictuality and generalized anxiety. Conclusions It is essential to find new solutions that can be promptly implemented, reconfiguring the way we humanize our hospital wards.Sorghum (Sorghum bicolor) fulfills the demand for bioenergy resources and also provides substantial diet calories to the world's population. Therefore, many biological studies use sorghum as a research model for improvement of the domesticated food and bioenergy crops. Furthermore, leveraging genome editing systems in a plethora of grass plant species has been extensively studied with no exception in sorghum. However, a protocol that details the genome editing strategies using CRISPR/Cas9 and that combines an efficient tissue culture and transformation platform in sorghum based on Agrobacterium-mediated DNA transfer has yet to be reported. Basic Protocol 1 Construction of CRISPR/Cas9 expression vector to analysis of CRISPR-edited plants Basic Protocol 2 Stable transformation of sorghum Support Protocol Management of sorghum plants in a greenhouse.Objective To determine the incidence of extra-pulmonary tuberculosis (EPTB) and examine the risk factors and the clinical features of the disease over a ten-year period. Methods Retrospective study of records of patients who were followed and registered in the TB registry programme in the health district of Gorgan, Iran from January 1, 2008, through December 31, 2017. Results Among 2280 TB records, 609 (26.71%) were EPTB. They were mostly female patients (53.7%) and residents in rural areas (56.5%) with a mean age of 40.55 years [±16]. The average age of female patients (37.55 years [±16.99]) was lower than of male patients (44.07 years [±20.59]). The median of the incidence rate was 7.5 per 100 000 inhabitants for EPTB; biopsy and pathology were the best methods for the detection of EPTB. The most frequent forms of EPTB were lymphatic TB (193/609 = 31.7%) and pleural TB (158/609 = 25.9%). In most cases (245/609 = 40.2%), one to three months elapsed between occurrence of symptoms and final confirmation of EPTB. The outcome of EPTB was weaker than of pulmonary TB (PTB). Conclusion Our most important finding was the increasing incidence of EPTB, which shows the importance of attention to this disease. Lymph node and pleural tissue were the most commonly infected tissues. Skeletal TB presents a challenge in the diagnosis and treatment of EPTB.A series of cyclic active-site-directed inhibitors of the NS2B-NS3 proteases from Zika (ZIKV), West Nile (WNV), and dengue-4 (DENV4) viruses has been designed. The most potent compounds contain a reversely incorporated d-lysine residue in the P1 position. Its side chain is connected to the P2 backbone, its α-amino group is converted into a guanidine to interact with the conserved Asp129 side chain in the S1 pocket, and its C terminus is connected to the P3 residue via different linker segments. The most potent compounds inhibit the ZIKV protease with Ki values less then 5 nM. Crystal structures of seven ZIKV protease inhibitor complexes were determined to support the inhibitor design. All the cyclic compounds possess high selectivity against trypsin-like serine proteases and furin-like proprotein convertases. Both WNV and DENV4 proteases are inhibited less efficiently. Nonetheless, similar structure-activity relationships were observed for these enzymes, thus suggesting their potential application as pan-flaviviral protease inhibitors.Objective Lower digit ratios between the lengths of fingers 2 (2D) and 4 (4D) (2D4D) are associated with superior distance running and athletic performance. We examined relationships between 2D4D, aerobic fitness, physical skills, and overall physical fitness of elite adolescent boy and girl distance runners. Methods Subjects were top five finishers for their sex and age in 10 or more races of 10 km or longer in Michigan in 1981. We calculated 2D4D of 15 girls and 11 boys from radiographs. Subject peak O2 consumption (VO2Peak ), ventilatory threshold (VT), and point of equivalent change (PEC) were collected during intermittent treadmill protocol tests. Performances on physical skills tests (flex-arm hang, broad jump, vertical jump, figure-8-run, sit ups, and sit-and-reach test) were collected in the laboratory. We examined the interrelationships between 2D4D, subject sex, aerobic fitness, physical skills test performance, and overall physical fitness, a composite of aerobic and physical skills performance with correlation, linear regression, t tests, and principle component analyses. Results Girls had significantly larger right hand (R) 2D4D than boys. Boys had greater VO2Peak by mass than girls. Boys with lower R2D4D had significantly greater VO2Peak and PEC. Girls with lower R2D4D had significantly greater VT. Factors associated with aerobic fitness explained most of the variation in composite physical fitness scores. Composite aerobic fitness, physical skills, and overall physical fitness scores of boys were negatively correlated with R2D4D. Solutol HS-15 Conclusions These data suggest that R2D4D may help predict distance running performance in girls and boys and overall physical fitness in boys and provide additional insights into the innate factors influencing youth physical fitness.
