In this review, we summarize the current knowledge on the origin and evolution of SARS-CoV-2 and the roles of key viral factors. We discuss the structure of RNA-dependent RNA polymerase (RdRp) of SARS-CoV-2 and its significance in drug discovery and explain the receptor recognition mechanisms of coronaviruses. Further, we provide a comparative analysis of the SARS-CoV and SARS-CoV-2 S proteins and their receptor-binding specificity and discuss the differences in their antigenicity based on biophysical and structural characteristics.Root treatment with oxathiapiprolin, benthiavalicarb or their mixture Zorvec-Endavia [ZE (3+7, w/w)] was shown to provide prolonged systemic protection against foliar oomycete pathogens attacking cucumber, tomato and basil. Here we report that these fungicides can effectively protect potato plants against late blight when applied to the soil in which such potato plants are grown. In two field experiments, performed in 2019 and 2020, potato plants grown in 64 L containers were treated with a soil drench of oxathiapiprolin, benthiavalicarb or ZE at 12.5, 25 or 50 mg ai/five plants in a container. Artificial inoculations with Phytophthora infestans revealed that such treated plants were protected against late blight in a dose-dependent manner all along the season. Interestingly, oxathiapiprolin persisted in the treated soil for at least 139 days, providing systemic protection against late blight to the following potato crops grown in that treated soils. Potato plants grown in loess soil in the field were either sprayed or drenched with ZE. Plants treated via the soil were significantly better protected against late blight compared to the plants treated by a spray. The data demonstrate a new strategy for season-long protection of potato against late blight by a single soil application of ZE. The systemic nature of oxathiapiprolin and benthiavalicarb composing ZE assures the translocation to the foliage of two fungicides with different modes of action. This shall minimize the risk of developing resistance against either fungicide in the treated crops.Besides understanding the effects of fishing on harvested fish stocks, effects on non-target species, habitats and seafloor integrity also need to be considered. Static fishing gears have often been mentioned as a lower impact fishing alternative to towed gears, although studies examining their actual impact on the seafloor are scarce. In this study, we aimed to describe fish trap movements on the seafloor related to soaking time and trap retrieval. Impacts on the seafloor of lightweight rectangular traps and heavier circular traps were compared. We used 3D video cameras to estimate sweeping motion on the seabed and penetration into the sediment during soaking time. The area and distance swept by each type of trap during retrieval was determined by a camera set up facing the sea bottom. The potential rotation of the traps around the mainline was assessed using an Acoustic Doppler Current Profiler. Results showed that no penetration and almost no movements could be detected during soaking time for either lightweight or heavy commercial traps, even for high tidal coefficient (maximum 6 cm). No rotation could be observed when the tide turned. The swept area covered by a trap during retrieval was low (maximum 2.04 m2) compared to towed fishing gear and other static gear.Based on the engineering background in which the rock surrounding a wellbore is affected by a thermal shock, impact disturbances from drilling vibration, cyclic heat extraction and high temperature during hydrothermal geothermal energy mining, the environmental conditions in the shaft wall rock are simulated by means of high temperature, cooling, immersing granite in water with different curing temperatures and applying impact loads. Additionally, an experimental study on the mechanical characteristics of circular granite specimens under radial impact loads and in the heat treatment and water curing conditions is carried out. The results show that the inner diameters of the rings, heating temperatures, curing water temperatures and cycle heating times are less affected than other parameters by the impact load-strain curves of circular granite, which can generally be divided into three sections, i.e., the initial straight stage, nonlinear ascent yield stage and post-peak nonlinear decline stage. The factors in the test weaken the capacity of the circular granite to resist the impact, but the sizes of the inner diameters of the rings play a leading role. Dynamic tensile strain is generated in the inner wall along the impact direction during the impact, while compressive strain is produced on the inner wall in the vertical impact loading direction. learn more By analysing the crack propagation and final failure mode of circular granite, it is found that dynamic tensile failures are generated, crack initiation starts from the inner wall along the impact loading direction, and the outer circle in the vertical direction lags behind. The crack starts early and develops quickly on one side of the transmission bar. Finally, the failure criterion is established on the basis of some assumptions and circular-granite deformation failure characteristics, and the parameters, measured by the Brazilian disk test, are reasonably verified via substitution into the failure criterion equation.DNA-binding Transcription Factors (TFs) play a central role in regulation of gene expression in prokaryotic organisms, and similarities at the sequence level have been reported. These proteins are predicted with different abundances as a consequence of genome size, where small organisms contain a low proportion of TFs and large genomes contain a high proportion of TFs. In this work, we analyzed a collection of 668 experimentally validated TFs across 30 different species from diverse taxonomical classes, including Escherichia coli K-12, Bacillus subtilis 168, Corynebacterium glutamicum, and Streptomyces coelicolor, among others. This collection of TFs, together with 111 hidden Markov model profiles associated with DNA-binding TFs collected from diverse databases such as PFAM and DBD, was used to identify the repertoire of proteins putatively devoted to gene regulation in 1321 representative genomes of Archaea and Bacteria. The predicted regulatory proteins were posteriorly analyzed in terms of their genomic context, allowing the prediction of functions for TFs and their neighbor genes, such as genes involved in virulence, enzymatic functions, phosphorylation mechanisms, and antibiotic resistance.
