Tetraploid plants often have altered rates of vegetative growth relative to their diploid progenitors. However, the molecular basis for altered growth rates remains a mystery. This study reports microRNA (miRNA) and gene expression differences in Populus tetraploids and counterpart diploids using RNA and miRNA sequencing. The results showed that there was no significant difference between young leaves in the expression of vegetative growth-related miRNAs. However, as leaves aged, the expression of auxin- and gibberellin-related miRNAs was significantly upregulated, while the expression of senescence-related miRNAs was significantly downregulated. The dose effect enhanced the negative regulation of the target genes with ARFs, GA20ox, GA3ox, and GAMYB being downregulated, and TCP and NAC being upregulated. As a result, the chloroplast degradation of tetraploid leaves was accelerated, the photosynthetic rate was decreased, and the synthesis and decomposition ability of carbohydrate was decreased. The vulcanizate structure of filled compounds is affected by filler-rubber interactions (FRI) and the chemical crosslink density (CCD) of the matrix rubber. In particular, in filled compounds using a silica-silane system, FRIs due to silica-rubber coupling are a major influencing factor for the vulcanizate structure and physical properties. In this study, the effect of sulfur variation on the vulcanizate structure of silica-filled solution styrene-butadiene rubber compounds using a sulfide-silane coupling agent was studied. Forskolin order The vulcanizate structure according to sulfur variation was quantitatively analyzed using the swelling test and Flory-Rehner and Kraus equations. As the sulfur content increased, both FRI and the CCD increased, and it was confirmed that sulfur variation influenced the silica-rubber coupling efficiency through increased FRI. In addition, field emission scanning electron microscope images showed that increased FRI contributed to improvements in silica dispersion, abrasion resistance, and energy loss characteristics.Next generation sequencing (NGS) in combination with phage surface display (PSD) are powerful tools in the newly equipped molecular biology toolbox for the identification of specific target binding biomolecules. Application of PSD led to the discovery of manifold ligands in clinical and material research. However, limitations of traditional phage display hinder the identification process. Growth-based library biases and target-unrelated peptides often result in the dominance of parasitic sequences and the collapse of library diversity. This study describes the effective enrichment of specific peptide motifs potentially binding to arsenic as proof-of-concept using the combination of PSD and NGS. Arsenic is an environmental toxin, which is applied in various semiconductors as gallium arsenide and selective recovery of this element is crucial for recycling and remediation. The development of biomolecules as specific arsenic-binding sorbents is a new approach for its recovery. Usage of NGS for all biopanning fractions allowed for evaluation of motif enrichment, in-depth insight into the selection process and the discrimination of biopanning artefacts, e.g., the amplification-induced library-wide reduction in hydrophobic amino acid proportion. Application of bioinformatics tools led to the identification of an SxHS and a carboxy-terminal QxQ motif, which are potentially involved in the binding of arsenic. To the best of our knowledge, this is the first report of PSD combined with NGS of all relevant biopanning fractions.Continuous cultures assure the invariability of environmental conditions and the metabolic state of cultured microorganisms, whereas batch-cultured cells undergo constant changes in nutrients availability. For that reason, continuous culture is sometimes employed in the whole transcriptome, whole proteome, or whole metabolome studies. However, the typical method for establishing uniform growth of a cell population, i.e., by limited chemostat, results in the enrichment of the cell population gene pool with mutations adaptive for starvation conditions. These adaptive changes can skew the results of large-scale studies. It is commonly assumed that these adaptations reflect changes in the genome, and this assumption has been confirmed experimentally in rare cases. Here we show that in a population of budding yeast cells grown for over 200 generations in continuous culture in non-limiting minimal medium and therefore not subject to selection pressure, remodeling of transcriptome occurs, but not as a result of the accumulation of adaptive mutations. The observed changes indicate a shift in the metabolic balance towards catabolism, a decrease in ribosome biogenesis, a decrease in general stress alertness, reorganization of the cell wall, and transactions occurring at the cell periphery. These adaptive changes signify the acquisition of a new lifestyle in a stable nonstressful environment. The absence of underlying adaptive mutations suggests these changes may be regulated by another mechanism.Since the novel coronavirus outbreak of SARS-CoV-2 from the first cases whereof were reported in Wuhan, China, in December 2019, our globalized world has changed enormously. On the 11th of March 2020, the World Health Organization (WHO) declared COVID-19 a pandemic, and nations around the world have taken drastic measures to reduce transmission of the disease. The situation is similar in Switzerland, a small high-income country in Central Europe, where the first COVID-19 case was registered on the 25th of February 2020. Through literature review as well as correspondence with public health professionals and experts in mathematical modeling, this case study focuses on the outbreak's impact on Switzerland and on the measures this country has implemented thus far. Along with the rapid spread of the virus, the political organization, economy, healthcare system, and characteristics of the country greatly influence the approach taken in facing the crisis. Switzerland appears to be structurally well-prepared, but, according to mathematical modeling predictions, in order to avoid total collapse of healthcare facilities, the measures taken by the Swiss Government need to reduce the virus transmission chain by at least 70%. Fortunately, updated models on April 22nd show evidence that the non-pharmaceutical measures invoked have decreased transmission by an estimated 89%, proving effective management by the federal government and allowing for progressive deconfinement measures.
