Results demonstrated that local dynamics differences originate from local structural heterogeneity.We herein described a selenylative cyclization reaction of enynes by the utilization of diselenides as radical sources. The visible-light irradiation of the reaction mixture enables the generation of the selenium atom radical to trigger the radical addition/cyclization/selenation sequences. Anticancer activity and drug resistance of chemotherapy are closely related to the redox state of tumor cells. Herein, five lipophilic Pt(IV) prodrugs were synthesized on the basis of the most widely used anticancer drug cisplatin, whose anticancer efficacy and drug resistance are closely related to the intracellular redox state. Subsequently, a series of cisplatin-sensitive and drug-resistant cell lines as well as three patient-derived primary ovarian cancer cells have been selected to screen those prodrugs. To verify if the disruption of redox balance can be combined with these Pt(IV) prodrugs, we then synthesized a polymer with a diselenium bond in the main chain for encapsulating the most effective prodrug to form nanoparticles (NP(Se)s). NP(Se)s can efficiently break the redox balance via simultaneously depleting GSH and augmenting ROS, thereby achieving a synergistic effect with cisplatin. In addition, genome-wide analysis via RNA-seq was employed to provide a comprehensive understanding of the changes in transcriptome and the alterations in redox-related pathways in cells treated with NP(Se)s and cisplatin. Thereafter, patient-derived xenograft models of hepatic carcinoma (PDXHCC) and multidrug-resistant lung cancer (PDXMDR) were established to evaluate the therapeutic effect of NP(Se)s, and a significant antitumor effect was achieved on both models with NP(Se)s. Overall, this study provides a promising strategy to break the redox balance for maximizing the efficacy of platinum-based cancer therapy.The spatial distribution, concentration, particle size, and polymer compositions of microplastics in Lake Michigan and Lake Erie sediment were investigated. Fibers/lines were the most abundant of the five particle types characterized. Microplastic particles were observed in all samples with mean concentrations for particles greater than 0.355 mm of 65.2 p kg-1 in Lake Michigan samples (n = 20) and 431 p kg-1 in Lake Erie samples (n = 12). Additional analysis of particles with size 0.1250-0.3549 mm in Lake Erie resulted in a mean concentration of 631 p kg-1. The majority of polymers in Lake Michigan samples were poly(ethylene terephthalate) (PET), high-density polyethylene (HDPE), and semisynthetic cellulose (S.S. Cellulose), and in Lake Erie samples were S.S. Cellulose, polypropylene (PP), and poly(vinyl chloride) (PVC). Polymer density estimates indicated that 85 and 74% of observed microplastic particles have a density greater than 1.1 g cm-3 for Lake Michigan and Lake Erie, respectively. The current study provided a multidimensional dataset on the spatial distribution of microplastics in benthic sediment from Lake Michigan and Lake Erie and valuable information for assessment of the fate of microplastics in the Great Lakes.Extreme precipitation events affect water quantity and quality in various regions of the world. Heavy precipitation in 2019 resulted in a record high area of unplanted agricultural fields in the U.S. and especially in the Maumee River Watershed (MRW). March-July phosphorus (P) loads from the MRW drive harmful algal bloom (HAB) severity in Lake Erie; hence changes in management that influence P export can ultimately affect HAB severity. In this study, we found that the 2019 dissolved reactive P (DRP) load from March-July was 29% lower than predicted, while the particulate P (PP) load was similar to the predicted value. Furthermore, the reduced DRP load resulted in a less severe HAB than predicted based on discharge volume. The 29% reduction in DRP loss in the MRW occurred with a 62% reduction in applied P, emphasizing the strong influence of recently applied P and subsequent incidental P losses on watershed P loading. Other possible contributing factors to this reduced load include lower precipitation intensity, altered tillage practices, and effects of fallow soils, but more data is needed to assess their importance. We recommend conservation practices focusing on P application techniques and timing and improving resiliency against extreme precipitation events.Chemical reaction networks (CRNs) have been proposed as an abstraction for molecular computing. DNA strand displacement (DSD) reactions are good candidates to realize this endeavor, since DNA strands can be wired to implement the desired dynamic behavior in a test tube. Specialists use simulators to help them design such chemical systems before experimental implementation. In this sense, we present the DNAr package, an alternative open-source tool, developed in R language, for users from multidisciplinary areas. The current version of our tool offers functions to simulate CRNs, convert a formal CRN into a DSD network, interpret results, export to Visual DSD, and create libraries. Here, we use the consensus CRN to show DNAr features and a neural network model to demonstrate scalability, simulating more than 600 chemical reactions in a few minutes.Recently, advances in the synthesis and development of multifunctional nanoparticle platforms have opened up great opportunities and advantages for specifically targeted delivery of genes of interest. BSA-coated niosome structures (NISM@B) can potentially improve the efficiency in vitro delivery of nucleic acid molecules and the transfection of genes. Few studies have reported the combined use of niosomes with nucleic acid as therapeutic agents or decoy oligodeoxynucleotides (ODNs). Herein, we synthesized NISM@B to encapsulate NANOG decoy ODN (NISM@B-DEC), after which the physicochemical characteristics and in vitro and in vivo properties of NISM@B-DEC were investigated. Our results regarding physicochemical characteristics revealed that the stable niosome nanocarrier system was successfully synthesized with a regular spherical shape and narrow size distribution with proper zeta-potential values and had an appropriate biocompatibility. The ODN release from the niosome nanocarrier system exhibited controlled and pH-dependent behavior as the best models to explain the ODN release profile.
