The triggering receptor expressed on myeloid cells 2 (TREM2) is a multifunctional surface protein that affects survival, migration, and phagocytic capacity of myeloid cells. Soluble TREM2 levels were found to be increased in early stages of sporadic and familial Alzheimer's disease (AD) probably reflecting a defensive microglial response to some initial brain damage. The disintegrin and metalloproteases (ADAM) 10 and 17 were identified as TREM2 sheddases. We demonstrate that meprin β is a direct TREM2 cleaving enzyme using ADAM10/17 deficient HEK293 cells. LC-MS/MS analysis of recombinant TREM2 incubated with meprin β revealed predominant cleavage between Arg136 and Asp137, distant to the site identified for ADAM10/17. We further demonstrate that the metalloprotease meprin β cleaves TREM2 on macrophages concomitant with decreased levels of soluble TREM2 in the serum of Mep1b-/- mice compared to WT controls. Isolated BMDMs from Mep1b-/- mice showed significantly increased full-length TREM2 levels and enhanced phagocytosis efficiency compared to WT cells. The diminished constitutive shedding of TREM2 on meprin β deficient macrophages could be rescued by ADAM stimulation through LPS treatment. Our data provide evidence that meprin β is a TREM2 sheddase on macrophages and suggest that multiple proteases may be involved in the generation of soluble TREM2. © 2020 The Authors. OBJECTIVES The goal of this prospective study was to establish RIs for plasma proteins and thyroid hormones, total tetraiodothyronine (tT4), free T4 (fT4), total triiodothyronine (tT3), and free T3 (fT3), as well as iodine blood levels in red-eared sliders (Trachemys scripta elegans) and map turtles (Graptemys spp.). METHODS Variables were measured over 1 ½ years to determine variations depending on sex and season, as well as any correlations between the parameters. A total of 131 blood samples from red-eared sliders and 51 blood samples from map turtles were collected from May 2016 to November 2017. The levels of tT4, fT4, and fT3 were measured using standardized autoanalyzer test systems; tT3 was measured by ELISA and iodine inductively coupled plasma mass spectrometry. Total protein was quantitated using the biuret method, and plasma protein fractions were measured using capillary zone electrophoresis. RESULTS The results showed significant variations in thyroid hormone levels and plasma protein fractions depending on sex and season. Total T4 and fT4 concentrations were significantly (P  less then  .05) correlated in both turtle species and sexes in all seasons. Thyroid hormone levels correlated with blood proteins in a few seasons, including a positive correlation between tT4 and fT4 in female red-eared sliders in fall and female map turtles in summer and fall. CONCLUSIONS This study demonstrates the importance of considering species, season, and sex variations when interpreting thyroid hormone and plasma protein levels. Empirical work on semantic processing has shown that emotion also plays an important role especially in abstract concepts; however, models integrating emotion along with linguistic and visual information are lacking. Here, we first improve on visual and affective representations, derived from state-of-the-art existing models, by choosing models that best fit available human semantic data and extending the number of concepts they cover. The TETA-WICF/MCR was characterized using zoom stereo microscope, scanning electron microscopy (SEM), Fourier transform infrared (FTIR) spectroscopy, X-ray diffractometer (XRD), and magnetic separation performance analysis. Experimental results confirmed successful layer by layer modification of chitosan, biosorbent water-insoluble corn flour (WICF), TETA onto the surface of magnetic beads. The mean diameter of the TETA-WICF/MCR was 2.63 mm with good magnetic-responsive ability. Subsequently, the adsorption performance of the TETA-WICF/MCR obtained toward patulin was assessed in batch adsorption system and the results demonstrated that the adsorption process was strongly depended on adsorbent dosage, contact time, temperature, and initial patulin concentration. This preference is surprising because pigeons fail to maximize the rate of food intake; they exhibit a suboptimal preference. We advance a new explanation, the Δ-∑ hypothesis, in which the difference in probability of reinforcement within terminal links (Δ) and the overall reinforcement probability rate of each alternative (∑) are the key variables responsible for such suboptimal preference. We tested the Δ-∑ hypothesis in two experiments. In Experiment 1, we manipulated the Δs while maintaining constant all other parameters of the task, in particular the ∑s. We predicted a preference for the alternative with the larger Δ. In Experiment 2, we examined the effect of the overall reinforcement probabilities, the ∑s, while maintaining constant all other parameters of the task, in particular the Δs.
