Radiomics has potential for reflecting the differences in glioma perfusion heterogeneity between arterial spin labeling (ASL) and dynamic susceptibility contrast (DSC) imaging. The aim of this study was to compare radiomic features of ASL and DSC imaging-derived parameters (cerebral blood flow, CBF) and assess radiomics-based classification models for low-grade gliomas (LGGs) and high-grade gliomas (HGGs) using their parameters. The ASL-CBF and DSC-relative CBF of 46 glioma patients were normalized (ASL-nCBF and DSC-nrCBF) for data analysis. For each map, 91 radiomic features were extracted from the tumor volume. Seventy-five radiomic features were significantly different (P less then 0.00055) between ASL-nCBF and DSC-nrCBF. Positive correlations were observed in 75 radiomic features between ASL-nCBF and DSC-nrCBF. Even though ASL imaging underestimated CBF compared with DSC imaging, there were significant correlations (P less then 0.00055) in the first-order-based mean, median, 90th percentile, and maximum. Texture analysis showed that ASL-nCBF and DSC-nrCBF characterized similar perfusion patterns, while ASL-nCBF could evaluate perfusion heterogeneity better. Estimating the developmental stage of dinosaur embryos is hindered by the lack of a quantitative method for age determination, by the scarcity of material, and by the difficulty in visualizing that material. Here we present the results of a broad inquiry, using 3D reconstructions from X-ray computed tomography data, into cranial ossification sequences in extant saurian taxa and in well-preserved embryos of the early branching sauropodomorph dinosaur Massospondylus carinatus. Our findings support deep-time conservation of cranial ossification sequences in saurians including dinosaurs, allowing us to develop a new method for estimating the relative developmental percentage of embryos from that clade. We also observe null-generation teeth in the Massospondylus carinatus embryos which get resorbed or shed before hatching, similar to those of geckos. These lines of evidence allow us to confidently estimate that the Massospondylus carinatus embryos are only approximately 60% through their incubation period, much younger than previously hypothesized. The overall consistency of our results with those of living saurians indicates that they can be generalized to other extinct members of that lineage, and therefore our method provides an independent means of assessing the developmental stage of extinct, in-ovo saurians.5-Fluorouracil (5-FU) is a chemotherapeutic drug widely used to treat colorectal cancer. 5-FU is known to gradually lose its efficacy in treating colorectal cancer following the acquisition of resistance. We investigated the mechanism of 5-FU resistance using comprehensive lipidomic approaches. We performed lipidomic analysis on 5-FU-resistant (DLD-1/5-FU) and -sensitive (DLD-1) colorectal cancer cells using MALDI-MS and LC-MRM-MS. In particular, sphingomyelin (SM) species were significantly up-regulated in 5-FU-resistant cells in MALDI-TOF analysis. Further, we quantified sphingolipids including SM and Ceramide (Cer) using Multiple Reaction Monitoring (MRM), as they play a vital role in drug resistance. We found that 5-FU resistance in DLD-1/5-FU colorectal cancer cells was mainly associated with SM increase and Cer decrease, which are controlled by acid sphingomyelinase (SMPD1). In addition, reduction of SMPD1 expression was confirmed by LC-MRM-MS analysis and the effect of SMPD1 in drug resistance was assessed by treating DLD-1 cells with siRNA-SMPD1. Furthermore, clinical colorectal cancer data set analysis showed that down-regulation of SMPD1 was associated with resistance to chemotherapy regimens that include 5-FU. Thus, from our study, we propose that SM/Cer and SMPD1 are new potential target molecules for therapeutic strategies to overcome 5-FU resistance.Large-scale anthropogenic disturbances can have direct and indirect effects on marine communities, with direct effects often taking the form of widespread injury or mortality and indirect effects manifesting as changes in food web structure. Here, we report a time series that captures both direct and indirect effects of the Deepwater Horizon Oil Spill (DWH) on northern Gulf of Mexico (nGoM) reef fish communities. We observed significant changes in community structure immediately following the DWH, with a 38% decline in species richness and 26% decline in Shannon-Weiner diversity. Initial shifts were driven by widespread declines across a range of trophic guilds, with subsequent recovery unevenly distributed among guilds and taxa. For example, densities of small demersal invertivores, small demersal browsers, generalist carnivores, and piscivores remained persistently low with little indication of recovery seven years after the DWH. Initial declines among these guilds occurred prior to the arrival of the now-widespread, invasive lionfish (Pterois spp.), but their lack of recovery suggests lionfish predation may be affecting recovery. Factors affecting persistently low densities of generalist carnivores and piscivores are not well understood but warrant further study given the myriad ecosystem services provided by nGoM reef fishes.The metamorphism of snow (snowmelt process) has a potential influence on chemical and physical process occurring within it. This study carried out a detailed study on the variation of dissolved organic matter (DOM) in different stages of snowmelt in a typical mountain glacier located at Tibetan Plateau through collecting four different surface snow/ice categories, i.e., fresh snow, fine firn, coarse firn, and granular ice during May to October in 2015. The dissolved organic carbon (DOC) was observed by lost 44% from fresh snow to fine firn and enriched 129% from fine firn to granular ice, reflecting the dynamic variability in DOC concentration during snow metamorphism. The absorbance properties of each snow category are positively correlated with DOC concentration. The result of excitation emission matrix fluorescence with parallel factor analysis (EEM-PARAFAC) and Fourier transform ion cyclotron resonance mass spectrometry (FT-ICR MS) highlighted the domination of lipid- and protein-like compounds in glacial-derived DOM.
