Surprises are not always welcome. We have studied total body water, quantified the volume and determined the extracellular space, interstitial, and plasma volumes. Nonetheless, estimating these parameters clinically (at the bedside) is frustratingly imprecise. The author ruminates over 50 years clinical experience in intensive care units and grapples with the literature.Drought stress often limits plant growth and global crop yields. Catalase (CAT)-mediated hydrogen peroxide (H2 O2 ) scavenging plays an important role in the adaptation of plant stress responses, but the transcriptional regulation of the CAT gene in response to drought stress is not well understood. Here, we isolated an APETALA2/ETHYLENE RESPONSIVE FACTOR (AP2/ERF) domain-containing transcription factor (TF), NtERF172, which was strongly induced by drought, abscisic acid (ABA), and H2 O2 , from tobacco (Nicotiana tabacum) by yeast one-hybrid screening. NtERF172 localized to the nucleus and acted as a transcriptional activator. Chromatin immunoprecipitation, yeast one-hybrid assays, electrophoretic mobility shift assays, and transient expression analysis assays showed that NtERF172 directly bound to the promoter region of the NtCAT gene and positively regulated its expression. Transgenic plants overexpressing NtERF172 displayed enhanced tolerance to drought stress, whereas suppression of NtERF172 decreased drought tolerance. Under drought stress conditions, the NtERF172-overexpressed lines showed higher catalase activity and lower accumulation of H2 O2 compared with wild-type (WT) plants, while the NtERF172-silenced plants showed the inverse correlation. Exogenous application of amino-1,2,4-triazole (3-AT), an irreversible CAT inhibitor, to the NtERF172-overexpression lines showed decreased catalase activity and drought tolerance, and increased levels of cellular H2 O2 . Knock down NtCAT in the NtERF172-overexpression lines displayed a more drought stress-sensitive phenotype than NtERF172-overexpression lines. We propose that NtERF172 acts as a positive factor in drought stress tolerance, at least in part through the regulation of CAT-mediated H2 O2 homeostasis.Introduction The periocular and temporal regions are important aspects of beauty. The presence of reticular veins in these areas is undesirable and can also interfere with injection of neurotoxins to treat squint lines. 1064nm NdYAG laser shows promise as an effective treatment modality with long-lasting effects. Methods A retrospective study of 35 consecutive patients seen over a 3-year period in a private cosmetic clinic affiliated to the University of Toronto for periocular and temporal reticular veins was conducted. They were all treated with 1064 nm NdYAG laser. Results A total of 35 patients with an average age of 45.8 years old received treatment. 13 had lower eyelids treated, 9 had temples treated, 12 had both lower eyelids and temples treated, and 1 had both upper and lower eyelids treated. The first case of upper eyelid vein being treated with the 1064 nm NdYAG laser with great success after only 1 treatment is reported. 80% of the patients required only 1 to 2 treatment sessions. The average satisfaction score was 7.9 out of 10. The average pain score was 5.0 out of 10. 91% perceived the treatment outcome as excellent or good with the longest result lasting 33 months. There were 4 complications 1 scabbing, 1 small burn, 1 skin darkening and 1 teary eye. Conclusion The study showed that the use of the long-pulsed, contact-cooled, variable spot-sized 1064 nm NdYAG laser is effective and safe, with good patient satisfaction and tolerability, and is able to achieve long-term results. Objective This study examined the presence of posttraumatic stress disorder (PTSD) as a predictor of treatment response to dialectical behavior therapy (DBT) across the primary outcomes of interest within DBT (i.e., borderline personality disorder [BPD] symptoms, deliberate self-harm, emotion regulation [ER] difficulties) and PTSD symptoms. Method Participants (N = 56) were consecutive admissions to an outpatient DBT clinic that completed diagnostic interviews at intake and self-report outcome measures at intake and every 3 months throughout the treatment. Results Patients with (vs. without) a PTSD diagnosis did not report greater clinical severity at intake on most outcome measures, with the exception of PTSD symptom severity and, among older patients only, ER difficulties. The presence of a PTSD diagnosis was not associated with poorer treatment response to DBT. buy NSC 125973 Instead, PTSD was associated with better response on the measure of BPD symptom severity. Conclusions The results suggest that patients with PTSD can benefit from DBT.Risk analysis as a field and discipline is about concepts, principles, approaches, methods, and models for understanding, assessing, communicating, managing, and governing risk. The foundation of this field and discipline has been subject to continuous discussion since its origin some 40 years ago with the establishment of the Society for Risk Analysis and the Risk Analysis journal. This article provides a perspective on critical foundational challenges that this field and discipline faces today, for risk analysis to develop and have societal impact. Topics discussed include fundamental questions important for defining the risk field, discipline, and science; the multidisciplinary and interdisciplinary features of risk analysis; the interactions and dependencies with other sciences; terminology and fundamental principles; and current developments and trends, such as the use of artificial intelligence. The Brassica-specific gene MS5 mediates early meiotic progression, and its allelic variants contribute to a valuable genic male sterility three-line hybrid production system in rapeseed (Brassica napus L.). However, the underlying mechanisms of its triallelic inheritance are poorly understood. Herein, we show that the restorer allele MS5a and the maintainer allele MS5c , are both necessary for male fertility in Brassica napus. The functional divergence of the MS5a and MS5c is strongly related to the sequence variations in their coding region and less strongly to their promoter regions. The male-sterile allele MS5b encodes a chimeric protein containing only the complete MS5 coiled coil (CC) domain, having lost the MS5 superfamily domain. Both MS5a and MS5c can form homodimers in the nucleus via the CC domain. MS5b can competitively interact with MS5a or MS5c to form nonfunctional heterodimers. Owing to the close transcript levels of the MS5b and MS5c in the MS5b MS5c , these heterodimers induced a dominant-negative effect of MS5b on the MS5c , resulting in a male-sterile phenotype.
