Based upon functional predictions, OF + DEHP treatment altered carbohydrate metabolism, while both OF and OF + DEHP affected biosynthesis of fatty acids and lipid metabolism. Co-occurrence network analysis revealed decreases in cluster size and a fracturing of the microbial community network into unconnected components and a loss of keystone species in the OF + DEHP treatment when compared to Control and OF treatments. Data suggest that the addition of DEHP in the diet may exacerbate microbial dysbiosis, a consequence that may explain in part its role as an obesogenic chemical. This study aimed to investigate the effect of mono-(2-ethylhexyl) phthalate (MEHP), one of the major phthalate metabolites that are widespread in aquatic environments, on reproductive dysfunction, particularly on endocrine activity in adult male and female zebrafish. For 21 days, the zebrafish were exposed to test concentrations of MEHP (0, 2, 10, and 50 μg/mL) that were determined based on the effective concentrations (ECx) for zebrafish embryos. Exposure to 50 μg/mL MEHP in female zebrafish significantly decreased the number of ovulated eggs as well as the hepatic VTG mRNA abundance when those of the control group. Meanwhile, in female zebrafish, the biosynthetic concentrations of 17β-estradiol (E2) and the metabolic ratio of androgen to estrogen were remarkably increased in all MEHP exposed group compared with those in the control group, along with the elevated levels of cortisol. However, no significant difference was observed between these parameters in male zebrafishes. Therefore, exposure to MEHP causes reproductive dysfunction in female zebrafishes and this phenomenon can be attributed to the alteration in endocrine activities. Moreover, the reproductive dysfunction in MEHP-exposed female zebrafishes may be closely associated with stress responses, such as elevated cortisol levels. To further understand the effect of MEHP on the reproductive activities of fish, follow-up studies are required to determine the interactions between endocrine activities and stress responses. Overall, this study provides a response biomarker for assessing reproductive toxicity of endocrine disruptors that can serve as a methodological approach for an alternative to chronic toxicity testing.Scots pine (Pinus sylvestris L.) is a widespread tolerant forest tree-species; however, its adaptability to environmental change differs among sites with various buffering capacity. In this study, we compared the spatial effects of aridity index (AI) and nitrogen deposition (ND) on biomass density in natural and man-made pine stands of differing soil fertility using geographically weighted multiple lag regression. Soil fertility was defined using soil series as zonal trophic (27.9%), acidic (48.2%), gleyed (15.2%) and as azonal exposed (2.5%), maple (2.4%), ash (0.8%), wet (2.1%) and peat (0.9%) under pine stands in the Czech Republic (Central Europe; 4290.5 km2; 130-1298 m a.s.l.). Annual AI and ND in every pine stand were estimated by intersection between raster and vector from 1 × 1 km grid for years 2000, 2003, 2007 and 2010 of severe non-specific forest damage spread. Biomass density was obtained from a MODIS 250 × 250 m raster using the enhanced vegetation index (EVI) for years 2000-2015, with a decrease in EVI indicating non-specific damage. Environmental change was assessed by comparing predictor values at EVI time t and t+λ. Non-specific damage was registered over 51.9% of total forest area. Less than 8.8% of damaged stands were natural and the rest (91.2%) of damaged stands were man-made. Pure pine stands were more damaged than mixed. The ND effect prevailed up to 2007, while AI dominated later. Temporal increasing ND effect under AI effectiveness led to the most significant pine stand damage in 2008 and 2014. Predictors from 2000 to 2007 afflicted 58.5% of non-specifically damaged stands at R2 0.09-0.76 (median 0.38), but from 2000 to 2010 afflicted 57.1% of the stands at R2 0.16-0.75 (median 0.40). The most damaged stands occurred on acidic sites. Mixed forest and sustainable management on natural sites seem as effective remediation reducing damage by ND.Bees are considered as important providers of ecosystem services, acting via pollination process in crops and native plants, and contributing significantly to the maintenance of biodiversity. However, the decrease of bee's population has been observed worldwide and besides other factors, this collapse is also related to the extensive use of pesticides. In this sense, studies involving the assessment of adverse effects and the uptake of pesticides by bees are of great concern. This work presents an analytical method for the determination of the insecticide abamectin and the fungicide difenoconazole in the stingless bee Melipona scutellaris exposed via oral and topic to endpoints concentrations of active ingredients (a.i.) alone and in commercial formulations and the discussion about its mortality and uptake. For this purpose, QuEChERS (Quick, Easy, Cheap, Efficient, Rugged and Safe) acetate modified method was used for extraction and pesticides were determined by LC-MS/MS. The validation parameters have included a linear range between 0.01 and 1.00 μg mL-1; and LOD and LOQ of 0.038 and 0.076 μg g-1 for abamectin and difenoconazole, respectively. selleck compound The uptake of tested pesticides via oral and topic was verified by the accumulation in adult forager bees, mainly when the commercial product was tested. Mortality was observed to be higher in oral exposure than in topic tests for both pesticides. For abamectin in a commercial formulation (a.i.) no differences were observed for oral or topic exposure. On the other hand, for difenoconazole, topic exposure had demonstrated higher accumulation in bees, according to the increase of received dose. Through the results, uptake and the possible consequences of bioaccumulated pesticides are also discussed and can contribute to the knowledge about the risks involving the exposure of bees to these compounds.Airborne particulate matter (PM) has become a serious health issue causing pulmonary diseases such as asthma. Due to the side effects and non-specificity of conventional drugs, there is a need to develop natural-product-based alternative treatments. Sargassum horneri is a brown alga shown to have anti-oxidant, anti-inflammatory, and anti-allergic effects. Thus, we sought to determine whether ethanol extract of Sargassum horneri (SHE) mitigates the effect of PM exposure on asthma development. To establish a mouse model of asthma, BALB/c mice were sensitized with ovalbumin (OVA, 10 μg) and challenged with PM (5 mg/m3) for 7 days consecutively. SHE (200, 400 mg/kg), Prednisone (5 mg/kg), or PBS was daily administrated orally before PM exposure. SHE mitigated PM exacerbated dendritic cell activation. More importantly, SHE restrained Th2 polarization by attenuating transcription factors GATA3 and STAT5, which further mitigated the expression of Th2 cytokines interleukin (IL)-4, IL-5, and IL-13 in the lung homogenates of PM-exacerbated asthmatic mice.
