RESULTS In normal chow rats, constant light exposure led to glucose abnormalities and dyslipidemia. In high-fat-fed rats, constant light exposure exacerbated glucose abnormalities, dyslipidemia, insulin resistance, and inflammation and aggravated steatohepatitis. Compared with HFD-LD rats, HFD-LL had decreased plasma sphingosine-1-phosphate and elevated liver concentrations of total ceramide and specific ceramide species (ceramide d180/240, ceramide d181/220, ceramide d181/240, and ceramide d181/241), which were associated with increased hepatocyte apoptosis. CONCLUSIONS Constant light exposure causes dysregulation of sphingolipids and promotes steatohepatitis in high-fat-fed rats. © 2020 Journal of Gastroenterology and Hepatology Foundation and John Wiley & Sons Australia, Ltd.In tumor necrosis factor (TNF) signaling, phosphorylation and activation of receptor interacting protein kinase 1 (RIPK1) by upstream kinases is an essential checkpoint in the suppression of TNF-induced cell death. Thus, discovery of pharmacological agents targeting RIPK1 may provide new strategies for improving the therapeutic efficacy of TNF. In this study, we found that 3-O-acetylrubianol C (3AR-C), an arborinane triterpenoid isolated from Rubia philippinesis, promoted TNF-induced apoptotic and necroptotic cell death. To identify the molecular mechanism, we found that in mouse embryonic fibroblasts, 3AR-C drastically upregulated RIPK1 kinase activity by selectively inhibiting IKKβ. Notably, 3AR-C did not interfere with IKKα or affect the formation of the TNF receptor1 (TNFR1) complex-I. Moreover, in human cancer cells, 3AR-C was only sufficient to sensitize TNF-induced cell death when c-FLIPL expression was downregulated to facilitate the formation of TNFR1 complex-II and necrosome. Taken together, our study identified a novel arborinane triterpenoid 3AR-C as a potent activator of TNF-induced cell death via inhibition of IKKβ phosphorylation and promotion of the cytotoxic potential of RIPK1, thus providing a rationale for further development of 3AR-C as a selective IKKβ inhibitor to overcome TNF resistance in cancer therpay. © 2020 Federation of American Societies for Experimental Biology.Practical chiroptical sensing with a small group of commercially available aromatic aldehydes is demonstrated. Schiff base formation between the electron-deficient 2,4-dinitrobenzaldehyde probe and either primary amines, diamines, or amino alcohols proceeds smoothly in chloroform at room temperature and is completed in the presence of molecular sieves within 2.5 hours. The substrate binding coincides with a distinct circular dichroism signal induction at approximately 330 nm, which can be correlated to the absolute configuration and enantiomeric composition of the analyte. The usefulness of this sensing method is highlighted with the successful sensing of 18 aliphatic and aromatic amines and amino alcohols and five examples showing quantitative %ee determination with good accuracy. © 2020 Wiley Periodicals, Inc.BACKGROUND Vulval conditions have been shown to have a significant impact on patients' quality of life and can affect their relationships. The Lady Garden Club (LGC) is peer support group that was set up by patients with vulval conditions and is supported by the vulval clinic physicians at Chelsea & Westminster Hospital. OBJECTIVES Our aim was to assess the efficacy of this peer support group and the physician contribution to it. this website We also aimed to assess potential unmet needs of partners, which in turn could affect our patients' experience and quality of life. METHODS An anonymised online Survey Monkey link was sent to LGC members. Questions included a Dermatology Life Quality Index (DLQI) section. RESULTS The response rate was 60% (26). Over half (54%) were members > 2 years. Diagnoses included 85% (22) LS, lichen planus, 8% (2) eczema/psoriasis, warts 8% (2), vulval cancer 4% (1) and vulvodynia 8% (2). All valued a vulval specialist leading the LGC. Women benefitted from open member questioning (84%), learning from others (81%), self-help tips (81%), more patient information (77%), latest research updates (69%), sharing concerns and fears (65%), sharing personal experiences (62%), peer support network (62%), discussion about sex & relationships (35%). A third used the buddy system by phone, 19% soon after diagnosis. The average DLQI was 6.84 (range 0-25). Half reported depression and 59% anxiety. Two thirds (68%) felt women with other vulval conditions would benefit from the LGC. Over half (54%) felt there was an unmet need for helping partners understand their genital conditions. CONCLUSION The survey concluded that the LGC provides several additional benefits to women with vulval conditions, within a safe forum led by a vulval specialist. Currently with no available cure, COPD is associated with irreversible elastic tissue degradation in lungs, which results in release of unusual amino acids, isodesmosine and desmosine. These biomarkers are potential key elements in enzyme-linked immunosorbent assay (ELISA), an analytical method, which can detect certain compounds including antigens and proteins in easy and affordable manner. In order to target a biomarker with ELISA, it is necessary to prepare its specific antibody, which can be achieved by immunization of host organism with appropriate antigen containing the biomarker. Although preparation of these types of conjugates has been published, desmosine and isodesmosine used by researchers are obtained from natural sources such as animal tissues. Here, we report the first synthetic preparation of isodesmosine and keyhole limpet hemocyanin (KLH) conjugate from commercially available chiral amino acids and carrier protein. Formation of the core pyridinium of isodesmosine was achieved through key reaction-Chichibabin pyridinium synthesis-to deliver a 1,2,3,5-tetrasubstituted pyridinium amino acid selectively. Further modifications involving KLH and maleimide linker provided the target conjugate, which could potentially invoke an immune response to produce anti-isodesmosine antibody for the ELISA system. © 2020 Wiley Periodicals, Inc.
