Patients withdepressive and anxiety disorders showed consistent dysregulation across biological stress systems, particularly for current episodes. To understand stress system functionality in affective disorders, an integrated approach capturing cumulative stress indices within and across biological stress systems is important.Usually, Euro banknotes are made from cotton substrates and their waste is disposed of in landfill or is incinerated. In order to valorize the end-of-life euro banknotes (ELEBs), the substrates were used in this research for cellulase production via submerged fungal fermentation (SFF), and the resultant fungal cellulase w s used in ELEBs hydrolysis process for extraction of glucose. The experiments were started by exposing the ELEBs to different types of pretreatments, including milling process, alkali (NaOH/urea solution), and acid leaching to remove any contamination (e.g. dyes) and to decrease the crystallinity of cellulose (the main element in cotton substrate) thus increasing the degradation rate during the fermentation process. The effect of pretreatments on the morphology and chemical composition of ELEBs was observed using Scanning Electron Microscope and Energy Dispersive Spectrometry. Afterwards, Trichoderma reesei-DSM76 was used for cellulase production from the treated ELEBs with high cellulase activity (12.97 FPU/g). The resultant cellulase was upscaled in a bioreactor and used in ELEBs hydrolysis. Finally, the results showed that the optimized pretreatment methods (milling followed by leaching process) significantly improved the cellulase activity and glucose recovery, which was estimated by 96%. According to the obtained results, the developed strategy has a great potential for conversion of ELEBs into a glucose product that could be used in biofuels and bioplastics applications.Umifenovir is an antiviral drug approved in China and Russia for the treatment of influenza. The available dosage form consists of capsules marketed under the brand name Arbidol®. Due to its broad spectrum, umifenovir may also be used in other viral contexts, alone or combined with other antiviral drugs. Although knowledge of umifenovir intrinsic stability may be useful for any potential development of other pharmaceutical forms for other routes of administration and for quality risk management, no data regarding this matter is available to date. Spautin-1 price In this study, the exploration of the molecule's behaviour under hydrolytic, oxidative and photolytic conditions was carried out experimentally and supported by density functional theory (DFT) studies. It comes out that umifenovir is sensitive to these stress conditions giving rise to 6 structurally characterized degradation products. The one-electron oxidation process produced on the sulphur atom is probably the main cause of umifenovir degradation with reference to the structures of the degradation products formed and the DFT data.To administer vitamin C (VC) with precision to patients with the coronavirus disease (COVID-19), we developed an ultra-high-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method to assess plasma VC concentrations. 31 patients with COVID-19 and 51 healthy volunteers were enrolled. VC stability was evaluated in blood, plasma, and precipitant-containing stabilizers. A proportion of 7.7 % of VC was degraded in blood at room temperature (RT) (approximately 20-25 °C) at 1.5 h post administration with respect to the proportion degraded at 0.5 h, but without statistical difference. VC was stable in plasma for 0.75 h at RT, 2 h at 4 °C, 5 days at -40 °C, and 4 h in precipitant-containing stabilizer (2 % oxalic acid) at RT. The mean plasma concentration of VC in patients with COVID-19 was 2.00 mg/L (0.5-4.90) (n = 8), which was almost 5-fold lower than that in healthy volunteers (9.23 mg/L (3.09. 35.30)) (n = 51). After high-dose VC treatment, the mean VC concentration increased to 13.46 mg/L (3.93. 34.70) (n = 36), higher than that in healthy volunteers, and was within the normal range (6-20 mg/L). In summary, we developed a simple UPLC-MS/MS method to quantify VC in plasma, and determined the duration for which the sample remained stable. VC levels in patients with COVID-19 were considerably low, and supplementation at 100 mg/kg/day is considered highly essential. The present work describes the development and validation of a first report including several non-invasive NMR schemes to identify parameters as local chemical environments, homo- and heteronuclear site-specific spin correlations, diffusion coefficient-dependent polydispersity indexes and quantification of identified peptide entities that composes a commercial human Dialyzable Leucocyte Extract (DLE), Transferon, an oral liquid formulation of low-molecular-weight peptides. The above parameters were useful indicators to verify reproducibility, consistency and homogeneity among the DLE batches manufactured at Good Manufacturing Practice (GMP) facilities and for batch-releasing purposes in a quality control laboratory. The results showed that peptide identity of the DLE is represented with both high reproducible one-dimensional proton spectra and diffusion coefficient distributions that predicts in turn a weight-average molecular weight of around 6.7-7.4 kDa and a mean polydispersity index of 1.13. The obtained NMR peptide fingerprint of the analyzed DLE allowed to i) confirm its structural homogeneity by line-shape analysis, ii) identify and quantify its peptide content within the total solution with qNMR methods iii) to confirm the robustness of the technique as a feasible alternative for routine analysis of Natural or non-Natural Complex Drugs, such as DLEs.DNA barcoding is a widely used tool for species identification and authentication. However, it may not be applicable to highly processed herbal products due to severe DNA fragmentation. The emergence of DNA metabarcoding provides an alternative way to solve the problem. In this study, we are the first to combine the use of adaptor ligation-mediated PCR method and metabarcoding to reveal species identities in herbal products. As an illustration, we applied the method on three Hedyotis herbal products collected from China and Thailand. Results showed that H. diffusa and H. corymbosa were present in the products which were consistent with their label claims. Our study indicated that the adaptor ligation-mediated PCR with metabarcoding approach is useful for authentication of highly processed herbal products.
