Conclusions Our results suggest that PEW may be used as a bioactive ingredient to prevent obesity and its related metabolic disorders by modulating the composition of gut microbiota. © The Author(s) 2020.Background Androgen receptor (AR) has been reported to play vital roles in exercise-induced increase of muscle mass in rats, but needs to be further verified and the mechanism behind remains unclear. As AR target genes, insulin growth factor-1 (IGF-1) and IGF-1 receptor (IGF-1R) promote muscle hypertrophy through activating PI3K/Akt- mammalian target of rapamycin (mTOR) pathway, a classic pathway of muscle hypertrophy. So the main purpose of this study was using AR antagonist flutamide to demonstrate AR's effect on training-induced muscle hypertrophy and its possible mechanism IGF-1/IGF-1R- PI3K/Akt- mTOR pathway? Methods Forty-eight Sprague Dawley male rats aged 7 weeks were randomly divided into six groups control (C), flutamide (F), resistance training (R), resistance training plus flutamide (R + F), endurance training (E), and endurance training plus flutamide (E + F) groups. Flutamide was used to block AR in rats. Rats in R and R + F groups fulfilled 3 weeks of ladder climbing with progressively increase and endurance training-induced muscle hypertrophy, which was mediated at least partly through IGF-1/IGF-1R- PI3K/Akt- mTOR pathway. © The Author(s) 2020.Background Higher choline and betaine levels have been linked to lower risk of liver cancer, whereas existing data in relation to hepatocellular carcinoma (HCC) prognosis are scarce. Our objective was to examine the associations of the serum choline and betaine with HCC survival. Methods 866 newly diagnosed HCC patients were enrolled in the Guangdong Liver Cancer Cohort. Serum choline and betaine were assessed using high-performance liquid chromatography with online electro-spray ionization tandem mass spectrometry. Liver cancer-specific survival (LCSS) and overall survival (OS) were calculated. Cox proportional hazards models were used to estimate the hazard ratios (HRs) and 95% confidence intervals (CIs). Results Serum choline levels were associated with better LCSS (T3 vs. T1 HR = 0.69, 95% CI 0.51-0.94; P -trend less then 0.05) and OS (T3 vs. T1 HR = 0.73, 95% CI 0.54-0.99; P -trend less then 0.05). The associations were significantly modified by C-reactive protein (CRP) levels but not by other selected prognostic factors including sex, age, etc. The favorable associations between serum choline and LCSS and OS were only existed among patients with CRP ≥3.0 mg/L. No significant associations were found between serum betaine levels and either LCSS or OS. Conclusions This study revealed that higher serum choline levels were associated with better HCC survival, especially in HCC patients with systemic inflammation status. No significant associations were found between serum betaine and HCC survival. Our findings suggest the benefits of choline on HCC survival. In our study, we investigated the effect of the Tunisian jojoba seed on a metabolic syndrome induced in rat by the High Fat diet and High Fructose (HFHF) and its renal and hepatic complications. Methods The rats were fed with HFHF or Normal Diet (ND) for a period of 8 weeks. After that, a switch from HFHF to ND or Normal Diet Jojoba (NDJ),(jojoba diet approach) or High Fat and High Fructose and Jojoba diet (HFHFJ) (nutraceutical approach) has been done. Metabolic disorder was evaluated by measuring the fasting body weight, glycemia and C-peptide and leptin. Oxidative stress parameters like ThioBarbituric Acid Reactive Substances (TBARS) and Total Antioxidant Capacity (TAOC) were analyzed in the plasma and renal and hepatic function were determined by the measure of creatinine and alanine transferase (ALT) respectively. Histological analysis was performed on the liver, kidney and pancreas. Results HFHF diet exhibited characteristics of metabolic syndrome presented by insulin resistance, hyperinsulinemia, hyperleptinemia, fat mass with hepatic steatosis and renal disorder. HFHF diet was associated with oxidative stress (OS) presented by an increase in TBARS and a decrease in TAOC. Adding jojoba seeds to HFHF rat group diet induced a decrease in body weight, fat mass (58 and 41%), insulin resistance (59 and 56%), oxidative stress (60 and 41%), liver steatosis (from a score = 3 to a score = 0) and renal complications (25 and 42%). This effect was emphasized with diet approach. Conclusion The results demonstrated the beneficial effect of jojoba against metabolic syndrome and oxidative stress, suggesting that jojoba could be used in the prevention and treatment of metabolic syndrome. Graphical abstract © The Author(s) 2020.Background Severe hypoxia induces a series of stress responses in mammals; however, subterranean rodents have evolved several adaptation mechanisms of energy metabolisms and O2 utilization for hypoxia. Mammalian brains show extreme aerobic metabolism. Following hypoxia exposure, mammals usually experience irreversible brain damage and can even develop serious diseases, such as hypoxic ischemic encephalopathy and brain edema. To investigate mechanisms underlying the responses of subterranean rodents to severe hypoxia, we performed a cross-species brain transcriptomic analysis using RNA sequencing and identified differentially expressed genes (DEGs) between the subterranean rodent Lasiopodomys mandarinus and its closely related aboveground species L. VE-821 brandtii under severe hypoxia (5.0% O2, 6 h) and normoxia (20.9% O2, 6 h). Results We obtained 361 million clean reads, including 69,611 unigenes in L. mandarinus and 69,360 in L. brandtii. We identified 359 and 515 DEGs by comparing the hypoxic and normoxia groups DEGs were associated with environmental information processing and misregulation in cancers. Conclusions L. mandarinus has evolved hypoxia adaptation by enhancing DNA repair, damage prevention, and augmenting sensing, whereas L. brandtii showed a higher risk of tumorigenesis and promoted innate immunity toward severe hypoxia. These results reveal the hypoxic mechanisms of L. mandarinus to severe hypoxia, which may provide research clues for hypoxic diseases. © The Author(s) 2020.
