The three field tests were reproducible, but the same test, with and without the use of PGA, did not show agreement. © 2020 Wiley Periodicals, Inc.BACKGROUND Mannose-binding lectin (MBL) is a complement protein involved in the innate immune system, and is associated with some chronic respiratory diseases including noncystic fibrosis (non-CF) bronchiectasis in adults. The aim of this study was to investigate the frequency of MBL2 gene polymorphisms in children with non-CF bronchiectasis, and the effect of MBL deficiency on disease severity. METHODS Fifty children with non-CF bronchiectasis (bronchiectasis group) and 50 healthy controls (control group) were included. The demographic findings, number of acute pulmonary exacerbations in the previous year, airway cultures, pulmonary function tests, and radiologic scores of the bronchiectasis group were recorded. DNA extraction was performed in both groups and MBL2 gene polymorphisms in codons 52, 54, 57 in exon 1 and H/L, Y/X in the promoter region were studied using real-time polymerase chain reaction. Haplotypes were made by genotypes, and MBL serum expression was classified according to the genotypes in tsis, MBL genotype was different from healthy controls. Here, we derived MSCs from amniotic fluid and compared their differentiation ability and immunosuppression effect on PHA-activated PBMC with those of MSCs isolated from umbilical cords. Amniotic fluid MSCs were isolated and cultured on commercial AFC medium and classic MSC medium, and the number and size of colonies were used to evaluate differences in primary and passaged culture. Rate of proliferation, population doubling time, cell morphology, cell surface markers and mRNA expression were measured in subcultured cells. ABBV-2222 Furthermore, a comparative study was performed with umbilical cord MSCs to assess the ability of differentiation and immunosuppressive effect of PHA-stimulated PBMCs. Amniotic fluid MSCs were isolated and expanded by three methods, and exhibited nearly all the characteristics of umbilical cord MSCs. Early inhaled corticosteroids seem to reduce the incidence of bronchopulmonary dysplasia, but the long-term outcome remains unknown. RESEARCH QUESTION The aim of this study was to evaluate the effect of early inhaled corticosteroids on chronic respiratory morbidity. METHODS Fifty-nine survivors from the Prague cohort included in Neonatal European Study of Inhaled Steroids underwent further follow-up comprising of respiratory morbidity monitoring during the first 2 years of life followed by objective lung function testing performed at the age of 5.9 years (range 5-7 years). Both outcomes were pursued and finalized before the unblinding of budesonide subgroups. RESULTS Fifty randomized (budesonide vs placebo group, 56% vs 44%) survivors were included in the study. Spirometry was successfully performed in 48 children. No statistically significant differences were found in the lung function test (forced expiratory flow [FEF] - FEF75 , FEF50, FEF25 , and FEF25-75; FEV1 , forced vital capacity [FVC], FEV1 /FVC) although mild trend to the improvement of expiratory flow pattern was observed in the budesonide group (median z-score of FEV1 /FVC -0.376 vs -0.983, P = .13; median z-score of FEF25-75 -1.004 vs -1.458, P = .13; median z-score of FEF75 -0.527 vs -0.996, P = .17). Children assigned to budesonide had a significantly lower rate of symptoms of chronic lung disease (34.6% vs 68.2%; P = .04) than children assigned to placebo. INTERPRETATION Our study suggests that early inhaled budesonide was associated with the trend to the improvement of functional lung parameters and with a lower rate of symptoms of chronic lung disease within the first 2 years of life. © 2020 Wiley Periodicals, Inc.Highly twisted structures of expanded porphyrin provide a prominent basis to unravel the relationship between aromaticity and chirality. Here we report the synthesis of bis-Ge(IV) complexes of [38]octaphyrin that display rigid figure-eight structures. Two bis-Ge(IV) [38]octaphyrin isomers with respect to the stereochemistry of the axial hydroxy groups on the germanium ions were obtained and found to be aromatic. Upon oxidation with MnO2, these [38]octaphyrin complexes were converted to single syn-type isomer of [36]octaphyrin with maintenance of figure-eight conformation. The enantiomers have been successfully separated by HPLC equipped with chiral stationary phase. While aromatic [38]octaphyrin Ge(IV) complexes showed quite large molar circular dichroism up to Δε = 1500 M-1cm-1 with a dissymmetry factor gabs of 0.035, weakly antiaromatic [36]octaphyrin Ge(IV) complexes underscored moderate values; Δε = 540 M-1cm-1 with gabs of 0.023. Thus, figure-eight octaphyrin scaffold has been proved to be an attractive platform for novel chiroptical materials with tunable aromaticity.
