Across the Quercus species, the relative level of seed desiccation sensitivity, which we predicted to be important for reducing the risk of drying related mortality, was independent of climate. For desiccation-sensitive seeds this suggests a more diverse range of strategies for minimizing desiccation risk than reported previously.Cephalostachyum pingbianense is the only woody bamboo species that can produce bamboo shoots in four seasons under natural conditions. So far, the regulatory mechanism of shoot bud differentiation and development is unknown. In the present study, indole-3-acetic acid (IAA), zeatin riboside (ZR), gibberellin A3 (GA3 ) and abscisic acid (ABA) contents determination, RNA sequencing and differentially expressed gene analysis were performed on dormant rhizome bud (DR), growing rhizome bud (GR), and germinative bud (GB) in each season. The results showed that the contents of IAA and ZR increased while ABA content decreased, and GA3 content was stable during bud transition from dormancy to germination in each season. Moreover, rhizome bud germination was cooperatively regulated by multiple pathways such as carbohydrate metabolism, hormone signal transduction, cell wall biogenesis, temperature response, and water transport. The inferred hub genes among these candidates were identified by protein-protein interaction network analyses, most of which were involved in hormone and carbohydrate metabolism, such as HK and BGLU4 in spring, IDH and GH3 in winter, GPI and talA/talB in summer and autumn. It is speculated that dynamic phytohormone changes and differential expression of these genes promote the release of rhizome bud dormancy and contribute to the phenological characteristics of full-year shooting. Moreover, the rhizome buds of C. pingbianense may not suffer from ecodormancy in winter. These findings would help accumulate knowledge on shooting mechanisms in woody bamboos and provide a physiological insight into germplasm conservation and forest management of C. pingbianense.

Optical coherence tomography angiography (OCTA) is a premium imaging modality for noninvasive microvasculature studies. Deep learning networks have achieved promising results in the OCTA reconstruction task, benefiting from their powerful modeling capability. However, two limitations exist in the current deep learning-based OCTA reconstruction methods (a) the angiogram information extraction is only limited to the locally consecutive B-scans; and (b) all reconstruction models are confined to the 2D convolutional network architectures, lacking effective temporal modeling. As a result, the valuable neighborhood information and inherent temporal characteristics of OCTA are not fully utilized. read more In this paper, we designed a neighborhood information-fused Pseudo-3D U-Net (NI-P3D-U) for OCTA reconstruction.

The proposed NI-P3D-U was investigated on an in vivo animal dataset by a cross-validation strategy under both fully supervised learning and weakly supervised learning pipelines. To demonstrate the OCTA reconstin potential for clinical applications.Exosomes, which are membrane-bound extracellular vesicles (EVs), are generated in the endosomal compartment of almost all eukaryotic cells. They are formed upon the fusion of multivesicular bodies and the plasma membrane and carry proteins, nucleic acids, lipids and other cellular constituents from their parent cells. Multiple factors influence their production including cell stress and injury, humoral factors, circulating toxins, and oxidative stress. They play an important role in intercellular communication, through their ability to transfer their cargo (proteins, lipids, RNAs) from one cell to another. Exosomes have been implicated in the pathophysiology of various diseases including cardiovascular disease (CVD), cancer, kidney disease, and inflammatory conditions. In addition, circulating exosomes may act as biomarkers for diagnostic and prognostic strategies for several pathological processes. In particular exosome-containing miRNAs have been suggested as biomarkers for the diagnosis and prognosis of myocardial injury, stroke and endothelial dysfunction. They may also have therapeutic potential, acting as vectors to deliver therapies in a targeted manner, such as the delivery of protective miRNAs. Transfection techniques are in development to load exosomes with desired cargo, such as proteins or miRNAs, to achieve up-regulation in the host cell or tissue. These advances in the field have the potential to assist in the detection and monitoring progress of a disease in patients during its early clinical stages, as well as targeted drug delivery.Sensing carbohydrate availability is essential for plants to coordinate their growth and development. In Arabidopsis thaliana, TREHALOSE 6-PHOSPHATE SYNTHASE 1 (TPS1) and its product, trehalose 6-phosphate (T6P), are important for the metabolic control of development. tps1 mutants are embryo-lethal and unable to flower when embryogenesis is rescued. T6P regulates development in part through inhibition of SUCROSE NON-FERMENTING1 RELATED KINASE1 (SnRK1). Here, we explored the role of SnRK1 in T6P-mediated plant growth and development using a combination of a mutant suppressor screen and genetic, cellular and transcriptomic approaches. We report nonsynonymous amino acid substitutions in the catalytic KIN10 and regulatory SNF4 subunits of SnRK1 that can restore both embryogenesis and flowering of tps1 mutant plants. The identified SNF4 point mutations disrupt the interaction with the catalytic subunit KIN10. Contrary to the common view that the two A. thaliana SnRK1 catalytic subunits act redundantly, we found that loss-of-function mutations in KIN11 are unable to restore embryogenesis and flowering, highlighting the important role of KIN10 in T6P signalling. The function of borealin proteins has been widely reported in the cell division of animals. Nonetheless, there is little research about their only known paralogue (cell division cycle associated 9, cdca9). In this study, cdca9 was investigated in loach (Misgurnus anguillicaudatus) for the first time. cdca9 was highly expressed in the embryo before the gastrula stage, and it was predominantly expressed in the ovary, especially in the oocytes of stage II. In conclusion, this study reveals a potential function of cdca9 in the early embryogenesis and ovarian development of fish.

As a lipid-soluble vitamin necessary for normal human physiology, vitamin D is mostly used in fortified foods, medicines and adjuvant treatment of diseases. However, taken in high doses, vitamin D can be toxic.

We treated RAW264.7 cells with a semi-inhibitory concentration (IC

) of vitamin D

. The metabolic changes in the treated cells were analyzed by

H nuclear magnetic resonance (NMR) and ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS).

After treatment of RAW264.7 cells with an IC

dose of 55 μm vitamin D

, tunor necrosis factor-α levels decreased significantly and remarkable metabolic differences were also observed, with 12 types of metabolites were identified by

H NMR and 87 identified by UPLC-MS/MS. Moreover, the metabolism of amino acids, sugars, lipids and other metabolic pathways were also affected.

Although vitamin D

is an indispensable nutrient in the body, excessive exposure has negative effects on cells and their metabolism. The present study will assist further analyses of the mechanism underlying vitamin D

toxicity. © 2022 Society of Chemical Industry.

However, the function of PPRs that only localize to the cytoplasm remains unknown. Here, we demonstrated that the rice (Oryza sativa) PPR protein CYTOPLASM-LOCALIZED PPR1 (OsCPPR1) contributes to pollen development and localizes to the cytoplasm. Knocking down OsCPPR1 led to abnormal plastid development in tapetal cells, prolonged tapetal programmed cell death (PCD) and tapetum degradation, and significantly reduced pollen fertility. Transcriptome analysis revealed that the transcript level of OsGOLDEN-LIKE1 (OsGLK1), which encodes a transcription factor that regulates plastid development and maintenance, was significantly higher in the OsCPPR1 knockdown plants compared to wild-type plants. We further determined that OsCPPR1 downregulates OsGLK1 transcription by directly binding to the single-stranded regions of OsGLK1 mRNAs. Overexpression of OsGLK1 resulted in abnormal tapetum and plastid development, similar to that seen in OsCPPR1 knockdown plants, and suppression of OsGLK1 partially restored pollen fertility in the OsCPPR1 knockdown plants. We therefore conclude that OsCPPR1 suppresses OsGLK1 in the regulation of plastid development and PCD in the tapetum. Our work revealed novel functions for a cytosolic PPR, demonstrating the diverse roles of PPRs in plants and identifying a new regulatory mechanism for regulating pollen development in rice.

Acute pancreatitis exhibits a rapid clinical progression which makes it difficult to observe in human; hence, an experimental animal model is needed. This preliminary study performed an induction of acute pancreatitis using glycodeoxycholic acid (GDOC) in an experimental macaque model.

GDOC injections (initial dose of 11.20 mg/kg) were administered in an escalating manner at specific time points. The injection was given along the bilio-pancreatic duct, followed by measurement of vital signs, serum amylase-lipase, TNF-α, procalcitonin, oxidative stress parameters, and microscopic and macroscopic findings.

The results indicated that acute pancreatitis occurred following induction with low-dose GDOC. Serum amylase and lipase levels increased with subsequent GDOC injections. Blood pressure and heart rate were elevated, indicating abdominal pain. Changes in TNF-α, procalcitonin, and oxidative stress values showed active inflammation. We observed histologic features of pancreatitis and as the dose increased, vasodilation of the splanchnic vasculatures was observed.

Small dose GDOC injection in the bilio-pancreatic duct may have a role to induce acute pancreatitis in Macaca nemestrina.

Mycobacteria interact with macrophages altering their polarization state, which propagates establishment of infection. Thus, molecular macrophage properties in mycobacterial infections are critical both for understanding the biology of the infections as well as identifying therapeutic targets. Here, we review recent advances in the understanding how altered macrophage polarization in mycobacterial infections may lead to the design of targeted therapies that may reprogram these macrophages for enhanced mycobactericidal function.
