Celeriac F1 hybrid seed production is currently complicated due to the instability of cytoplasmic male sterile lines. To develop alternative alloplasmic CMS lines, an asymmetric protoplast fusion and hybrid screening methodology was established. Celeriac suspension cells protoplasts were used as the acceptor and carrot, coriander and white celery mesophyll protoplasts as the donor for protoplast fusion experiments. Acceptor cytoplasmic inheritance was inhibited by iodoacetamide treatment and donor nuclear genome inheritance was prevented by UV exposure. Protoplasts were selectively stained and fused using electroporation and polyethylene glycol, and candidate hybrid shoots were obtained. One chloroplast and three mitochondrial markers that could distinguish acceptor and donors organelles were used to characterize over 600 plants obtained after fusion events, without identifying any cybrid. In order to increase the testing efficiency a high number of micro plantlets were pooled and hence the presence of the carrot specific Atp1 marker in one of the pooled samples was detected. We demonstrated that fusion took place between celeriac and a carrot indicating that the creation of viable hybrids is possible although at a very low frequency. These findings open the path for new cytoplasmic hybridization and the isolation of novel CMS lines of celeriac.Surfing is one additional sport proposed by the Tokyo 2020 Organizing Committee. Surprisingly, substantial efforts to understand surfing energetics are recent, and the impact of a single surfing paddling cycle on fatigue and energy cost is still not clear. Since surfing paddling technique is highly specific, experiments in real practice conditions are necessary to provide deeper insights. Through a biophysical approach, biomechanical and energetics responses of surfing paddling were quantified and compared from 16 competitive male surfers (23.5 ± 10.0 years old, 65.3 ± 11.4 kg and 1.72 ± 0.01 m) during two sets (PRE and POST) of 10 s all-out tethered paddling plus 20 m sprint paddling, interposed by 6 min of endurance paddling. Faster surfers presented lower energy cost during sprint PRE (r2 = 0.30, p = 0.03) and endurance (r2 = 0.35, p = 0.02) relative surfing paddling velocities. FEN1-IN-4 concentration Although the energy cost was higher for a lower velocity at maximal paddling velocity POST, the energy cost of surfing paddling increased with absolute velocity according to a power function (R2 = 0.83). Our results suggest that fatigue seems to occur even following a single surfing paddling cycle. By measuring the refracted angle of the acoustic beam transmitted through this metafluid shaped as a prism, we determine the acoustic index to water according to Snell's law. These experimental data are then compared with an excellent agreement to calculations performed in the framework of Multiple Scattering Theory showing that the emergence of negative refraction depends on the volume fraction [Formula see text] of the resonant micro-beads. For diluted metafluid ([Formula see text]), only positive refraction occurs whereas negative refraction is demonstrated over a broad frequency band with concentrated metafluid ([Formula see text]).We aimed to explore sex differences in the association of childhood socioeconomic position (SEP) with the level of cognitive performance and the rate of cognitive decline. We studied 84,059 individuals (55% women; mean age 64 years) from the Survey on Health, Ageing and Retirement in Europe. Sex differences in the association of childhood SEP (household characteristics at age 10) with the level of cognitive performance (verbal fluency, immediate recall, delayed recall) were analysed using multilevel linear regression. Structural equation modelling tested education, depressive symptoms and physical state as mediators. The relationship between childhood socioeconomic advantage and disadvantage and the rate of cognitive decline was assessed using linear mixed-effects models. Higher childhood SEP was associated with a higher level of cognitive performance to a greater extent in women (B = 0.122; 95% CI 0.092-0.151) than in men (B = 0.109; 95% CI 0.084-0.135). The strongest mediator was education. Childhood socioeconomic disadvantage was related to a higher rate of decline in delayed recall in both sexes, with a greater association in women. Strategies to prevent impaired late-life cognitive functioning, such as reducing childhood socioeconomic disadvantages and improving education, might have a greater benefit for women.Upon insemination, sperm cells are exposed to components of the female reproductive tract (FRT) fluids, such as urea and epidermal growth factor (EGF). It has been shown that both urea and EGF use EGF receptor signaling and produce reactive oxygen species (ROS) that are required at certain levels for sperm capacitation and acrosome reaction. We therefore hypothesized that during bovine sperm capacitation, a high level of urea and EGF could interfere with sperm function through overproduction of ROS. High-level urea (40 mg/dl urea is equal to 18.8 mg/dl of blood urea nitrogen) significantly increased ROS production and TUNEL-positive sperm (sperm DNA fragmentation, sDF) percentage, but decreased HOS test score, progressive motility, acrosome reaction and capacitation. The EGF reversed the negative effects of urea on all sperm parameters, with the exception of ROS production and DNA fragmentation, which were higher in urea-EGF-incubated sperm than in control-sperm. The developmental competence of oocytes inseminated with urea-EGF-incubated sperm was significantly reduced compared to the control. A close association of ROS production or sDF with 0-pronuclear and sperm non-capacitation rates was found in the network analysis. In conclusion, EGF enhanced urea-reduced sperm motility; however, it failed to reduce urea-increased sperm ROS or sDF levels and to enhance subsequent oocyte competence. The data suggests that any study to improve sperm quality should be followed by a follow-up assessment of the fertilization outcome.
