The present study evaluated the effects of the tension-free vaginal tape (TVT) procedure in patients who had previously undergone pelvic floor repair (study group), compared to women who underwent TVT insertion only (control group). The study group comprised 84 patients who underwent the TVT procedure but had previously also undergone surgical POP repair. The control group consisted of 250 women in whom the TVT was inserted. The primary objective was to compare the objective cure rate and the secondary objective was to compare the subjective cure rate in both groups. Negative pad test was achieved in over 91% in both groups. Objective and subjective cure rates were compared, as well as complication rates. Significant improvement was observed in the postoperative 1-h pad test in all patients. In all patients, we observed significant improvement in the quality of life, with no differences between the groups. No differences were found in the occurrence of postoperative urinary retention, urgency and frequency of daytime micturition, or vaginal erosion between the groups. The current results demonstrate that the two-step approach to pelvic reconstruction and anti-incontinence surgery is as safe and effective as primary TVT implantation.Gully erosion is a form of natural disaster and one of the land loss mechanisms causing severe problems worldwide. This study aims to delineate the areas with the most severe gully erosion susceptibility (GES) using the machine learning techniques Random Forest (RF), Gradient Boosted Regression Tree (GBRT), Naïve Bayes Tree (NBT), and Tree Ensemble (TE). The gully inventory map (GIM) consists of 120 gullies. Of the 120 gullies, 84 gullies (70%) were used for training and 36 gullies (30%) were used to validate the models. Fourteen gully conditioning factors (GCFs) were used for GES modeling and the relationships between the GCFs and gully erosion was assessed using the weight-of-evidence (WofE) model. The GES maps were prepared using RF, GBRT, NBT, and TE and were validated using area under the receiver operating characteristic(AUROC) curve, the seed cell area index (SCAI) and five statistical measures including precision (PPV), false discovery rate (FDR), accuracy, mean absolute error (MAE), and root mean squared error (RMSE). Nearly 7% of the basin has high to very high susceptibility for gully erosion. Validation results proved the excellent ability of these models to predict the GES. Of the analyzed models, the RF (AUROC = 0.96, PPV = 1.00, FDR = 0.00, accuracy = 0.87, MAE = 0.11, RMSE = 0.19 for validation dataset) is accurate enough for modeling and better suited for GES modeling than the other models. Therefore, the RF model can be used to model the GES areas not only in this river basin but also in other areas with the same geo-environmental conditions.Applying electrical stimulation (ES) could affect different cellular mechanisms, thereby producing a bactericidal effect and an increase in human cell viability. Despite its relevance, this bioelectric effect has been barely reported in percolated conductive biopolymers. In this context, electroactive polycaprolactone (PCL) scaffolds with conductive Thermally Reduced Graphene Oxide (TrGO) nanoparticles were obtained by a 3D printing method. Under direct current (DC) along the percolated scaffolds, a strong antibacterial effect was observed, which completely eradicated S. aureus on the surface of scaffolds. Notably, the same ES regime also produced a four-fold increase in the viability of human mesenchymal stem cells attached to the 3D conductive PCL/TrGO scaffold compared with the pure PCL scaffold. These results have widened the design of novel electroactive composite polymers that could both eliminate the bacteria adhered to the scaffold and increase human cell viability, which have great potential in tissue engineering applications.BACKGROUND The role of vitamin D and omega-3 long chain polyunsaturated fatty acids (omega-3 LCPUFA) in improving core symptoms of autism spectrum disorder (ASD) in children has been investigated by a few randomised controlled trials and the results are mixed and inconclusive. The response to treatment with these nutrients is heterogenous and may be influenced by inflammatory state. As an exploratory analysis, we investigated whether inflammatory state would modulate the effect of these nutrients on core symptoms of ASD. Methods Seventy-three New Zealand children with ASD (2.5-8.0 years) completed a 12-month randomised, double-blind, placebo-controlled trial of vitamin D (VID, 2000 IU/day), omega-3 LCPUFA; (OM, 722 mg/day docosahexaenoic acid), or both (VIDOM). Non-fasting baseline plasma interleukin-1β (IL-1β) was available for 67 children (VID = 15, OM = 21, VIDOM = 15, placebo = 16). Children were categorised as having undetectable/normal IL-1β ( 0.10); OM and VIDOM (P = 0.01) for SRS-awareness. When only children with elevated IL-1β were included, five outcomes showed greater improvements OM (P = 0.01) for SRS-total; OM (P = 0.03) for SRS-social communicative functioning; VID (P = 0.01), OM (P = 0.003) and VIDOM (P = 0.01) for SRS-awareness. CONCLUSION Inflammatory state may have modulated responses to vitamin D and omega-3 LCPUFA intervention in children with ASD, suggesting children with elevated inflammation may benefit more from daily vitamin D and omega-3 LCPUFA supplementation.
