In comparison to hERGWT channels, hERGW410R channels have markedly decreased total and surface expressions. W410R mutation dramatically reduces hERG channel currents (IKr) and shifts its steady-state activation curve to depolarization. Moreover, hERGW410R channels make dominant-negative effects on hERGWT channels. Significantly, we find hERG channel blocker E-4031 could partially rescue the function of hERGW410R channels by increasing the membrane expression. By using in silico model, we reveal that hERGW410R channels obviously elongate the repolarization of human ventricular myocyte action potentials. Collectively, W410R mutation decreases the currents of hERG channel, because of diminished membrane expression of mutant channels, that subsequently leads to elongated repolarization of cardiomyocyte, which might induce the pathogenesis of LQT2. Furthermore, E-4031 could partially rescue the decreased activity of hERGW410R channels. Thus, our work identifies a novel loss-of-function mutation in KCNH2 gene, which might provide a rational basis for the management of LQT2.In areca nut and husk, 14 elements (As, Ca, Cd, Cl, Co, Cu, K, Mg, Mn, Na, Rb, Sb, and Zn) were determined, while 34 elements including rare earth elements were detected in the corresponding soil samples using instrumental neutron activation analysis and atomic absorption spectrometry methods, whereas the concentration levels of Hg in tested samples are negligible, perhaps, below the detection limits. Bet-hedging is a complex evolutionary strategy involving morphological, eco-physiological, (epi)genetic and population dynamics aspects. We review these aspects in flowering plants and propose further research needed for this topic. It has evolved in organisms subjected to variable cues from the external environment, be they abiotic or biotic stresses such as irregular rainfall or predation. In flowering plants, bet-hedging is exhibited by hundreds of species and is mainly exerted by reproductive organs, in particular seeds but also embryos and fruits. The main example of bet-hedging in angiosperms is diaspore heteromorphism in which the same individual produces different seed/fruit morphs in terms of morphology, dormancy, eco-physiology and/or tolerance to biotic and abiotic stresses in order to 'hedge its bets' in unpredictable environments. The objective of this reviet-hedging strategies in plants 1) understanding ecological trade-offs among different traits; 2) producing more comprehensive phylogenetic analyses to understand the diffusion and evolutionary implications of this strategy; 3) clarifying epigenetic mechanisms related to bet-hedging and plant responses to environmental cues; and 4) applying multi-omics approaches to study bet-hedging at different levels of detail. Water limitation affects the quantity and quality of plants available to herbivores as food, and can also affect higher trophic-level consumers through variability in prey quality and reduced availability of suitable habitats. Our study assessed the response of an arthropod community to water limited wheat (Triticum aestivum L.) in a field setting. We used rainout shelters to exclude precipitation, irrigated raised bed plots to create three levels of water availability, and monitored arthropod community development over 8 weeks. First, we compared arthropod communities in habitats with different levels of water limitation and found that community composition was reliant on the magnitude of the water stress. This difference was largely due to the loss of piercing-sucking herbivores and predators in high-stress environments. Next, we focused on aphids and their natural enemies to investigate the underlying mechanisms driving community responses using structural equation modeling (SEM). Aphid abundance was negatively affected by water limitation, and this response was primarily associated with stress-induced plant physiological changes and not plant biomass or natural enemy abundance. Natural enemy abundance was also reduced in water-limited habitats, but natural enemies responded to plant biomass and not prey availability. These effects were exacerbated as water stress increased. The absence of natural enemy effects on aphids indicates that top-down predation effects were dampened by strong bottom-up effects of plant water limitation. This study revealed the importance of considering water stress intensity when predicting outcomes of droughts for arthropod communities.
