mCSM-membrane will be an invaluable and dedicated resource for investigating the effects of single-point mutations on membrane proteins through a freely available, user friendly web server at http//biosig.unimelb.edu.au/mcsm_membrane.FATCAT 2.0 server (http//fatcat.godziklab.org/), provides access to a flexible protein structure alignment algorithm developed in our group. In such an alignment, rotations and translations between elements in the structure are allowed to minimize the overall root mean square deviation (RMSD) between the compared structures. This allows to effectively compare protein structures even if they underwent structural rearrangements in different functional forms, different crystallization conditions or as a result of mutations. The major update for the server introduces a new graphical interface, much faster database searches and several new options for visualization of the structural differences between proteins.Digital PCR provides high sensitivity and unprecedented accuracy in DNA quantification, but current approaches require dedicated instrumentation and have limited opportunities for multiplexing. Here, we present an isothermal platform for digital enumeration of DNA reaction products in multiplex via standard fluorescence microscopy. Circular DNA strands, which may result from a wide range of molecular detection reactions, are captured on streptavidin-coated surfaces via hybridized biotinylated primers, followed by rolling circle amplification (RCA). The addition of 15% polyethylene glycol 4000 during RCA resulted in uniform, easily recorded reaction products. Immobilized DNA circles were visualized as RCA products with 100% efficiency, as determined by droplet digital PCR. Therefore, to balance clinical usage/availability and wastage in a tertiary hospital setting without a trauma center, an innovative system model was established. This system reduced wastage by transferring platelet unit approaching their expiration date to a nearby facility (with the same blood supplier and a trauma center) before expiration, when there is no anticipated need for the product at the original location. Methods A review of data to determine the degree of platelet wastage and wastage costs one year before implementation of this measure in October 2017 and one year after implementation of this measure. Results Since the implementation of this measure, no platelet units have expired on the shelf. In contrast, from October 2016 to October 2017, before implementation of platelet transfer, platelet products expired regularly. Conclusion This new system model is highly effective in maintaining platelet inventory without wastage.Early onset diabetes has adverse transgenerational effects, yet in-person National Diabetes Prevention Programs (NDPPs) have low reach among adults of peak reproductive age. We examined participation and weight loss with online NDPPs for younger versus older adults. Solera Health, Inc., collected data from 12,966 adults who enrolled in a yearlong online NDPP from 2015 to 2018. We used general linear models and logistic regression to assess differences between younger and older adults ( less then 45 vs. ≥45 years) in session initiation (logging in), session completion (activities approximating intensity of in-person classes), and weight loss, overall and according to engagement thresholds. Almost all (N = 12,497, 96%) individuals who enrolled initiated ≥1 session(s), but fewer (N = 2,408, 19%) completed ≥4 sessions over ≥9 months, achieving 4.5% weight loss on average. Among all enrollees with ≥2 weights (N = 10,161), younger men and women lost less weight (1.8% and 1.7%, respectively) than older men (3.3%) and women (2.7%; all p less then .05). Among all enrollees who completed ≥4 sessions over ≥9 months, weight loss did not differ between older men (4.3%), older women (4.0%), and younger men (3.5%), but younger women achieved less weight loss (3.0%) than older adults (all p less then .001). Online programming supports NDPP reach and weight loss, although younger adults completed fewer sessions and young women achieved less weight loss than older adults. Efforts to increase ongoing engagement among younger adults are needed to prevent early onset of diabetes and adverse transgenerational effects.Background Patients with inflammatory bowel disease (IBD) may be at increased risk of adverse neonatal outcomes. The aim of this study was to determine pooled incidences and risk factors for these outcomes. Methods Medline, Embase, and Cochrane Library were searched through May 2019 for studies reporting adverse neonatal outcomes in IBD. Odds ratios (ORs) with 95% confidence intervals (CIs) were calculated. Results The pooled incidence of preterm birth, low birth weight, congenital anomalies, and infants transferred to the neonatal intensive care unit was 8.6% (95% CI, 7.0%-10.1%), 8.9% (95% CI, 7.3%-10.5%), 2.1% (95% CI, 1.6%-2.6%), and 4.9% (95% CI, 2.9%-6.9), respectively. Compared with healthy controls, patients with IBD were more likely to deliver infants with low birth weight ( less then 2500 grams; OR, 2.78; 95% CI, 1.16-6.66) and infants admitted to the intensive care unit (OR, 3.33; 95% CI, 1.83-6.05). Patients with Crohn's disease had an increased incidence of congenital anomalies (OR, 3.03; 95% CI, 1.43-6.42). Among IBD patients, active disease was associated with increased incidence of preterm birth (OR, 2.06; 95% CI, 1.21-3.51), low birth weight (OR, 2.96; 95% CI, 1.54-5.70), and small for gestational age (OR, 2.62; 95% CI, 1.18-5.83). Antitumor necrosis factor (anti-TNF) use during pregnancy was associated with an increased incidence of neonatal intensive care unit admission (OR, 2.42; 95% CI, 1.31-4.45) and low birth weight (OR, 1.54; 95% CI, 1.01-2.35). Conclusions Patients with IBD, particularly with active disease or requiring anti-TNF therapy, may be at increased risk of developing adverse neonatal outcomes.
