y of such cases in groups of patients where standard approaches to prosthetics were applied. In particular, aesthetic defects found in the main group of patients were 2.5 times less frequent than in the comparison group, and repeated prosthetics was performed 2.7 times less frequently.

 Application of the developed comprehensive approach to planning of prosthodontic rehabilitation provides for significant reduction in the frequency of complications and aesthetic defects, as well as cases of repeated need for prosthetics.

Conduct a histological comparison of the pulp response to different materials, with a focus on the continuity and morphology of the mineralized barrier after direct pulp capping.

 One hundred and eight maxillary first molars of 54 Wistar rats were subject to direct pulp capping and divided into three groups according to the materials used calcium hydroxide (CH), mineral trioxide aggregate (MTA), and Biodentine. All cavities were sealed, and the animals were euthanized at 7, 14, and 21 days. Descriptive histological evaluation of the inflammation and formation of the mineralized barrier was performed.

 Statistical analyses were performed using the Kruskal-Wallis test, which was complemented by the Dunn test; differences with

< 0.05 were considered statistically significant.

 The results showed that MTA and Biodentine elicited less intense inflammatory reactions than CH. With respect to the formation and quality of the dentin barrier formed, differences were observed at 21 days between the analyzed groups; the best results being obtained following treatment with MTA and Biodentine.

 MTA and Biodentine induced formation of a more continuous and uniform mineralized barrier with less intense pulp response than CH.

 This article aimed to study the effect of different concentrations of nano zinc oxide particles on the color change of MDX4-4210 facial silicone elastomer after artificial aging.

 Silicone specimens (

= 150) were fabricated by incorporating intrinsic pigments and divided into three groups-white, yellow, and red, each group consisting of 50 specimens (

= 50). In each color, specimens were subdivided into five subgroups according to the quantity of zinc oxide nanoparticles (0, 0.5, 1.0, 1.5, and 2.0% weight), where the 0% weight served as the control in each group. #link# All specimens were then subjected to artificial aging using an accelerated aging machine chamber for 12, 24, 48, and 72 hours. L*a*b* values of specimens were noted after a different aging period by a spectrophotometer and ∆

* was calculated.

 Two-way repeated analysis of variance (ANOVA) was done to examine the effects under test conditions (concentration and aging time) of each color group. Then color, concentration, and the aging period were subjected to three-way repeated ANOVA to investigate the effects of different colors and concentrations on ∆

*. Bonferroni's test was performed to identify differences between groups. The significant level was at

= 0.05.

 The control group showed significantly higher ∆

* values than the test groups. The 1.5% test group showed significantly lower ∆

* compared with the others. The 0.5 to 2.0% of nano zinc oxide significantly decreased the color change of the silicone elastomer (

< 0.05), but there were no significant differences among groups.

 Incorporation of 1.5% of nano zinc oxide can improve the color stability of silicone prosthesis (MDX4-4210).

 The current research was conducted to evaluate the frequency of color-vision deficiency among dental students of King Khalid University College of Dentistry, Saudi Arabia.

 A cross-sectional study was performed among 203 dental students working as interns at male and female dental clinics of King Khalid University College of Dentistry (KKUCOD), Saudi Arabia. The Ishihara color-vision deficiency (CVD) test with 24 plates was used for diagnosis of the problem. Analysis of the data was done by performing Chi-square tests using SPSS software version 20.

 The frequency of total CVD was found to be 3.9%. While the association of gender with total CVD was observed to be statistically nonsignificant, a statistically significant relation was drawn with red-green color deficiency. Out of the total of 203 patients, 44 males were identified with red-green color deficiency, whereas only three females were found to have this condition, therefore revealing that CVD is more prevalent in males. Age was found to have a significant association with red-green color vision deficiency, protanopia, and total CVD.

 A total of 20.19% male dental students had red-green color vision deficiency compared to 1.4% in female students. The dental students must be aware of their congenital color vision deficiency and its impact on their professional life. Screening of such dental students and professionals is quite important so that they can tackle the color vision problems well without having detrimental effect on their future dental practice.

 Conservative approaches to esthetically treat stained arrested caries lesions (s-ACLs) have not been explored in clinical studies. This study aims to investigate the efficacy of in-office dental bleaching agent, as a conservative approach, to esthetically treat s-ACLs.

 Twelve patients (

= 46) presented with s-ACLs were treated with 40% hydrogen peroxide (in-office bleaching protocol; 20 minutes × 3). Color values were measured using a spectrophotometer (CIE L*a*b*), aided with digital photography to assess visual color change clinically. Measurements were taken for each specimen at baseline and immediately after bleaching.

 The color change calculated before and after bleaching for each dental substrate was analyzed using paired

-test (α = 0.05).

 The bleached s-ACLs had a significant increase in

* values (

< 0.001), and a significant decrease in both

* (

= 0.001) and

* (

= 0.007) values, indicating lighter color improvement (bleaching efficacy). The baseline mean of

*,

*, and

* values were 61.5, 2, and 15.4, respectively, and after bleaching were 67.7, 1.4, and 13.3, respectively, with a mean increase in ∆

of >7.9, which resulted in a visible clinical stain improvement as orange/light brown stains were removed completely, while gray/black stains improved to a lesser extent.

 Significant color improvement was observed when the in-office bleaching protocol (40% hydrogen peroxide) was used in orange/brown s-ACLs. However, it showed lesser improvement in gray/black s-ACLs.

 Microsurgery requires repeated practice and training to achieve proficiency, and there are a variety of curriculums available. This study aims to determine the importance of an expert instructor to guide students through procedures. We compared student proficiency across two microsurgery courses one with (Columbia University, United States [CU] cohort) and one without a dedicated microsurgery instructor (University of Thessaloniki, Greece [UT] cohort).

 Students were divided into two cohorts of 22 students (UT cohort) and 25 students (CU cohort). Student progress was evaluated by examining patency (lift-up and milking tests), anastomotic timing, and quality (Anastomosis Lapse Index [ALI]) of end-to-end arterial and venous anastomoses on day 1 and again on day 5. Chi-squared tests evaluated patency immediately and 30 minutes postoperation.

-Tests evaluated anastomotic timing and ALI scores. link2

-Values < 0.05 were considered significant.

 We evaluated progress within and between each cohort. Within t of progress and proficiency level attained at the conclusion of the course. We suggest students who are seeking to maximize proficiency in microsurgical procedures enroll in courses with an expert instructor.Leukemogenesis is characterized by chromosomal rearrangements with additional molecular disruptions, yet the cooperative mechanisms are still unclear. Using whole-exome sequencing of a pair of monozygotic twins who were discordant for childhood acute lymphoblastic leukemia (ALL) with ETV6-RUNX1 (E/R) gene fusion successively after birth, we identified the R209C mutation of G protein subunit α o1 (GNAO1) as a new ALL risk loci. Moreover, GNAO1 missense mutations are recurrent in ALL patients and are associated with E/R fusion. Ectopic expression of the GNAO1 R209C mutant increased its GTPase activity and promoted cell proliferation and cell neoplastic transformation. Combined with the E/R fusion, the GNAO1 R209C mutation promoted leukemogenesis through activating PI3K/Akt/mTOR signaling. Reciprocally, activated mTORC1 phosphorylated p300 acetyltransferase, which acetylated E/R and thereby enhanced the E/R transcriptional activity of GNAO1 R209C. Thus, our study provides clinical evidence of the functional cooperation of GNAO1 mutations and E/R fusion, suggesting GNAO1 as a therapeutic target in human leukemia.Huntington disease (HD) is a neurodegenerative disorder that is caused by a CAG repeat expansion in HTT. The length of this repeat, however, only explains a proportion of the variability in age of onset in patients. Genome-wide association studies have identified modifiers that contribute toward a proportion of the observed variance. By incorporating tissue-specific transcriptomic information with these results, additional modifiers can be identified. link3 We performed a transcriptome-wide association study assessing heritable differences in genetically determined expression in diverse tissues, with genome-wide data from over 4000 patients. Functional validation of prioritized genes was undertaken in isogenic HD stem cells and patient brains. Enrichment analyses were performed with biologically relevant gene sets to identify the core pathways. HD-associated gene coexpression modules were assessed for associations with neurological phenotypes in an independent cohort and to guide drug repurposing analyses. Transcriptomic analyses identified genes that were associated with age of HD onset and displayed colocalization with gene expression signals in brain tissue (FAN1, GPR161, PMS2, SUMF2), with supporting evidence from functional experiments. This included genes involved in DNA repair, as well as novel-candidate modifier genes that have been associated with other neurological conditions. Further, cortical coexpression modules were also associated with cognitive decline and HD-related traits in a longitudinal cohort. In summary, the combination of population-scale gene expression information with HD patient genomic data identified novel modifier genes for the disorder. Further, these analyses expanded the pathways potentially involved in modifying HD onset and prioritized candidate therapeutics for future study.
