Methods All patients referred to our hospital, from July 2018 to July 2019, who were arthroscopic candidates due to sport-related pathologies, underwent ultrasound scan 1 day prior to surgery. Ultrasound assessment was performed by an expert radiologist, with a 13-MHz probe, located transversely proximal to the patella in different degrees of knee flexion to assess trochlear lesion grade and thickness. Arthroscopic examination of all patients was performed by an experienced orthopedic knee surgeon (second author). Sensitivity and specificity of ultrasound were calculated. Results A total of 48 patients were involved in the study with a mean age of 33.2 years (SD 9.7), between 19 and 51 years of age. Patients were 81% male (39 patients). The sensitivity of ultrasound in grading of trochlear cartilage lesion was 100%, meanwhile its specificity was 88.2% (30 cases had normal cartilage while this figure was 34 in arthroscopy). It can play an important role as an outpatient diagnostic workup in patients with anterior knee pain.Organosolv pretreatment with two ethanol concentrations (25% and 50%, v/v) was performed to improve enzymatic saccharification of poplar sawdust. It was found that lower ethanol concentration (25%, v/v) pretreatment resulted in a higher enzymatic digestibility of poplar (38.1%) due to its higher xylan removal and similar lignin removal ratios, compared to that pretreated with 50% (v/v) ethanol pretreatment (27.5%). However, the residual lignin still exhibited a strong inhibition on enzymatic hydrolysis of organosolv-pretreated poplar (OP). Bio-surfactant preparations including tea saponin (TS), TS crude extract, and tea seed waste were applied in enzymatic hydrolysis of OP, due to their potential ability of reducing enzyme non-productive binding on lignin. Their optimal loadings in enzymatic hydrolysis of OP were optimized, which indicated that adding 0.075 g/g glucan of TS improved the 72-h glucose yield of OP by 48.3%. Moreover, adding TS crude extract and tea seed waste exhibited the better performance than TS for improving enzymatic hydrolysis of OP. It was verified that the presence of protein in TS crude extract and tea seed waste accounted for the higher improvement. More importantly, the directly application of tea seed waste in enzymatic hydrolysis could achieve the similar improvement on enzymatic hydrolysis of OP, where chemosynthetic surfactant (PEG6000) was added. The residual enzyme activities in supernatant of enzymatic hydrolysis were also determined to reveal the changes on enzyme adsorption after adding surfactants. High-throughput sequencing of the 16S rRNA gene and denaturing gradient gel electrophoresis showed that Vibrionaceae was persistent with high relative abundances in the intestine from cultivated shrimp during all the studied stages. The massive mortality observed at day 63 could be related to an overabundance of different Operational Taxonomic Units (OTUs) of Vibrio, Shewanella and Clostridium. Principal coordinate analysis (PCoA) showed variations in microbial structure at different culture times. These findings suggest that OTUs of different taxa contributed to the community switch from healthy to diseased individuals, questioning the hypothesis that single bacterial species is the cause of disease outbreaks. This study provided data to improve the understanding of disease outbreaks during shrimp farming.
