We found that the negative control groups, treated postsurgically with systemic steroids, showed a significant suppression of both the inflammatory response as well as the osteogenetic activity, i.e. were associated with significant general anti-anabolic effects, even when steroids were used only at low dose level. The local anti-cytokine treatment, however, was able to significantly enhance new bone formation activity, i.e. the anabolic activity, over positive control values with BMP-2 only. However, the anti-cytokine treatment was unable to reduce the local inflammatory and swelling responses.The goal of this study was to determine the injury profiles of Canadian children who presented to the Emergency Department from 1990 to 2016 due to an injury caused while traveling in a form of land transportation that did not require child restraint. A case series was conducted using data from the electronic Canadian Hospitals Injury Reporting and Prevention Program (eCHIRPP). Children who were injured while travelling on land transportation for which child restraint is not required, who presented to a Canadian Emergency Department that participates in eCHIRPP between April 1, 1990 to August 29, 2016, were included. Overall, 1856 children sustained 2139 injuries (mean age 9.8 years (SD 4.5), 45.5% male). The majority of children were injured on a school bus (49.3%). The most commonly injured body part was the head or neck (52.6%). The most common type of injury was a superficial or open wound (33.1%), followed by traumatic brain injury (19.3%). Overall, 39.4% of injuries required no treatment in hospital. Aim To examine the interplay between AKT1 and cannabis use in terms of the cognitive performance of the general population. Methods Our sample consisted of 389 Spanish university students. Sustained attention was measured via the Continuous Performance Test-Identical Pairs, immediate and delayed verbal memory with the Logical Memory subtest of the Wechsler Memory Scale, and working memory with the Wisconsin Card Sorting Test. Lifetime cannabis use frequency was assessed and individuals were classified as cannabis users or non-users. Two single nucleotide polymorphisms of the AKT1 gene were genotyped and, according to previous studies, each subject was defined as a carrier of two, one or no copies of the haplotype (rs2494732(C)-rs1130233(A)). Multiple linear regressions were conducted to test the effect of the genetic variability and cannabis use (and their interaction) on cognitive performance. Results An effect of the AKT1 haplotype was found on attention scores individuals with two copies of the haplotype performed better (β=0.18, p less then 0.001 (adjusted for false discovery rate)), while neither cannabis nor the AKT1-cannabis interaction was associated with attention. No effect of AKT1, cannabis or the AKT1-cannabis interaction was found on verbal memory or working memory. Conclusions Our study provides additional evidence that AKT1 modulates cognitive performance. However, in our non-clinical sample, the previously reported interaction between cannabis use and the AKT1 gene was not replicated.Latent class models have been widely used in longitudinal studies to uncover unobserved heterogeneity in a population and find the characteristics of the latent classes simultaneously using the class allocation probabilities dependent on predictors. However, previous latent class models for longitudinal data suffer from uncertainty in the choice of the number of latent classes. In this study, we propose a Bayesian nonparametric latent class model for longitudinal data, which allows the number of latent classes to be inferred from the data. The proposed model is an infinite mixture model with predictor-dependent class allocation probabilities; an individual longitudinal trajectory is described by the class-specific linear mixed effects model. The model parameters are estimated using Markov chain Monte Carlo methods. The proposed model is validated using a simulated example and a real-data example for characterizing latent classes of estradiol trajectories over the menopausal transition using data from the Study of Women's Health Across the Nation.In Europe, a demographic transition has occurred meaning that countries have larger older adult populations who are increasingly living alone and receiving homecare rather than institutional care. Given that living alone is the greatest individual risk factor for fire mortality amongst older adults and that large differences exist in terms of both fire mortality risk and living arrangements between countries, this study investigates the association between co-living rates and fire mortality rates among older adults in Europe. Freely available datasets with aggregated European data on fire mortality, living arrangements among older adults, population statistics and GDP were analyzed using Poisson regression models. The results show that fire-related mortality rates amongst older adults in Europe is correlated with living arrangements after adjusting for GDP. Specifically, in Europe, when the share of older adults living alone increases by one percentage point, fire mortality rates increase by roughly 4 percent for both sexes.
