In forebrain, stress increased the gene expressions of Egf-Erbb1 and Nrg1/Nrg2-Erbb4 pathways more clearly in 129Sv, whereas the corresponding proteins were significantly elevated in Bl6. We suggest that not only inhibited activity of the DA system, but also reduced activity of EGF family and NMDA receptor signaling underlies higher susceptibility to stress in 129Sv. Altogether, this study underlines the better suitability of 129Sv for modelling neuropsychiatric disorders than Bl6.Currently the only effective measure against tick-borne encephalitis (TBE) is vaccination. Despite the high efficacy of approved vaccines against TBE, rare cases of vaccine failures are well documented. Both host- and virus-related factors can account for such failures. In this work, we studied the influence of mouse strain and sex and the effects of cyclophosphamide-induced immunosuppression on the efficacy of an inactivated TBE vaccine. We also investigated how an increased proportion of non-infectious particles in the challenge TBE virus would affect the protectivity of the vaccine. The vaccine efficacy was assessed by mortality, morbidity, levels of viral RNA in the brain of surviving mice, and neutralizing antibody (NAb) titers against the vaccine strain and the challenge virus. Two-dose vaccination protected most animals against TBE symptoms and death, and protectivity depended on strain and sex of mice. Immunosuppression decreased the vaccine efficacy in a dose-dependent manner and changed the vaccine-induced NAb spectrum. The vaccination protected mice against TBE virus neuroinvasion and persistence. However, viral RNA was detected in the brain of some asymptomatic animals at 21 and 42 dpi. Challenge with TBE virus enriched with non-infectious particles led to lower NAb titers in vaccinated mice after the challenge but did not affect the protective efficacy.Two novel xylanolytic enzymes, a xylanase and a β-xylosidase, were simultaneously isolated and characterized from the extracellular medium of Byssochlamys spectabilis ATHUM 8891 (anamorph Paecilomyces variotii ATHUM 8891), grown on Brewer's Spent Grain as a sole carbon source. They represent the first pair of characterized xylanolytic enzymes of the genus Byssochlamys and the first extensively characterized xylanolytic enzymes of the family Thermoascaceae. In contrast to other xylanolytic enzymes isolated from the same family, both enzymes are characterized by exceptional thermostability and stability at low pH values, in addition to activity optima at temperatures around 65 °C and acidic pH values. Applying nano-LC-ESI-MS/MS analysis of the purified SDS-PAGE bands, we sequenced fragments of both proteins. Based on sequence-comparison methods, both proteins appeared conserved within the genus Byssochlamys. Xylanase was classified within Glycoside Hydrolase family 11 (GH 11), while β-xylosidase in Glycoside Hydrolase family 3 (GH 3). The two enzymes showed a synergistic action against xylan by rapidly transforming almost 40% of birchwood xylan to xylose. These data give insight into the fine regulation of steroid genes when several isoforms are present.Mechanical properties of powder bed fusion processed unalloyed copper are reported majorly in the as-fabricated condition, and the effect of post-processes, common to additive manufacturing, is not well documented. In this study, mechanical properties of unalloyed copper processed by electron beam powder bed fusion are characterized via room temperature quasi-static uniaxial tensile test and Vickers microhardness. Tensile samples were extracted both perpendicular and parallel to the build direction and assigned to three different conditions as-fabricated, hot isostatic pressing (HIP), and vacuum annealing. In the as-fabricated condition, the highest UTS and lowest elongation were obtained in the samples oriented perpendicular to the build direction. These were observed to have clear trends between sample orientation caused primarily by the interdependencies between the epitaxial columnar grain morphology and dislocation movement during the tensile test. Texture was insignificant in the as-fabricated condition, and its effect on the mechanical properties was outweighed by the orientation anisotropy. The fractographs revealed a ductile mode of failure with varying dimple sizes where more shallow and finely spaced dimples were observed in the samples oriented perpendicular to the build direction. EDS maps reveal that grain boundary oxides coalesce and grow in HIP and vacuum-annealed specimens which are seen inside the ductile dimples and contribute to their increased ductility. Overall, for the post-process parameters chosen in this study, HIP was observed to slightly increase the sample's density while vacuum annealing reduced the oxygen content in the specimens.
