Pooled 12-month primary patency per limb was 90% (143/160; 95% confidence interval [CI] 0.84-0.95), freedom from CD-TLR 96% (124/130; 95% CI 0.91-0.99), limb salvage rate 97% (156/160; 95% CI 0.94-1.00), survival rate 90% (112/125; 95% CI 0.82-0.96), and AFS rate 89% (110/125; 95% CI 0.81-0.94). Subgroup analyses of included Absorb BVS studies showed similar results. All studies were assessed as moderate quality.

In this study, we tested whether mutations in the methylation pathway genes ten-eleven-translocation 2 (TET2) and DNA methyltransferase gene 3A (DNMT3A) improve the responses of patients with myelodysplastic syndrome (MDS) to decitabine.

We retrospectively sequenced the TET2 and DNMT3A genes from 70 patients diagnosed with de novo MDS between June 2008 and December 2011 and treated with a 5-day regimen of decitabine (290 cycles). We then analyzed treatment outcomes.

Patients with hematological improvement survived longer than those without hematological improvement (22.9 months vs. 10.9 months, p = 0.006). Among the 70 patients, 12 (17.1%) carried TET2 or DNMT3A mutations. The baseline characteristics of patients with wild type or mutated genes were similar. Patients with mutations in TET2 or DNMT3A had a higher overall response rate than those with the wild type genes (82.3% vs. 46.6%, p = 0.023). Multivariate analysis demonstrated that the TET2 or DMNT3A mutation status was associated with improved treatment responses and better overall survival among patients receiving decitabine.

These results demonstrate that TET2 mutations enhance the treatment response of MDS patients to hypomethylating agents like decitabine.

Especially, when it comes to deep reinforcement learning-based path planning, high-dimensionality makes it difficult for existing reinforcement learning-based methods to have efficient exploration which is crucial for successful training. The recently proposed soft actor-critic (SAC) is well known to have good exploration ability due to the use of the entropy term in the objective function. Motivated by this, in this paper, a SAC-based path planning algorithm is proposed. The hindsight experience replay (HER) is also employed for sample efficiency and configuration space augmentation is used in order to deal with complicated configuration space of the multi-arms. To show the effectiveness of the proposed algorithm, both simulation and experiment results are given. Because of PTSD-related stigma, firefighters may search for information online. The current study evaluated the quality, readability, and completeness of PTSD online resources, and to determine how the online treatment recommendations align with current evidence. Google.ca (Canada) searches were performed using four phrases 'firefighter PTSD', 'firefighter operational stress', 'PTSD symptoms', and 'PTSD treatment'. The 75 websites identified were assessed using quality criteria for consumer health information (DISCERN), readability and health literacy statistics, content analysis, and a comparison of treatments mentioned to the current best evidence. The average DISCERN score was 43.8 out of 75 (indicating 'fair' quality), with 9 'poor' websites (16-30), 31 'fair' websites (31-45), 26 "good" websites (46-60), and nine excellent websites (61-75). The average grade level required to understand the health-related content was 10.6. The most mentioned content was PTSD symptoms (48/75 websites) and PTSD treatments (60/75 websites). The most frequently mentioned treatments were medications (41/75 websites) and cognitive behavioural therapy (40/75 websites). Cognitive behavioural therapy is supported by strong evidence, but evidence for medications appears inconsistent in current systematic reviews. Online PTSD resources exist for firefighters, but the information is challenging to read and lacks evidence-based treatment recommendations.
