On the basis of our earlier discovered natural product penipyridone G with potential lipid-lowering utility, 35 penipyridone derivatives were designed, synthesized and characterized. Based on the oleic acid-induced HepG2 cell lipid accumulation model, compounds 12c, 14, 15f, 15k, 15o, 15p and 16f showed potent lipid-lowering activities among the synthetic compounds at 10 μM. In particular, compounds 4, 15k, 15o showed significant activities on inhibiting lipid accumulation in insulin resistant HepG2 cells, and these three compounds were safe and non-toxic within the concentration range of 400 μM. In comparison, 15o possessed the best lipid-lowering activity. Compared with the vehicle group, the triglyceride inhibition rate of 15o was about 30.2%, and the total cholesterol inhibition rate was about 14.8% at 20 μM, which was equipotent to Simvastatin. Our research indicates that 15o may serve as a promising lead compound for the development of hypolipidemic drugs.Phosphoinositides are an important class of anionic, low abundance signaling lipids distributed throughout intracellular membranes. The plasma membrane contains three phosphoinositides PI(4)P, PI(4,5)P2, and PI(3,4,5)P3. Of these, PI(4)P has remained the most mysterious, despite its characterization in this membrane more than a half-century ago. Fortunately, recent methodological innovations at the chemistry-biology interface have spurred a renaissance of interest in PI(4)P. Here, we describe these new toolsets and how they have revealed novel functions for the plasma membrane PI(4)P pool. We examine high-resolution structural characterization of the plasma membrane PI 4-kinase complex that produces PI(4)P, tools for modulating PI(4)P levels including isoform-selective PI 4-kinase inhibitors, and fluorescent probes for visualizing PI(4)P. The proposed method has been used for the diagnosis of pneumonia caused by the 2019 novel coronavirus disease (COVID-19) using chest X-ray image (CXI) and chest computer tomography image (CCTI). The FBSED method is used to decompose CXI and CCTI into sub-band images (SBIs). The SBIs are then used to train various pre-trained convolutional neural network (CNN) models separately using a transfer learning approach. The combination of SBI and CNN is termed as one channel. Deep features from each channel are fused to get a feature vector. Different classifiers are used to classify pneumonia caused by COVID-19 from other viral and bacterial pneumonia and healthy subjects with the extracted feature vector. The different combinations of channels have also been analyzed to make the process computationally efficient. For CXI and CCTI databases, the best performance has been obtained with only one and four channels, respectively. The proposed model was evaluated using 5-fold and 10-fold cross-validation processes. The average accuracy for the CXI database was 100% for both 5-fold and 10-fold cross-validation processes, and for the CCTI database, it is 97.6% for the 5-fold cross-validation process. Therefore, the proposed method may be used by radiologists to rapidly diagnose patients with COVID-19.

Although the psychological effects of the COVID-19 pandemic have been investigated, there is no study comparing the effects between the peaks. This study aims to compare the levels of anxiety, depression, and stress of healthcare workers struggling with pandemic between the first and second peaks.

A total of 2460 healthcare workers, 1051 from the first peak period and 1409 from the second peak period, were included in the study. The first peak measurements of the participants were made between 07.04.2020 and 05.05.2020 and the second peak measurements were made between 22.11.2020 and 20.12.2020 according to the peak period in Turkey. Depression-Anxiety-Stress-21(DASS-21) scale was applied to the participants online by the purpose of the study.

The mean age of the participants was 32.63±7.70, and 66.5% of them were female. A statistically significant difference was found between the income status (p<0.001), lifestyle (p<0.001) and COVID-19 test result (p<0.001), DASS-21 Depression (p<0.001, t=-5.311), Anxiety (p<0.001, t=-8.244), Stress (p<0.001, -10.056) and total(p<0.001, t=-8.719) scores of the two groups.

The authenticity of the starting plant material was confirmed using the nuclear ribosomal internal transcribed spacer as a molecular marker, DNA content and chromosome number. Phenolic constituents were analyzed using high-performance liquid chromatography to complement species-specific metabolic profiles. In several Hypericum representatives, the pharmacologically important compounds, including naphthodianthrones; phloroglucinol derivatives; chlorogenic acid; and some classes of flavonoids, particularly the flavonols rutin and hyperoside, flavanol catechin, and flavanones naringenin and naringin, were reported for the first time. Comparative multivariate analysis of chemometric data for seedlings cultured in vitro and acclimated to the outdoor conditions revealed a strong genetically predetermined interspecific variability in phenolic compound content.
