OBJECTIVES AND METHODS M. canis strains were collected from animal hosts that tested positive (Group I; n=13) and negative (Group II; n=14) to this pathogen following itraconazole (ITC) therapy. In vitro ITC susceptibility was assessed according to the Clinical Laboratory Standards Institute (CLSI M38-A2) methodology using conidia, hypha-conidia and arthroconidia at 3 and 7 days of incubation in order to assess the most suitable inoculum and incubation time. Successively, ketoconazole (KTC), voriconazole (VRC) terbinafine (TRB), posaconazole (PSZ), fluconazole (FLC) and griseofulvin (GRI) susceptibilities were assessed using the chosen inoculum. RESULTS The MIC values of ITC after three day-incubation were equal than those recorded after seven day-incubation. ITC MICs were ≤1μg/ml for strains from Group II and >1μg/ml for those of Group II only when conidia were used. All strains showed high susceptibility to VRC, POS, TEB and low susceptibility to ITC, KTC, GRI and FLC regardless of the source and incubation time. CONCLUSIONS AND CLINICAL IMPORTANCE Results suggest that correlation between the in vitro results and clinical outcome was observed only by incubating conidia for 3 days at 30±2°C.These conditions might be most suitable to assess in vitro susceptibility of M. canis and assist in determining the occurrence of drug resistance and cross -resistance phenomena. All rights reserved.BACKGROUND We herein investigate the safety and efficacy of single-agent anti-rejection regimens in a mouse vascularized composite allotransplantation (VCA) model. METHODS Orthotopic hind-limb transplantations (Balb/c→C57BL/6) were performed using 6-8 week old mice. A thirty-day regimen of either rapamycin, tacrolimus (both 1, 3, 5mg/kg/d) or cyclosporine (25, 35, 50mg/kg/d) was used. Primary endpoints were animal and graft survival, and secondary chimerism and regulatory T cell levels. RESULTS For rapamycin and tacrolimus given at 1, 3, and 5mg/kg/day, median graft survival time (MST) was 23d(18-28d), 30d(23-30d), and 30d(30-30d) and 14d(13-18d), 30d(16-30d), and 30d(30-30d) respectively. For cyclosporine dosed at 25 and 35mg/kg/day, MST was 15d(12-18d) and 21d(14-27d). Toxicity from CsA 50mg/kg led to 100% mortality. Mixed chimerism levels were higher in rapamycin treated animals than in tacrolimus treated recipients (p=0.029). Tacrolimus was superior in preventing leukocyte recruitment to the allograft. CONCLUSIONS In murine VCA, no standardized immunosuppressive regimen exists, limiting comparability of outcomes and survival. Our data demonstrate that rapamycin and tacrolimus maintenance treatment at 5mg/kg/d both yielded allograft survival ( less then grade 3 rejection) in all animals. On postnatal day (PND) 21, forty-five male Wistar rats were divided in 2 groups, according to their diet (1-Control) receiving standard lab chow or (2-Palatable Diet) receiving both standard chow plus palatable diet ad libitum. At PND 60, behavioral tests were performed to investigate memory in distinct tasks. Hippocampal plasticity markers were investigated at PND 28 in half of the animals, and after the behavioral tests. Palatable diet consumption induced an impairment in memory, aversive or not, and increased Na+ , K+ -ATPase activity, both at PND 28, and in the adulthood. Synaptophysin, brain-derived neurotrophic factor (BDNF), and protein kinase B (AKT) and phosphorylated AKT were reduced in the hippocampus at PND 28. However, at PND 75, this diet consumption led to increased hippocampal levels of synaptophysin, spinophilin/neurabin-II, and decreased BDNF and neuronal nitric oxide synthase.
