This study investigated the variability in the worst-case scenario (WCS) and suggested a framework to improve the definition and guide further investigation. Optical tracking data from 26 male players across 38 matches were analysed to determine the WCS for total distance, high-speed running (>5.5 m.s-1) and sprinting (>7.0 m.s-1) using a 3-minute rolling window. Position, total output, previous epoch, match half, time of occurrence, classification of starter vs substitute, and minutes played were modelled as selected contextual factors hypothesized to have associations with the WCS. Linear mixed effects models were used to account for cross-sectional observations and repeated measures. Unexplained variance remained high (total distance R2 = 0.53, high-speed running R2 = 0.53 and sprinting R2 = 0.40). Intra-individual variability was also high (total distance CV = 4.6-8.2%; high-speed CV = 15.6-37.8% and Sprinting CV = 21.1-76.4%). The WCS defined as the maximal physical load in a given time-window, produces unstable metrics lacking context, with high variability. Furthermore, training drills targetting this metric concurrently across players may not have representative designs and may underprepare athletes for complete match demands and multifaceted WCS scenarios. Using WCS as benchmarks (reproducing similar physical activity for training purposes) is conceptually questionable.Preadipocyte proliferation is a critical and precisely orchestrated procedure in adipogenesis, which is highly regulated by microRNAs (miRNAs). A previous study identified that the expression of miR-33a is different in intramuscular fat (IMF) tissues from steers and bulls. In the present study, miR-33a was overexpressed in bovine preadipocytes, and a total of 781 differentialy expressed genes were found, including 348 upregulated and 433 downregulated genes. Gene Ontology and Kyoto Encyclopaedia of Genes and Genomes (KEGG) pathway analyses of the differentially expressed genes enriched cell division and cell cycle respectively. MiR-33a overexpression decreased the rate of preadipocyte proliferation. Synchronously, the mRNA and protein expression levels of proliferation-related marker genes, including cyclin B1 (CCNB1) and proliferating cell nuclear antigen (PCNA), were decreased. In contrast, inhibiting miR-33a increased the rate of preadipocyte proliferation, and expression levels of CCNB1 and PCNA. Furthermore, based on luciferase reporter assays, miR-33a targeted directly cyclin-dependent kinase 6 (CDK6)-3'UTR and inhibited CDK6 protein expression. Interestingly, the silencing of CDK6 inhibited bovine preadipocyte proliferation and proliferation-related genes. Therefore, miR-33a inhibits the proliferation of bovine preadipocytes. CDK6 is the target gene of miR-33a and may be involved in the effects of miR-33a on bovine preadipocyte proliferation.Dietary exposure to heavy metals is threatening human health worldwide. In this study, the concentration of cadmium, mercury, arsenic and chromium in 258 samples of brown rice, grown in Yangtze River Delta where the soils were low-level contaminated, were investigated. In 12 (4.6%) and 10 (3.9%) rice samples the concentrations of Cd and Hg, respectively, exceeded the limit for food. ANOVA showed that Cd and Hg concentrations in rice grains collected from Nanjing and Jiaxing were higher than in the less developed city Yancheng. Students' t-test showed Cd and Hg were accumulated in hybrid rice higher than in conventional rice. The hazard quotients (HQs) showed a low risk from rice consumption. Conventional rice was recommended to cultivate to reduce the current risk in the soil defined as safe use level in Yangtze River Delta.Purpose The objective of this study was to evaluate a plyometric conditioning activity (3 sets of 5 countermovement jumps, [CA]) for twitch properties and voluntary knee extension. Methods After a familiarization session, fourteen highly trained sprint athletes, 12 men (23.25 ± 7.17 years) and 2 women (23.0 ± 2.8 years) performed 2 experiments, each in a randomized order (crossover design). In one experiment, the time-course of twitch contractile properties was evaluated with and without the previous CA at 2, min intervals to 10 min of recovery. In the second session, maximal voluntary knee extension was evaluated at the same recovery intervals, for control and experimental condition in random order. Results Mixed-model ANOVA with Bonferroni post-hoc revealed significant differences between pre-test and 2 min (p  0.05). No significant difference was observed for voluntary RTD following CA. Conclusion The plyometric CA increased twitch peak torque and RTD consistent with PAP; however, there was no effect of CA on voluntary RTD of knee extension at any time after the plyometric CA. Even with PAP confirmed, we observed that the CA fails to improve isometric RTD of quadriceps femoris muscles. HighlightsA plyometric CA significantly increased twitch peak torque (at 2 and 4 min) and twitch rate of torque development (at 2 min) of quadriceps femoris muscles, indicating postactivation potentiation (PAP).No effect was observed for twitch contraction time and ½ relaxation time after the CA.No improvement was observed on voluntary rate of torque development evaluated at the same time intervals.Influenza H3N8 viruses have been recovered frequently from wild bird species, including Anseriformes (primarily from migratory ducks) and Charadriiformes (primarily from shorebirds). However, little attention has been given to the transmission ability of H3N8 avian influenza viruses among mammals. Here, we study the potential human health threat and the molecular basis of mammalian transmissibility of H3N8 avian influenza viruses isolated from wild bird reservoirs. We classified eight H3N8 viruses into seven different genotypes based on genomic diversity. Six of eight H3N8 viruses isolated naturally from wild birds have acquired the ability to bind to the human-type receptor. PLX8394 However, the affinity for α-2,6-linked SAs was lower than that for α-2,3-linked SAs. Experiments on guinea pigs demonstrated that three viruses transmitted efficiently to direct-contact guinea pigs without prior adaptation. Notably, one virus transmitted efficiently via respiratory droplets in guinea pigs but not in ferrets. We further found that the PB1 S524G mutation conferred T222 virus airborne transmissibility between ferrets.
