KEY FINDINGS Meriolin1 could exert the antitumour activity on SH-SY5Y cells by inducing cell cycle arrest, cell autophagy, the mitochondrion-dependent cell apoptosis and targeting the Akt/MAPKs signalling pathway. CONCLUSIONS Meriolin1 might be a promising therapeutic candidate for neuroblastoma. BTK inhibitor chemical structure © 2020 Royal Pharmaceutical Society.INTRODUCTION Preimplantation genetic testing (PGT) is growing in importance and volume internationally. International societies such as the European Society for Human Reproduction and Embryology compile international results and these data are published in scientific journals. We present the first compilation of practices, quality measuress and outcome data from Nordic clinics performing PGT. MATERIAL AND METHODS We conducted a structured online survey of PGT practices in the Nordic countries to compare clinical and laboratory techniques, outcomes and quality measures applied in Nordic clinics. The survey was designed by the authors and answered by the authors and members of the study group. The outcome data represents results from 2018. Results and details were clarified through iteration with responding clinics while maintaining anonymity. Response rate in the study was 80%, with 8 of 10 clinics performing PGT responding. RESULTS Most of the PGT cycles in the Nordic countries are funded through the public huntries and most of them use local laboratories for genetic analyses of the biopsies. Laboratory procedures are largely in accordance with international guidelines and the outcome after PGT in terms of clinical pregnancy per transfer is comparable to results in international reports. © 2020 Nordic Federation of Societies of Obstetrics and Gynecology.Nearly two in five youth with major depressive disorder fail to respond to first-line interventions. As such, treatment-resistant depression represents a formidable challenge for clinicians and researchers. In fact, even considering the diagnosis of treatment-resistant depression in children and adolescents requires (a) defining treatment-resistant depression and, by extension, treatment failure; (b) defining recovery; (c) understanding its developmental trajectory; and in addition to (d) understanding the evidence for treatment interventions in this population. Accumulating data suggest that treatment-resistant depression is heterogeneous and that this heterogeneity may inform interventions. Additionally, these data suggest that substantially more nuance is needed in evaluating the 'adequacy' of prior treatments whether they are psychotherapeutic or psychopharmacologic. Last, adjunctive interventions that focus on neuromodulation, glutamatergic, GABAergic, and inflammatory pathways remain poorly understood in youth with treatment-resistant depression despite very significant advances in adults with treatment-resistant depression. © 2020 The Authors. Insight in the competitive advantage of lactic acid bacteria over other fermentative bacteria in a mixed culture enables ecology-based process design and can aid the development of sustainable and energy-efficient bioprocesses. Here we demonstrate the enrichment of lactic acid bacteria in a controlled sequencing batch bioreactor environment using a glucose-based medium supplemented with peptides and B vitamins. A mineral medium enrichment operated in parallel was dominated by Ethanoligenens species and fermented glucose to acetate, butyrate and hydrogen. The complex medium enrichment was populated by Lactococcus, Lactobacillus and Megasphaera species and showed a product spectrum of acetate, ethanol, propionate, butyrate and valerate. An intermediate peak of lactate was observed, showing the simultaneous production and consumption of lactate, which is of concern for lactic acid production purposes. This study underlines that the competitive advantage for lactic acid-producing bacteria primarily lies in their ability to attain a high biomass specific uptake rate of glucose, which was two times higher for the complex medium enrichment when compared to the mineral medium enrichment. The competitive advantage of lactic acid production in rich media can be explained using a resource allocation theory for microbial growth processes. © 2020 The Authors. Biotechnology and Bioengineering published by Wiley Periodicals, Inc.AIM The study aimed to assess the effectiveness of an assertiveness training program on psychological wellbeing and work engagement among novice psychiatric nurses. DESIGN A quasi-experimental design was utilized (one group pre/post assessment). SETTING The study was conducted at The Abbasia hospital for mental health in Cairo, Egypt. SUBJECTS The subjects of the study were 36 novice nurses who were starting their careers the hospital during 2017/2018. TOOLS The current study used four tools for collecting the data socio-demographic data sheet, Rathus Assertiveness Schedule, Riff's Psychological Well-Being Scale, and Utrecht Work Engagement Scale. RESULTS The study results revealed a statistically significant difference between measure one and measure two intervention program regarding assertiveness skills, psychological well-being, and work engagement. Also, there was a significant positive correlation between the total mean scores of assertiveness skills and total mean scores of psychological well-being. CONCLUSIONS This single-group feasibility study demonstrated that assertiveness training for novice nurses seems feasible. It may achieve a favorable outcome in developing assertiveness skills and improving psychological wellbeing. RECOMMENDATIONS Further randomized controlled trials with more extended follow-up periods are required. © 2020 Wiley Periodicals, Inc.Synapses are enriched in the cytoskeletal protein actin, which determines the shape of the pre- and postsynaptic compartments, organizes the neurotransmitter release machinery, and provides a framework for trafficking of components. In the postsynaptic compartment, interactions with actin or its associated proteins are also critical for the localization and activity of synaptic neurotransmitter receptors and ion channels. Actin binding proteins, including spectrin and α-actinin, serve as molecular linkages between the actin cytoskeleton and a diverse collection of receptors, including the NMDA receptor (NMDAR) and voltage-gated Na+ channels. The actin cytoskeleton can regulate neurotransmitter receptors and ion channels by controlling their trafficking and localization at the synapse and by directly gating receptor channel opening. We highlight evidence that synaptic actin couples physically and functionally to the NMDAR and supports its activity. The molecular mechanisms by which actin regulates NMDARs are only just emerging, and recent advancements in light and electron microscopy-based imaging techniques should aide in elucidating these mechanisms.
