Infantile pneumonia (IP) is an acute lower respiratory infection and brings a heavy burden to children health. Circular RNAs (circRNAs) participate in the regulation of pneumonia process. In this research, the effects of circ_0038467 in regulating lipopolysaccharide (LPS)-induced cell injury and underlying mechanism were revealed. Results showed that circ_0038467 expression and TLR4 protein level were upregulated, while miR-195-5p expression was downregulated in LPS-induced MRC-5 cells. Circ_0038467 silencing restored LPS-mediated inhibition on cell proliferation and promotion on apoptosis and inflammatory response. Additionally, circ_0038467 acted as a sponge of miR-195-5p, which was further revealed to target TLR4. MiR-195-5p inhibitor reversed circ_0038467 silencing-mediated influences under LPS treatment. Furthermore, LPS-activated NF-κB pathway was partly blocked by circ_0038467 silencing, which was restrained by TLR4 overexpression. Circ_0038467 silencing protected MRC-5 cells from LPS-induced injury by miR-195-5p/TLR4 axis through NF-κB pathway, providing a theoretical basis for circRNA-directed IP therapy.Mat and liquid-type electric insecticide vaporizers continuously emit insecticides into the surrounding air. Because both the target insects, mostly mosquitoes, and humans are exposed to those insecticides, it is crucial to understand and monitor their deposition and spatial distribution in treated areas. In the current study, we examined the evaporation of insecticides from seven commercial liquid and mat vaporizers and measured their knock-down and insecticidal activity against the adult females of the Asian tiger mosquito, Aedes albopictus (Skuse). Electric vaporizers from three manufactures had differences in their heaters and active ingredients. Most had continued evaporation during hourly and daily monitoring; however, some liquid vaporizers failed to continue emission to their designated end periods. KRT-232 manufacturer Overall, mosquitoes located at the upper position in a Peet-Grady chamber and in a field-simulation room exhibited faster knock-down activity than did mosquitoes in other areas, indicating that the insecticides accumulated on the ceiling area. Although most of mat and liquid vaporizers had less then 60 min of average KT90 values when tested in the Peet-Grady chamber (1.8 × 1.8 × 1.8 m), they failed to have any knock-down in 2 h of observation in the field-simulation room (6.8 × 3.4 × 2.7 m) but showed 72. 8 ± 11.7% and 56. 7 ± 7.3% knock-down in the mat and liquid vaporizers, respectively, in 3 h of operation. Further study will be required to examine whether this relatively limited efficacy can be compensated by other physiological and behavioral effects, including disruption in host-seeking or blood-sucking activities.

To establish an ultra-high performance liquid chromatography (UPLC) method for simultaneous determination of umbelliferonel, nodakenin, psoralen, xanthotoxin and bergapten contents in Peucedanum decursivum Radix.

The analysis was achieved on a Symmetry®C18 column (4.6mm×250mm, 5μm), with acetonitrile and water as the mobile phase in gradient elution mode. The column temperature was maintained at 30°C, with flow rate 1.0mL·min-1. The injection volume of sample was 10μL. The ultraviolet detection wavelength was set at the maximum absorption wavelength 325nm for umbelliferonel and nodakenin, 259nm for psoralen, xanthotoxin and bergapten, respectively.

The five kinds of coumarins in Peucedanum decursivum Radix were separated well and the linear relation was obtained (R2≥0.9998). The average recoveries were 101.31, 105.27, 90.85, 106.42 and 90.19%, respectively, with Relative standard deviation (RSD) 3.07, 3.17, 1.62, 2.53 and 4.54%, respectively.

The established method was accurate and feasible, which could be used as the basis of quality control of Peucedanum decursivum Radix.

ITS, β-tubulin, and calmodulin DNA sequencing was performed for species identification. Broth microdilution susceptibility testing for amphotericin B, voriconazole, posaconazole, itraconazole, isavuconazole, anidulafungin, caspofungin, and micafungin was done according to CLSI for all strains. A. fumigatus was most frequently recovered from clinical specimens, while A. niger was commonly encountered in soil, both followed by A. flavus in second place. A total of 60 (30%) cryptic species were identified, with A. tubingensis and A. tamarii being the most commonly found. The decreased susceptibility to amphotericin B and azoles was 32% for both, and were mainly led by A. fumigatus, whereas this percentage decreased to 9% for caspofungin particularly in A. terreus. More than 75% of cryptic species were susceptible in vitro to all antifungals. Multi-azole decreased susceptibility was detected only in 7 isolates. Given that antifungal resistance in Aspergillus spp. is an increasing worldwide threat that causes major challenges in the clinical management of aspergillosis, these data highlight the need for continuous epidemiological surveillance of these pathogens for the implementation of locally-adequate treatment strategies.Microbial communities can have dramatically different compositions even among similar environments. This might be due to the existence of multiple alternative stable states, yet there exists little experimental evidence supporting this possibility. Here, we gathered a large collection of absolute population abundances capturing population dynamics in one- to four-strain communities of soil bacteria with a complex life cycle in a feast-or-famine environment. This dataset led to several observations (i) some pairwise competitions resulted in bistability with a separatrix near a 11 initial ratio across a range of population densities; (ii) bistability propagated to multi-stability in multispecies communities; and (iii) replicate microbial communities reached different stable states when starting close to initial conditions separating basins of attraction, indicating finite-sized regions where the dynamics are unpredictable. The generalized Lotka-Volterra equations qualitatively captured most competition outcomes but were unable to quantitatively recapitulate the observed dynamics.
