To evaluate the plaque morphologic quantification reliability of 3D whole-brain VWI in patients via comparison with 3D targeted VWI and 2D turbo spin-echo (TSE).

Prospective.

Thirty-three patients with symptomatic ICAD.

A 3D and 2D TSE acquired at 3.0 T.

Each participant underwent two VWI sessions with an interval of 7-10 days. Three readers identified in consensus all the plaques on both whole-brain and targeted 3D VWI. Their lumen and vessel wall area and volume, plaque burden, percent stenosis, and vessel wall remodeling were measured for by two independent readers. At each culprit plaque determined by a radiologist, the lumen and vessel wall area, plaque burden, plaque-to-wall contrast ratio (CR), and plic quantification of ICAD and outperforms 3D targeted VWI.

Remarkably, many patient-derived antibodies that potently inhibit viral infection harbor few to no mutations from the germline, suggesting that naïve antibody libraries are a viable means for discovery of novel SARS-CoV-2 neutralizing antibodies. Here, we used a yeast surface-display library of human naïve antibodies to isolate and characterize three novel neutralizing antibodies that target the RBD one that blocks interaction with angiotensin-converting enzyme 2 (ACE2), the human receptor for SARS-CoV-2, and two that target other epitopes on the RBD. These three antibodies neutralized SARS-CoV-2 spike-pseudotyped lentivirus with IC50 values as low as 60 ng/ml in vitro. Using a biolayer interferometry-based binding competition assay, we determined that these antibodies have distinct but overlapping epitopes with antibodies elicited during natural COVID-19 infection. Taken together, these analyses highlight how in vitro selection of naïve antibodies can mimic the humoral response in vivo, yielding neutralizing antibodies and various epitopes that can be effectively targeted on the SARS-CoV-2 RBD.This study aimed to examine the surface characteristics of low shrinkage composites and adhesion of Streptococcus mutans and Streptococcus mitis to these materials. Control material (glass) and three low shrinkage composites (Charisma Diamond, Kalore GC, Beatiful II LS) were used. After polishing procedure was applied to composite specimens, surface roughness (SR), surface free energy (SFE), and contact angle measurements were performed. Surfaces of composite were analyzed using scanning electron microscope and energy-dispersive X-ray spectroscopy. After pellicle formation with artificial saliva, S. mutans and S. mitis biofilms were incubated in 5% CO2 for 24 h at 37°C and were analyzed using confocal laser scanning microscopy. The lowest SR and highest SFE values were found in the control group. While the contact angle of control was statistically lower than composites, statistically difference was not found between composite groups. S. mutans adhesion of composites was significantly lower than control group, but there was no significant difference between composites.
