Episodic memory entails the storage of events together with their spatio-temporal context and retrieval comprises the subjective experience of a link between the person who remembers and the episode itself. We used an encoding procedure with mobile-phones to generate experimentally-controlled episodes in the real world object-images were sent to the participants' phone, with encoding durations up to 3 weeks. In other groups of participants, the same objects were encoded during the exploration of a virtual town (45 min) or using a standard laboratory paradigm, with pairs of object/place-images presented in a sequence of unrelated trials (15 min). At retrieval, we tested subjective memory for the objects (remember/familiar) and memory for the context (place and time). We found that accurate and confident context-memory increased the likelihood of "remember" responses, in all encoding contexts. We also tested the participants' ability to judge the temporal-order of the encoded episodes. Using a model of temporal similarity, we demonstrate scale-invariant properties of order-retrieval, but also highlight the contribution of non-chronological factors. We conclude that the mechanisms governing episodic memory retrieval can operate across a wide range of spatio-temporal contexts and that the multi-dimensional nature of the episodic traces contributes to the subjective experience of retrieval.This manuscript presents GazeBase, a large-scale longitudinal dataset containing 12,334 monocular eye-movement recordings captured from 322 college-aged participants. Participants completed a battery of seven tasks in two contiguous sessions during each round of recording, including a - (1) fixation task, (2) horizontal saccade task, (3) random oblique saccade task, (4) reading task, (5/6) free viewing of cinematic video task, and (7) gaze-driven gaming task. Here, we develop Single-Cell Chromosome Conformation Calculator (Si-C) within the Bayesian theory framework and apply this approach to reconstruct intact genome 3D structures from single-cell Hi-C data of eight G1-phase haploid mouse ES cells. The inferred 100-kb and 10-kb structures consistently reproduce the known conserved features of chromatin organization revealed by independent imaging experiments. Compared to other cancer subtypes with similar incidences and death tolls, our understanding of the somatic mutations driving cutaneous squamous cell carcinoma is limited. The main challenge is that these tumors have high mutation burdens, primarily a consequence of UV-radiation-induced DNA damage from sunlight, making it difficult to distinguish driver mutations from passenger mutations. We overcame this challenge by performing a meta-analysis of publicly available sequencing data covering 105 tumors from 10 different studies. Moreover, we eliminated tumors with issues, such as low neoplastic cell content, and from the tumors that passed quality control, we utilized multiple strategies to reveal genes under selection. In total, we nominated 30 cancer genes. Among the more novel genes, mutations frequently affected EP300, PBRM1, USP28, and CHUK. Collectively, mutations in the NOTCH and p53 pathways were ubiquitous, and to a lesser extent, mutations affected genes in the Hippo pathway, genes in the Ras/MAPK/PI3K pathway, genes critical for cell-cycle checkpoint control, and genes encoding chromatin remodeling factors. Serum lipid levels have been widely studied in PD with controversial results among different populations and age groups. The present study is aimed at determining if the serum lipid profile could be influenced by the genetic background of PD patients. We included 403 PD patients (342 sporadic PD patients, 30 GBA-associated PD patients, and 31 LRRK2-associated PD patients) and 654 healthy controls (HCs). Total cholesterol, HDL, LDL, and triglycerides were measured in peripheral blood. Analysis of covariance adjusting for sex and age (ANCOVA) and post hoc tests were applied to determine the differences within lipid profiles among the groups. Multivariate ANCOVA revealed significant differences among the groups within cholesterol and LDL levels. GBA-associated PD patients had significantly lower levels of total cholesterol and LDL compared to LRRK2-associated PD patients and HCs. The different serum cholesterol levels in GBA-associated PD might be related to diverse pathogenic mechanisms.
