Shoot contained small amount of these phenolic lipids; their average content was 3.23% of level obtained in the whole seedling. Moreover, 41.38% of resorcinolic lipids of seven-day-old shoot was accumulated in part of leaf covered by coleoptile. Interestingly, a removal of 1.07% of the primary pool of kernel alkylresorcinols by short-term washing (10 s) of wheat seed with acetone before planting decreased their level only in seed of seven-day old seedling. Compared to the respective controls, this treatment did not affect the amount of these lipids in the green part of seedling that proved that de novo synthesis of 5-n-alkylresorcinols takes place in shoots. The very similar homolog profiles of these lipids in four- and seven-day-old shoots turned out to be markedly less diversified than those found in respective seed samples. Compared to the mature wheat caryopsis, the rise in the content of very-long-chain homologs was observed only in the oldest shoot. Their increased accumulation was probably connected with formation of cuticular layer providing the defensive barrier against various phytopathogens.

This study aimed to investigate the role of prolonged second stage of labour and second stage caesarean section on the risk of spontaneous preterm birth (sPTB) in a subsequent pregnancy.

This was a retrospective cohort study of nulliparous women with two consecutive singleton deliveries between 2014 and 2017 at a tertiary centre. In the vaginal delivery cohort, subsequent pregnancy outcomes for women with a prolonged second stage (>2h) were compared with those with a normal second stage (≤2h). In the caesarean delivery cohort, women with a first stage or a second stage were compared with the vaginal delivery cohort. The primary outcome was subsequent sPTB.

A total of 821 women met inclusion criteria, of which 74.8% (614/821) delivered vaginally and 25.2% (207/821) delivered by caesarean section. There was no association between a prolonged second stage in the index pregnancy and subsequent sPTB (aOR 0.70, 95% CI 0.13-3.83, p=0.7). The risk of subsequent sPTB was threefold for those with a second stagnt pregnancy.

Human milk supports the development of a beneficial newborn intestinal microflora. We have shown previously that human milk had reduced bacteria but unchanged nutrient composition when stored at-20°C for up to nine months. We suspected declining bacterial colony counts were manifestations of bacterial dormancy and not failure of survival. We investigated differences in selected bacterial colony counts (lactobacillus, bifidobacteria, staphylococcus, streptococcus and enterococcus) in human milk stored for 2 and 12weeks at-20°C in either manual or automatic defrost freezers and whether reduced bacterial counts at 12weeks were the result of dormancy or failure of survival.

Freshly expressed milk was obtained from mothers in the NICU, divided into aliquots and stored for 2 and 12weeks at-20°C in either automatic or manual defrost freezers. Subsequently, duplicate aliquots, one thawed and the other thawed and maintained at room temperature for 4h, were plated to assess bacterial colony counts.

Significant declines in bacterial colony counts were seen from 2 to 12weeks freezer storage for all bacteria. There were no differences in colony counts between freezer types. Once thawed, no further bacterial growth occurred.

ODM201 At present there is insufficient evidence from clinical trials to determine the safety and efficacy of HCQ as a treatment for COVID-19. However, since the World Health Organisation declared COVID-19 a pandemic in March 2020, the US Food and Drug Administration issued an Emergency Use Authorisation to allow HCQ and Chloroquine (CQ) to be distributed and used for certain hospitalised patients with COVID-19 and numerous clinical trials are underway around the world, including the UK based RECOVERY trial, with over 1000 volunteers. The validation of a liquid chromatography tandem mass spectrometry (LC-MS/MS) method for the simultaneous determination of HCQ and two of its major metabolites, desethylchloroquine (DCQ) and di-desethylchloroquine (DDCQ), in whole blood is described. Methods Blood samples were deproteinised using acetonitrile. HCQ, DCQ and DDCQ were chromatographically separated on a biphenyl column with gradient elution, at a flow rate of 500 μL/min. The analysis time was 8 min. Results For each analyte linear calibration curves were obtained over the concentration range 50-2000 μg/L, the lower limit of quantification (LLOQ) was 13 μg/L, the inter-assay relative standard deviation (RSD) was less then 10% at 25, 800 and 1750 μg/L and mean recoveries were 80, 81, 78 and 62% for HCQ, d4-HCQ, DCQ and DDCQ, respectively. Conclusion This method has acceptable analytical performance and is applicable to the therapeutic monitoring of HCQ, evaluating the pharmacokinetics of HCQ in COVID-19 patients and supporting clinical trials.

An online cross-sectional survey gathered demographic (age, gender, ethnicity, education, relationship, housing and employment status, employment type, and isolation status) and clinical characteristics using validated tools (self-reported stress, anxiety, depression, and contamination/hand hygiene obsessive-compulsive symptoms).
