However, a successful early diagnosis is only possible with the existence of discriminative features. In this study, a new descriptor based on the number of colors was developed in order to successfully diagnose lesions of melanoma. The number of colors is the main feature in the identification of melanoma-type skin lesions. The user must select a threshold value when calculating the number of colors of the lesion. The incorrect threshold value selection of non-expert users disrupts the aforementioned feature and also leads to significant diagnostic errors. In this study, it was revealed that color counting threshold values have a significant effect on the distinctiveness of the number of colors. In the three dermoscopic databases, color counting threshold values that provide the maximum distinctiveness on melanoma and benign lesions were determined as 0 and 0.123 respectively. By using these color counting threshold values, the number of colors for each sample in the data sets was calculated separately. Following that, a novel attribute called the number of color difference was defined as a function of color counting threshold values. Experiments using only the proposed new descriptor yielded 52.7% higher f-measure and 84.5% higher true-positive performance than the number of colors used in the literature. The results obtained in this study revealed the importance of accurately determining the number of colors the lesions had and states that the applied color counting threshold significantly influences the classification results. Thereby, a new method is proposed for determining the critical color counting threshold. We claim that the classical ABCD rule should be improved by our new descriptor. Rhein (Rh) is an effective anti-inflammatory drug with proven efficacy in in-vitro and in-vivo models. pH sensitive Rh and NH4HCO3 laden poly (lactic-co-glycolic acid (PLGA) nanoparticles (NPs) (Rh-PLGA-NPs@NH4) are developed for an effective treatment of OA. The Rh-PLGA-NPs@NH4 are prepared along with Rh-PLGA-NPs as a control by double emulsion method. Rh-PLGA-NPs@NH4 was characterized for their size, shape, morphology and encapsulation efficiency (EE). The effect of pH on release of Rh from Rh-PLGA-NPs@NH4 was studied at different pH. Further, the cytotoxicity effect of Rh-PLGA-NPs@NH4 on THP-1 cells were evaluated. Anti-inflammatory efficacy was evaluated on LPS stimulated THP-1 cells and the release of pro-inflammatory cytokines was evaluated and compared with control. The size of Rh-PLGA-NPs@NH4 and Rh-PLGA-NPs was found to be 190.7 ± 1.2 nm and 134.6 ± 2.4 nm respectively with poly dispersity (PDI) 0.14 and 0.15. The zeta potential of Rh-PLGA-NPs@NH4 was found to be -22 ± 1.12 mV. Rh-PLGA-NPs@NH4 were uniform, smooth and spherical shape as confirmed using electron microscopy analysis. Rh-PLGA-NPs@NH4 release the Rh more effectively in the low pH of synovial fluid environment (SFE). Aiming to improve the productivity of this bacteriocin, we evaluated several potential factors that could stimulate the synthesis of bovicin HC5 and selected variants of Streptococcus equinus (Streptococcus bovis) HC5 with enhanced bacteriocin production by adaptive laboratory evolution (ALE). The highest production of the bacteriocin (1.5-fold) was observed when Strep. equinus HC5 was cultivated with lactic acid (100 mmol/L). For the ALE experiment, Strep. equinus HC5 cells were subjected to acid-shock (pH 3.0 for 2 h) and maintained in continuous culture for approximately 140 generations (40 days) in media with lactic acid (100 mmol/L) and pH-controlled at 5.5 ± 0.2. An adapted variant was selected showing a distinct phenotype (sedimentation, pigmentation) compared with the parental strain. Bacteriocin production increased 2-fold in this adapted Strep. equinus HC5 variant, which appears to be associated with changes in the cell envelope of the adapted variant and enhanced bacteriocin release into the culture media.
