Most described alcohol screening as routine, with clinicians viewed as trustworthy and objective. Patients enacted a range of self-regulatory techniques to limit their drinking but perceived such strategies as learned through experience rather than based on clinical advice. However, most saw alcohol advice as a valuable component of preventative healthcare, especially those experiencing co-occurring health conditions. CONCLUSIONS Despite strong acceptance of the screening role played by primary care clinicians, patients have less confidence in the effectiveness of alcohol advice. Primary care-based alcohol brief advice needs to reflect how individuals actually drink, and harness strategies that patients already commonly employ, such as self-regulation, to boost its relevance.BACKGROUND There has been a shift from institutional care towards home care, and from formal to informal care to contain long-term care (LTC) costs in many countries. However, substitution to home care or informal care might be harder to achieve for some conditions than for others. Therefore, insight is needed in differences in LTC use, and the role of potential informal care givers, across specific conditions. We analyze differences in LTC use of previously independent older patients after a fracture of femur and stroke, and in particular examine to what extent having a partner and children affects LTC use for these conditions. METHODS Using administrative data on Dutch previously independent older people (55+) with a fracture of femur or stroke in 2013, we investigate their LTC use in the year after the condition takes place. We use administrative treatment data to select individuals who were treated by a medical specialist for a stroke or femoral fracture in 2013. SF2312 molecular weight Subsequent LTC use is measured as using noprevalence of specific conditions within the older people population into account when designing long-term care policies.BACKGROUND The replication crisis hit the medical sciences about a decade ago, but today still most of the flaws inherent in null hypothesis significance testing (NHST) have not been solved. While the drawbacks of p-values have been detailed in endless venues, for clinical research, only a few attractive alternatives have been proposed to replace p-values and NHST. Bayesian methods are one of them, and they are gaining increasing attention in medical research, as some of their advantages include the description of model parameters in terms of probability, as well as the incorporation of prior information in contrast to the frequentist framework. While Bayesian methods are not the only remedy to the situation, there is an increasing agreement that they are an essential way to avoid common misconceptions and false interpretation of study results. The requirements necessary for applying Bayesian statistics have transitioned from detailed programming knowledge into simple point-and-click programs like JASP. Stilles and regarding their ability to detect an existing effect. CONCLUSION Based on the results, two of the commonly used indices can be recommended for more widespread use in clinical and biomedical research, as they improve the type I error control compared to the classic two-sample t-test and enjoy multiple other desirable properties.BACKGROUND Protein succinylation has recently emerged as an important and common post-translation modification (PTM) that occurs on lysine residues. Succinylation is notable both in its size (e.g., at 100 Da, it is one of the larger chemical PTMs) and in its ability to modify the net charge of the modified lysine residue from + 1 to - 1 at physiological pH. The gross local changes that occur in proteins upon succinylation have been shown to correspond with changes in gene activity and to be perturbed by defects in the citric acid cycle. These observations, together with the fact that succinate is generated as a metabolic intermediate during cellular respiration, have led to suggestions that protein succinylation may play a role in the interaction between cellular metabolism and important cellular functions. For instance, succinylation likely represents an important aspect of genomic regulation and repair and may have important consequences in the etiology of a number of disease states. In this study, we developed DeepSuccinylSite, a novel prediction tool that uses deep learning methodology along with embedding to identify succinylation sites in proteins based on their primary structure. RESULTS Using an independent test set of experimentally identified succinylation sites, our method achieved efficiency scores of 79%, 68.7% and 0.48 for sensitivity, specificity and MCC respectively, with an area under the receiver operator characteristic (ROC) curve of 0.8. In side-by-side comparisons with previously described succinylation predictors, DeepSuccinylSite represents a significant improvement in overall accuracy for prediction of succinylation sites. CONCLUSION Together, these results suggest that our method represents a robust and complementary technique for advanced exploration of protein succinylation.BACKGROUND This study explored the effects of physical activity and sedentary behaviour on the decline of cognitive ability among the elderly. To compensate for the limitations of self-reported physical activity, objective measures were used. METHODS A cross-sectional survey of 308 aged people mean 68.66 ± 5.377 years, in Nanjing, China, was conducted. Physical activity was measured using the ActiGraph GT3X+, and cognitive function was measured using the Montreal Cognitive Assessment. RESULTS The overall participant model, adjusted for age, BMI, education, and monthly average income, found that light physical activity (β = 0.006, p 0.05). Further, light physical activity only affects the cognitive ability of elderly females (β = 0.006, p  less then  0.05). There was an inverted 'U' association between moderate-vigorous physical activity and cognitive ability. The association models found that moderate-vigorous physical activity in the 22.13 min·day- 1~38.79 min·day- 1 range affected cognitive ability most beneficially, with the highest beta coefficient among all groups (β = 0.
