Since a nucleophilic sp2 boron species can be generated in situ under the combined action of an inorganic base, B2pin2 and methanol, research on base-promoted nucleophilic borylation of unsaturated compounds has attracted significant attention. A series of multi-borylated compounds, such as alkyl 1,2-bis(boronates), gem-diborylalkanes, and 1,1,2-tris(boronates), are constructed based on this strategy. These multi-borylated compounds can in turn undergo selective protodeboronation, creating a variety of useful boron-containing compounds. This Feature article documents the development of base-promoted domino-borylation-protodeboronation (DBP) strategies and their applications in organic synthesis.Antibody-targeted nanoparticles have shown exceptional promise as delivery vehicles for anticancer drugs, although manufacturability challenges have hampered clinical progress. These include the potential for uncontrolled and random antibody conjugation, resulting in masked or inactive paratopes and unwanted Fc domain interactions. To circumvent these issues, we show that the interchain disulfide of cetuximab F(ab) may be selectively re-bridged with a strained alkyne handle, to permit 'click' coupling to azide-capped nanoparticles in a highly uniform and oriented manner. When compared to conventional carbodiimide chemistry, this conjugation approach leads to the generation of nanoparticles with a higher surface loading of cetuximab F(ab) and with markedly improved ability to bind to the target epidermal growth factor receptor. Moreover, we show that entrapment of a camptothecin payload within these nanoparticles can enhance drug targeting to antigen-expressing pancreatic cancer cells, resulting in superior cytotoxicity versus the conventional nanoformulation. Collectively, this work highlights the critical need to develop refined methods for the construction of targeted nanoparticles that will accelerate their clinical translation through improved performance and manufacturability.We synthesized all major saturated and unsaturated 2-alkyl-4(1H)-quinolone N-oxides of Pseudomonas and Burkholderia, quantified their native production levels and characterized their antibiotic activities against competing Staphylococcus aureus. However, in order to further improve and develop new cellulose nanomaterials, a better understanding of the interplay between the self-assembled materials and their building blocks is crucial. This paper describes the structure-property relationships between cellulose nanofibrils (CNFs) and their resulting self-assembled structures in the form of hydrogels and aerogels. Rheological experiments revealed that the transition from viscous to elastic state with the corresponding evolution of the properties of the CNF dispersion depends on the aspect ratio and can be described in terms of the dynamic overlap concentration. The elastic shear modulus was dependent on the aspect ratio at very low CNF concentrations, reaching a plateau, where only the concentration of CNFs was relevant. This transition point in shear modulus was exploited to determine the mesh size of the fibril network, which was found to be in excellent agreement with predictions from scaling arguments. These findings highlight the possibility to tune the self-assembled materials response directly from the bottom-up by the CNF particle structure and thus, suggest new assembly routes starting directly from the CNF design.Patterned liquid crystal (LC) configurations find widespread applications in functional devices such as lenses, gratings, displays and soft-robots. In combination with external stimuli such as an applied electric field, photo-alignment at the surfaces offers an attractive way to stabilize different LC structures in the bulk of a device. Herein, a planar LC cell is developed using a photo-alignment layer at the bottom substrate and a rubbed nylon film at the top substrate. Patterned planar photo-alignment is achieved by modulating the linear polarization with a spatial light modulator (SLM) and projecting the pattern onto the bottom substrate. A ring pattern is written into the photo-alignment layer with a continuous rotation between an inner radius and an outer radius. In the other regions the alignment is parallel to the rubbing direction at the top substrate. The recommended and most widely used preservation procedure for MSI is freezing samples in isopentane and storing them at temperatures below -80 °C. On the other hand, the most common and general method for preserving biological samples in clinical practice is fixation in paraformaldehyde. Special types of samples prepared from these fixed tissues that are used for histology and immunohistochemistry are free-floating sections. It would be very beneficial if the latter procedure could also be applicable for the samples intended for subsequent MSI analysis. In the present work, we optimized and evaluated paraformaldehyde-fixed free-floating sections for the analysis of lipids in mouse brains and used the sections for the study of lipid changes in double transgenic APP/PS1 mice, a model of Alzheimer's-like pathology.
