The range of the mosquito Aedes aegypti continues to expand, putting more than two billion people at risk of arboviral infection. The sterile insect technique (SIT) has been used to successfully combat agricultural pests at large scale, but not mosquitoes, mainly because of challenges with consistent production and distribution of high-quality male mosquitoes. We describe automated processes to rear and release millions of competitive, sterile male Wolbachia-infected mosquitoes, and use of these males in a large-scale suppression trial in Fresno County, California. In 2018, we released 14.4 million males across three replicate neighborhoods encompassing 293 hectares. At peak mosquito season, the number of female mosquitoes was 95.5% lower (95% CI, 93.6-96.9) in release areas compared to non-release areas, with the most geographically isolated neighborhood reaching a 99% reduction. This work demonstrates the high efficacy of mosquito SIT in an area ninefold larger than in previous similar trials, supporting the potential of this approach in public health and nuisance-mosquito eradication programs.An amendment to this paper has been published and can be accessed via a link at the top of the paper.Education and intelligence are highly correlated and inversely associated with schizophrenia. Counterintuitively, education genetically associates with an increased risk for the disease. To investigate why, this study applies a multivariable Mendelian randomization of intelligence and education. For those without college degrees, older age of finishing school associates with a decreased likelihood of schizophrenia-independent of intelligence-and, hence, may be entangled with the health inequalities reflecting differences in education. A different picture is observed for schooling years inclusive of college more years of schooling increases the likelihood of schizophrenia, whereas higher intelligence distinctly and independently decreases it. This implies the pleiotropy between years of schooling and schizophrenia is horizontal and likely confounded by a third trait influencing education. A multivariable Mendelian randomization of schooling years and bipolar disorder reveals that the increased risk of schizophrenia conferred by more schooling years is an artefact of bipolar disorder - not education.Although cochlear venous insufficiency has been considered to cause sudden sensorineural hearing loss (SSHL), there is insufficient clinical evidence to support this hypothesis. We sought to determine whether there is a correlation between draining patterns of the dural venous sinuses and the side of the affected ear in SSHL, as well as hearing recovery. The medical records of 109 patients diagnosed with unilateral SSHL were retrospectively reviewed. Magnetic resonance images and pure tone audiometry were performed in all patients. We measured the dominance of the inferior petrosal sinus (IPS) and transverse-sigmoid sinus (TS/SS) ipsilateral to the affected ear. Most patients were characterized by asymmetric venous drainage (IPS, 53.2%; TS/SS, 81.7%). The dominant side of the IPS or TS/SS was independent of the side of the affected ear for all patients in this study. However, in 35 patients with early recovery within 2 weeks, the dominant side of TS/SS was significantly associated with the side of the affected ear (p = 0.011). Moreover, the dominance of both the IPS and TS/SS influenced hearing outcomes at 3 months. Dominant TS/SS ipsilateral to the affected ear, particularly in the presence of ipsilateral hypoplastic IPS, is associated with a favorable hearing prognosis of SSHL.Lignin is a heterogeneous aromatic polymer responsible for cell wall stiffness and protection from pathogen attack. However, lignin represents a bottleneck to biomass degradation due to its recalcitrance related to the natural cell wall resistance to release sugars for fermentation or further processing. A biological approach involving genetics and molecular biology was used to disrupt lignin pathway synthesis and decrease lignin deposition. Here, we imaged three-dimensional fragments of the petioles of wild type and C4H lignin mutant Arabidopsis thaliana plants by synchrotron cryo-ptychography. Feeding is the principal vehicle of transmission of Candidatus Liberibacter asiaticus (Ca. Las) by the Asian citrus psyllid (ACP), Diaphorina citri. This study aimed to predict putative ACP effectors that may act on the Huanglongbing (HLB) pathosystem. Bioinformatics analysis led to the identification of 131 candidate effectors. Gene expression investigations were performed to select genes that were overexpressed in the ACP head and modulated by Ca. Las. To evaluate the actions of candidate effectors on D. citri feeding, six effectors were selected for gene silencing bioassays. Double-stranded RNAs (dsRNAs) of the target genes were delivered to D. citri adults via artificial diets for five days. RNAi silencing caused a reduction in the ACP lifespan and decreased the salivary sheath size and honeydew production. Moreover, after dsRNA delivery of the target genes using artificial diet, the feeding behaviors of the insects were evaluated on young leaves from citrus seedlings. These analyses proved that knockdown of D. citri effectors also interfered with ACP feeding abilities in planta, causing a decrease in honeydew production and reducing ACP survival. Electrical penetration graph (EPG) analysis confirmed the actions of the effectors on D. citri feeding behaviors. These results indicate that gene silencing of D. citri effectors may cause changes in D. citri feeding behaviors and could potentially be used for ACP control.Ixodid ticks are ectoparasites that feed exclusively on blood as their source of nutrients. Although ticks spend most of their life off the host, until now it has been assumed that the blood and the water vapor are the only sources of water to maintain water balance and prevent desiccation. Here we report for the first time that adult lone star ticks, Amblyomma americanum, also actively drink nutrient-free water, which greatly increases their survival. The volume of ingested water is greater in females than males (0.55 ± 0.06 vs 0.44 ± 0.07 µl) and most likely due to differences in tick size. Water uptake occurs through mouthparts and it can be later observed in the salivary glands and the midgut. We also exploited this behavior by adding a variety of inorganic compounds and microorganisms to water. Addition of inorganic salts to drinking water such as KH2PO4 + NaCl+KNO3 resulted in 100% tick mortality within 3 days. As a proof of concept for using the water drinking as a delivery route of toxic reagents for ticks, we also show that adding Pseudomonas aeruginosa to drinking water quickly leads to tick death. Over the past 15,000 years, the phenotype and genotype of multiple animal species, such as dogs, pigs, sheep, goats, cattle and horses, have been substantially altered during their adaptation to the human niche. Recent methodological innovations, such as improved ancient DNA extraction methods and next-generation sequencing, have enabled the sequencing of whole ancient genomes. These genomes have helped reconstruct the process by which animals entered into domestic relationships with humans and were subjected to novel selection pressures. Here, we discuss and update key concepts in animal domestication in light of recent contributions from ancient genomics.During their annual cycles, animals face a series of energetic challenges as they prioritise different life history events by engaging in temporally and potentially spatially segregated reproductive and non-breeding periods. Investigating behaviour and energy use across these periods is fundamental to understanding how animals survive the changing conditions associated with annual cycles. We estimated year-round activity budgets, energy expenditure, location, colony attendance and foraging behaviour for surviving individuals from a population of common guillemots Uria aalge. Despite the potential constraints of reduced day lengths and sea surface temperatures in winter, guillemots managed their energy expenditure throughout the year. Values were high prior to and during the breeding season, driven by a combination of high thermoregulatory costs, diving activity, colony attendance and associated flight. Guillemots also exhibited partial colony attendance outside the breeding season, likely supported by local resources. Bobcat339 in vivo Additionally, there was a mismatch in the timing of peaks in dive effort and a peak in nocturnal foraging activity, indicating that guillemots adapted their foraging behaviour to the availability of prey rather than daylight. Our study identifies adaptations in foraging behaviour and flexibility in activity budgets as mechanisms that enable guillemots to manage their energy expenditure and survive the annual cycle.Coronavirus disease 2019 (COVID-19), caused by the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), was first reported in Wuhan, Hubei Province, China in late December 2019. We re-analysed 640 throat swabs collected from patients in Wuhan with influenza-like-illness from 6 October 2019 to 21 January 2020 and found that 9 of the 640 throat swabs were positive for SARS-CoV-2 RNA by quantitative PCR, suggesting community transmission of SARS-CoV2 in Wuhan in early January 2020.BACKGROUND To evaluate the impact of injection frequency on yearly visual outcomes of patients treated with intravitreal aflibercept for neovascular age-related macular degeneration (nAMD) over a period of 5 years in a tertiary ophthalmic centre. DESIGN Single centre, retrospective cohort study. PARTICIPANTS Consecutive treatment-naive nAMD patients initiated on aflibercept injections 5 years ago. METHODS The Moorfields OpenEyes database was searched for consecutive patients who were initiated on intravitreal aflibercept for nAMD in 2013-14 and the visual acuity (VA) in Early Diabetic Retinopathy Study (ETDRS) letters and injection records per year were recorded for a period of 5 years. Analyses of the whole cohort and a sub-sample of 5-year completers were done. The cohort was further grouped into Group A (on continuous treatment), Group B (early cessation of treatment) and Group C (interrupted treatment) to evaluate the relation between treatment frequency and visual outcomes. MAIN OUTCOME MEASURES The primary end point was change in VA at 5 years; secondary outcomes included proportion of eyes that gained or maintained VA, number of injections received and the effect of treatment frequency.
